    Notizen: Summary The triglycerides of animal fats from milk, meat or cheese can also be characterized by the gaschromatographic pattern of the fatty acid methylesters (FAME) after conversion by an unsophisticated transesterification procedure. Even from the FAME pattern falsifications of commercial animal fats can be revealed by the fast separation of the FAME using a standard CW 20 M (polyethyleneglycol) capillary column. Characteristic patterns of FAME originating from different animals are shown. The patterns of the FAME from different races of the same animal are very similar, so that the characterisation according to the type of animal is not disturbed.

    Notizen: Summary Trimethylsulfonium hydroxide (TMSH) can convert fatty acids into the corresponding fatty acid methyl esters (FAMEs) in a single step. These fatty acids may also be bound in biomolecules such as phospholipids and/or glycerides. Complex mixtures of saturated and unsaturated FAMEs which may contain hydroxy and cylopropyl groups are obtained by trans-esterification; they can easily be separated in most cases by capillary GC. When FAMEs are generated from different microorganisms e.g. bacteria the patterns of the chromatograms are characteristic. Examples of characteristic patterns of bacteria with different cell wall structures are shown. The described method of transesterification can also be applied directly to blood serum without sophisticated sample pretreatment. The profiles of the chromatograms match well those described in the literature obtained by other methods of trans-esterification or sample preparation.

    Notizen: Sampling techniques for practical quantitative capillary GC have to meet certain principal requirements. Both the absolute and the relative peak areas (e.g. column loads) must be reproducible with high precision and at high accuracy; discrimination of certain constituents according to their volatility should not take place on sampling. On the basis of systematic studies, the three most reliable sampling techniques used for GC analyses with the aim of achieving precise and accurate quantitative data proved to be the following: On-column, injection, splitless PTV injection, and an optimized version of split sampling called “cooled needle split” injection. The on-column technique can be optimized by using precolumns with wider internal diameters and without stationary phase coatings to overcome the problems of large liquid sampling volumes and for automation. The PTV technique should only be used in the splitless mode because discrimination cannot be suppressed completely with the split mode. All three of the techniques can be operated automatically, either to avoid “human interference”, i.e. to improve precision or for unattended operation to save man-power.
