    Notes: The secretion of the skin glands of the ‘orange-thighed frog’ Litoria xanthomera contains seven peptides. One of these is the known hypotensive peptide caerulein. Two new peptides, caerin 1.6 [GLFSVLGAVAKHVLPHVVPVIAEKL(NH2)], and caerin 1.7 [GLFKVLGSVAKHLLPHVAPVIAEKL(NH2)] show antibacterial properties. Two other peptides lack the first two amino acid residues of caerins 1.6 and 1.7 and show no antibacterial activity. The identification of the peptides in Litoria xanthomera confirms that this species is related to Litoria caerula, Litoria gilleni and Litoria splendida but not as closely as those three species are related to each other. © 1997 European Peptide Society and John Wiley & Sons, Ltd.

    Notes: Six peptides have been isolated and characterized from the dorsal glands of the tree frog Litoria genimaculata. One of these is the known hypotensive peptide caerulein; the others have been named maculatins. The amino acid sequences of the maculatin peptides have been determined using a combination of fast atom bombardment mass spectrometry and automated Edman sequencing. Four of the maculatin peptides show antibiotic activity, with maculatin 1.1 [GLFGVLAKVAAHVVPAIAEHF(NH2;)] showing the most pronounced activity, particularly against Gram-positive organisms. Maculatin 1.1 resembles the known caerin 1 antibiotic peptides, except that four of the central amino acid residues (of the caerin 1 system) are missing in maculatin 1.1. A comparison of the antibiotic activity of maculatin 1.1 with those of caerin 1.1 is reported. ©1998 European Peptide Society and John Wiley & Sons, Ltd.

    Notes: Correlation of a quantum many-body state makes the one-particle density matrix nonidempotent. Therefore, the Shannon entropy of the natural occupation numbers measures the correlation strength on the one-particle level. Here, it is shown how this general idea of a correlation entropy must be adapted for two-electron systems in view of conservation laws which mix Slater determinants even in the noninteracting limit. Results are presented for the correlation entropy s of H2 as a function of the nucleus-nucleus separation R. In the ground state, the entropy of the spatial factor of the wave function maximizes 1.7 bohr beyond the Coulson-Fischer separation. The role of the correlation entropy in density functional theory is also discussed. © 1997 John Wiley & Sons, Inc.

    Notes: Because of the high degeneracy of hyperspherical harmonics, a method is needed for selecting the most important ones for inclusion in hyperangular basis sets. Such a method was developed by M. Fabre de la Ripelle, who showed that the most important harmonics are λ-projections of the product of the potential and a zeroth-order wave function; and he gave these the name, “potential harmonics.” In the present study we develop Fourier-transform-based methods for generating potential harmonics and for evaluating matrix elements between them. These methods are illustrated by a small calculation on three-body Coulomb systems with a variety of mass ratios. © 1997 John Wiley & Sons, Inc.

    Notes: Electrochemiluminescence (ECL) studies of the chemiluminescent (CL) polymer diazoluminomelanin (DALM) biosynthesized in nitrate reductase transfected Escherichia coli JM109 bacteria revealed noteworthy anodic ECL and even more intense cathodic ECL. Bacterial DALM (BD) ECL was also assessed in the presence of 100 ppm of 33 different metal and non-metal ions which revealed specific anodic, but not cathodic, enhancements of BD ECL with Ag+, Hg2+ and Ru3+. The precursors and intermediate polymers which comprise DALM, such as luminol, 3-amino-L-tyrosine (3-AT), aminomelanin (AM) and diazomelanin (DM) were screened for ECL enhancement against the same set of elemental ions. Significant anodic ECL enhancements were observed for luminol with Hg2+ in the presence of tripropylamine (TPA), but not for any other DALM component in combination with other elemental ions, either anodically or cathodically. Comparison of BD with luminol in the presence and absence of TPA and Hg2+ revealed very different ECL activity patterns and suggested different mechanisms for BD and luminol ECL. © 1998 John Wiley & Sons, Ltd.

    Notes: Minisatellites provide not only the basis for DNA fingerprinting and DNA profiling but also extremely informative systems for analysing processes of tandem repeat turnover in the human genome. Minisatellite instability appears to involve distinct mutation processes in somatic and germline cells; in the germline, mutation is frequently dominated by inter-allelic conversion-like events most likely occurring at meiosis and apparently regulated by cis-acting mutation initiator elements. Attempts to define these initiators in transgenic mice have so far been thwarted by what appears to be a major human/mouse barrier to the inter-species transfer of repeat instability. Minisatellites not only show high frequency spontaneous mutation in the germline, but also appear to be very sensitive to mutation induction by ionizing radiation, both in experimentally irradiated mice and in human populations exposed following the Chernobyl disaster; the mechanisms of mutation induction by radiation remain enigmatic.

    Notes: The marron (Cherax tenuimanus) is one of the few species of freshwater crayfish native to Australia that is suitable for aquaculture and occurs only in the southwest of Western Australia. This study describes polymorphic microsatellite markers which differentiate marron populations from several geographically distinct regions (including rivers and streams, dams, and commercial marron farms) throughout Western Australia. Twenty microsatellite loci, primarily of the (CA)n. (GT)n type, were isolated and sequenced from a marron cosmid library. Three of these loci were characterised further. Two loci exhibited extensive polymorphism and one was monomorphic. The polymorphic loci exhibited Mendelian codominant inheritance in the family group comprising two individual parents and approximately 100 offspring bred for this study. These loci permitted differentiation between the five geographically distinct populations studied and thus provide a basis for genetic characterisation of marron stock in Western Australia.

    Notes: In the last six years, the Developmental Therapeutics Program (DTP) of the US National Cancer Institute (NCI) has screened over 60 000 chemical compounds and a larger number of natural product extracts for their ability to inhibit growth of 60 different cancer cell lines representing different organs of origin. Whereas inhibition of the growth of one cancer cell type gives no information on drug specificity, the relative growth inhibitory activities against 60 different cells constitute patterns that encode detailed information on mechanisms of action and resistance (as reviewed in Boyd and Paull, Drug Devel. Res. 1995, 34, 19-109 and Weinstein et al., Science 1997, 275, 343-349). In order to correlate the patterns of activity with properties of the cells, we and other laboratories are characterizing the cells with respect to a large number of factors at the DNA, mRNA, and protein levels. As part of that effort, we have developed a two-dimensional gel electrophoresis (2-DE) protein expression database covering all 60 cell types (Buolamwini et al., submitted). Here we present analyses of the correlations among protein spots (i) in terms of their patterns of expression and (ii) in terms of their apparent relationships to the pharmacology of a set of 3989 screened compounds. The correlations tend to be stronger for the latter than for the former, suggesting that the spots have more robust signatures in terms of the pharmacology than in terms of expression levels.     Notes: The relative yields of the products of isomerization of D,L-glyceraldehyde in D2O with intramolecular transfer of a hydride ([1-1H]DHA) and with proton transfer ([1-2H]DHA) were determined by high-resolution 1H NMR analyses. A study of the catalysis of this isomerization by deuteroxide ion, buffer anions and Zn2+ established the following: (1) isomerization with proton and hydride transfer occurs at approximately equal rates in dilute solutions of sodium deuteroxide; (2) Brønsted bases catalyze isomerization with proton transfer in a bimolecular reaction; (3) Zn2+ catalyzes isomerization with hydride transfer in a bimolecular reaction; and (4) Zn2+ and acetate ion react in concert to catalyze isomerization with proton transfer in a termolecular reaction. These results show that the various pathways for isomerization with proton and hydride transfer proceed via transition states of similar energies, so that there is no strong imperative for enzymatic catalysis by any particular reaction mechanism. The relevance of these results to the mechanism of action of xylose isomerase is discussed briefly. © 1998 John Wiley & Sons, Ltd.
