    Notizen: Binding, internalization, and degradation of 125I-labeled, antiproliferative, or nonantiproliferative heparan sulfate by human embryonic lung fibroblasts was investigated. Both L-iduronate-rich, antiproliferative heparan sulfate species as well as L-iduronate-poor, inactive ones were bound to trypsin-releasable, cell-surface sites. Both heparan sulfate types were bound with approximately the same affinity to one high-affinity site (Kd approximately 10-8 M) and to one (Kd approximately 10-6 M), respectively. Results of Hill-plot analysis suggested that the two sites are independent. Competition experiments with unlabeled glycosaminoglycans indicated that the binding sites had a selective specificity for sulfated, L-iduronate-rich heparan sulfate. Dermatan sulfate, which is also antiproliferative, was weakly bound to the cells. The antiproliferative effects of heparan and dermatan sulfate appeared to be additive. Hence, the two glycosaminoglycans probably exert their effect through different mechanisms. At concentrations above 5 μg/ml (approximately 10-7 M), heparan sulfate was taken up by human embryonic lung fibroblasts, suggesting that the low-affinity site represents an endocytosis receptor. The antiproliferative effect of L-iduronate-rich heparan sulfate species was also exerted at the same concentrations. The antiproliferative species was taken up to a greater degree than the inactive one, suggesting a requirement for internalization. However, competition experiments with dextran sulfate suggested that both the high-affinity and the low-affinity sites are involved in mediating the antiproliferative effect. Structural analysis of the inactive and active heparan sulphate preparations indicated that although sulphated L-iduronate appears essential for antiproliferative activity, it is not absolutely required for binding to the cells. Degradation of internalized heparan sulfate was analyzed by polyacrylamide gel electrophoresis using a sensitive detection technique.     Notizen: We have modified a method (Gilles et al: Anal. Biochem., 159:109-113, 1986) for measuring cell number, that is basec on the binding of crystal violet to cell nuclei and used it to assay effects of various glycosaminoglycans on growth of human lung fibroblasts. The procedure was modified by growing cells in microcultures (96 well microplates) and by measuring the amount of adsorbed dye using a microplate photometer after solubilisation of the cells with detergent. There was a linear relationship between absorbance and cell number measured by a Coulter Counter. The method is rapid and sensitive and can be used for screening many samples as well as measuring growth rates at low initial cell densities. Even the low growth rates obtained in the absence of serum can be detected. Human lung fibroblasts were growth arrested by serum deprivation and then grown for periods of up to 4 days in the presence of serum and exogenously added glycosaminoglycans (range, 0.1-100 μg/ml). Hyaluronan, chondroitin sulfate, and dextran sulfate were without effects, whereas dermatan sulfate, certain heparan sulfates, and heparin suppressed growth (20%-50% inhibition). The antiproliferative activity of dermatan sulfate increased with increasing iduronate content. Certain heparan sulfates with moderately high sulfate and L-iduronate contents were better inhibitors than heparin despite the fact that the latter glycan has even higher sulfate and L-iduronate contents. The antiprolifera-tive effect of exogenous glycans appeared after a lag period of 3-4 days, suggesting that they interfered with factors produced by the cells. Furthermore, the degree of inhibition was greater when the initial cell density was low. Above a certain level of seeded cells (approx. 10,000 cells/well), there was no inhibition by any of the glycosaminoglycans. It is possible that exogenous glycans cannot overcome endogenous growth-promoting effects in densely seeded cultures.

    Beschreibung: Tidal disruption events (TDEs) are transient flares produced when a star is ripped apart by the gravitational field of a supermassive black hole (SMBH). We have observed a transient source in the western nucleus of the merging galaxy pair Arp 299 that radiated 〉1.5 x 10 52 erg at infrared and radio wavelengths but was not luminous at optical or x-ray wavelengths. We interpret this as a TDE with much of its emission reradiated at infrared wavelengths by dust. Efficient reprocessing by dense gas and dust may explain the difference between theoretical predictions and observed luminosities of TDEs. The radio observations resolve an expanding and decelerating jet, probing the jet formation and evolution around a SMBH.

    Beschreibung: Compact neutron star binary systems are produced from binary massive stars through stellar evolution involving up to two supernova explosions. The final stages in the formation of these systems have not been directly observed. We report the discovery of iPTF 14gqr (SN 2014ft), a type Ic supernova with a fast-evolving light curve indicating an extremely low ejecta mass (0.2 solar masses) and low kinetic energy (2 x 10 50 ergs). Early photometry and spectroscopy reveal evidence of shock cooling of an extended helium-rich envelope, likely ejected in an intense pre-explosion mass-loss episode of the progenitor. Taken together, we interpret iPTF 14gqr as evidence for ultra-stripped supernovae that form neutron stars in compact binary systems.

    Beschreibung: We present new Hubble Space Telescope images of high-velocity H-alpha and Lyman-alpha emission in the outer debris of SN 1987A. The H-alpha images are dominated by emission from hydrogen atoms crossing the reverse shock. For the first time we observe emission from the reverse shock surface well above and below the equatorial ring, suggesting a bipolar or conical structure perpendicular to the ring plane. Using the H-alpha imaging, we measure the mass flux of hydrogen atoms crossing the reverse shock front, in the velocity intervals (7,500 〈 V(sub obs) 〈 2,800 km/s) and (1,000 〈 V(sub obs) 〈 7,500 km/s), M(sub H) = 1.2 10(exp 3) M/ y. We also present the first Lyman-alpha imaging of the whole remnant and new Chandra X-ray observations. Comparing the spatial distribution of the Lyman-alpha and X-ray emission, we observe that the majority of the high-velocity Lyman-alpha emission originates interior to the equatorial ring. The observed Lyman-alpha/H-alpha photon ratio, R(L-alpha/H-alpha) approx. = 17, is significantly higher than the theoretically predicted ratio of approx. = 5 for neutral atoms crossing the reverse shock front. We attribute this excess to Lyman-alpha emission produced by X-ray heating of the outer debris. The spatial orientation of the Lyman-alpha and X-ray emission suggests that X-ray heating of the outer debris is the dominant Lyman-alpha production mechanism in SN 1987A at this phase in its evolution.

    Beschreibung: Supernova (SN) explosions are crucial engines driving the evolution of galaxies by shock heating gas, increasing the metallicity, creating dust, and accelerating energetic particles. In 2012 we used the Atacama Large Millimeter/ Submillimeter Array to observe SN1987A, one of the best-observed supernovae since the invention of the telescope. We present spatially resolved images at 450 m, 870 m, 1.4 mm, and 2.8 mm, an important transition wavelength range. Longer wavelength emission is dominated by synchrotron radiation from shock-accelerated particles, shorter wavelengths by emission from the largest mass of dust measured in a supernova remnant (〉0.2 Solar Mass). For the first time we show unambiguously that this dust has formed in the inner ejecta (the cold remnants of the exploded star's core). The dust emission is concentrated at the center of the remnant, so the dust has not yet been affected by the shocks. If a significant fraction survives, and if SN 1987A is typical, supernovae are important cosmological dust producers.

    Beschreibung: We present the most sensitive ultraviolet observations of Supernova 1987 A to date. Imaging spectroscopy from the Hubble Space Telescope-Cosmic Origins Spectrograph shows many narrow (Delta v approximates 300 km/s) emission lines from the circumstellar ring, broad Delta v approximates 10-20 x 10(exp 3) km/s) emission lines from the reverse shock, and ultraviolet continuum emission. The high signal-to-noise ratio (〉40 per resolution element) broad Ly-alpha emission is excited by soft X-ray and EUV heating of mostly neutral gas in the circumstellar ring and outer supernova debris. The ultraviolet continuum at lambda 〉 1350 A can be explained by H-I two-photon (2s(exp 2)S(sub 1/2)-l(exp 2)S(sub 1/2)) emission from the same region. We confirm our earlier, tentative detection of N V lambda 1240 emission from the reverse shock and present the first detections of broad He II lambda1640, C IV lambda 1550, and N IV ] lambda1486 emission lines from the reverse shock. The helium abundance in the high-velocity material is He/H = 0.14 +/- 0.06. The N V /H alpha line ratio requires partial ion-electron equilibration (T(sub e)/T(sub p) approximately equal to 0.14-0.35). We find that the N/C abundance ratio in the gas crossing the reverse shock is significantly higher than that in the circumstellar ring, a result that may be attributed to chemical stratification in the outer envelope of the supernova progenitor. The N/C abundance may have been stratified prior to the ring expUlsion, or this result may indicate continued CNO processing in the progenitor subsequent to the expUlsion of the circumstellar ring.

    Beschreibung: When a massive star explodes as a supernova, substantial amounts of radioactive elements-primarily Ni-56, Ni-57 and Ti-44 are produced. After the initial from shock heating, the light emitted by the supernova is due to the decay of these elements. However, after decades, the energy powering a supernova remnant comes from the shock interaction between the ejecta and the surrounding medium. The transition to this phase has hitherto not been observed: supernovae occur too infrequently in the Milky Way to provide a young example, and extragalactic supernovae are generally too faint and too small. Here we report observations that show this transition in the supernova SN 1987A in the Large Magellan Cloud. From 1994 to 200l, the ejecta faded owing to radioactive decay of Ti-44 as predicted. Then the flux started to increase, more than doubling by the end of 2009. We show that this increase is the result of heat deposited by X-rays produced as the ejecta interacts with the surrounding material. In time, the X-rays will penetrate farther into the ejects, enabling us to analyse the structure and chemistry of the vanished star.

    Beschreibung: We report four years of radio and X-ray monitoring of the Type IIn supernova SN 2006jd at radio wavelengths with the Very Large Array, Giant Metrewave Radio Telescope and Expanded Very Large Array at X-ray wavelengths with Chandra, XMM-Newton and Swift-XRT. We assume that the radio and X-ray emitting particles are produced by shock interaction with a dense circumstellar medium. The radio emission shows an initial rise that can be attributed to free-free absorption by cool gas mixed into the nonthermal emitting region external free-free absorption is disfavored because of the shape of the rising light curves and the low gas column density inferred along the line of sight to the emission region. The X-ray luminosity implies a preshock circumstellar density approximately 10(exp 6) per cubic meter at a radius r approximately 2 x 10(exp 16) centimeter, but the column density inferred from the photoabsorption of X-rays along the line of sight suggests a significantly lower density. The implication may be an asymmetry in the interaction. The X-ray spectrum shows Fe line emission at 6.9 keV that is stronger than is expected for the conditions in the X-ray emitting gas. We suggest that cool gas mixed into the hot gas plays a role in the line emission. Our radio and X-ray data both suggest the density profile is flatter than r2 because of the slow evolution of the unabsorbed emission.

    Beschreibung: We present an observational study of the Type IIn supernovae (SNe IIn) 2005ip and 2006jd. Broad-band UV, optical and near-IR photometry, and visual-wavelength spectroscopy of SN 2005ip complement and extend upon published observations to 6.5 years past discovery. Our observations of SN 2006jd extend from UV to mid-infrared wavelengths, and like SN 2005ip, are compared to reported X-ray measurements to understand the nature of the progenitor. Both objects display a number of similarities with the 1988Z-like subclass of SN IIn including: (i) remarkably similar early- and late-phase optical spectra, (ii) a variety of high ionization coronal lines, (iii) long-duration optical and near-IR emission and, (iv) evidence of cold and warm dust components. However, diversity is apparent including an unprecedented late-time r-band excess in SN 2006jd. The observed di erences are attributed to di erences between the mass-loss history of the progenitor stars. We conclude that the progenitor of SN 2006jd likely experienced a signi cant mass-loss event during its pre-SN evolution akin to the great 19th century eruption of eta Carinae. Contrarily, as advocated by Smith et al. (2009), we nd the circumstellar environment of SN 2005ip to be more consistent with a clumpy wind progenitor.
