    Notes: Linear oligogalacturonic acids (1,4-linked α-D-galacturonic acid oligomers), obtained by partial acid hydrolysis of orange polygalacturonides, were studied by negative-ion electrospray ionization mass spectrometry, without prior sample derivatization. After preparative separation using high-resolution anion-exchange chromatography, some fractions enriched in uronic acids were desalted, transferred into a methanol+water solution adjusted to pH 10, and directly submitted to electrospray ionization mass spectrometry in the negative-ion recording mode. Clear molecular mass assignments of the oligomers, covering a degree of polymerization between 4 and 7, were obtained.

    Notes: Increased differential pumping improves the simultaneous sequential operation of ion-beam neutralization, neutral dissociation, neutral product ionization, and ion product dissociation for neutralization-reionization (NR) mass spectrometry. Following Porter's developments for neutralized ion-beam spectroscopy, instrument modifications allow a wider variety of metals (vaporization up to 800 °C) to be used for neutralization and eliminate multiplier instabilities seen previously using neutralization metals such as sodium. Irreproducibility of recovered precursor ion signals using metal vapors of low ionization potential has been traced to mercury background. Double focusing of the ionized neutral beam yields NR mass spectra of the highest resolution yet reported.

    Notes: Positive ion fast atom bombardment mass spectrometry (FABMS) of in situ N-phosphorylated oligopeptides showed intense quasi-molecular ions together with the successive alkene loss fragment ions, which afford multiple checks of the unequivocal reality of the relative molecular mass of the tested samples. More interesting, in a novel cleavage pattern only the N-phosphoryl fragment ions gave intense peaks, the C-terminal series ions being suppressed. For each of the N-terminal ions, losses of alkenes also occur to provide multiple checks for the existence of these ions. The FABMS of the in situ N- phosphorylated oligopeptides might provide an easily accessible routine method for peptide sequencing.

    Notes: Several approaches have been proposed for the use of mass spectra from collisionally activated dissociation (CAD) to determine the isomeric composition of mass analyzed ions. As originally proposed, measurement at collision gas pressures (≍ 30% transmittance) yielding maximum CAD cross-sections provides improved precision, whereas differential isomeric scattering should not be an appreciable problem, despite earlier warnings. For the C2H4O+· ions from butan-1,3-diol, this CAD method found [CH3CHO+·] = 50 ± 4%, in agreement with values of 44 ± 6 and 45 ± 6% from two methods of neutralization-reionization mass spectrometry, but in contrast with 66 ± 5% from a proposed CAD methodology using high transmittance measurements. The agreement of the three methods also shows that the principle of linear superposition of spectra is applicable to CAD as well as to electron ionization mass spectra.

    Notes: The chemical ionization mass spectra of cyclic glycols and monosaccharides using formaldehyde dimethyl acetal as reagent gas have been studied. In the gas phase, the stereospecific reaction of methylene acetal formation was observed. From the relative abundances of the characteristic ions the stereoisomers of these compounds may be definitely distinguished.

    Notes: A technique for adhesion of fine particles (ca. 5 μ) of the 405 white diatomaceous support on the inner wall of glass tube with organic glue [poly (diethylaminoethyl methacrylate) acetate] is described. The coated tube was drawn to capillary column by using a machine designed according to Desty's. The capillary column was then coated with stationary phase (such as squaiane, QF-1, Dexsil 300, PEG 20 M, OV-1, SE 30 and etc.) by the conventional dynamic or static method. This preparation method has proved easy to duplicate and is simple. The coating efficiency (UTE %) is within 60-90. The theoretical plate number per meter of an SE 30 column is about 4000, and per meter of a PEG 20 M column is about 2500. The columns thus prepared have been successfully used to analyze petroleum hydrocarbons, essential oils, petrochemicals, pheromone, steroid metabolites and others.

    Notes: A new method to prepare polymeric PLOT columns by using in situ polymerization technology is described. The method involves a straightforward in situ polymerization of the monomer. The polymer produced is directly coated on the metal tubing. This eliminates many of the steps needed in conventional polymeric PLOT column preparation. Our method is easy to operate and produces very reproducible columns, as shown previously (T. C. Shen, J. Chromatogr. Sci. 30, 239, 1992). The effects of solvents, tubing pretreatments, initiators and reaction temperatures in the preparation of PLOT columns are studied. Several columns have been developed to separate (1) highly polar compounds, such as water and ammonia or water and HCN, and (2) hydrocarbons and inert gases. A recent improvement has allowed us to produce bonded polymeric PLOT columns. These were studied, and the results are included also. © 1995 John Wiley & Sons, Inc.

    Notes: A method is presented for the analysis of shellfish tissue for domoic acid, a neurotoxic amino acid responsible for cases of amnesic shellfish poisoning. Tissue extracts are first taken through a two-stage solid-phase extraction clean-up, using reversed-phase and strong cation exchange cartridges. A two-stage derivatization, using N-methyl-bis-trifluoroacetamide followed by either N-methyl-tert-butyldimethylsilyltrifluoroacetamide or N, O-bis-trimethylsilyltrifluoroacetamide, is then used to produce an N-trifluoroacetyl-O-silyl derivative which can be analyzed by mass spectrometry with introduction via direct inlet probe, moving-belt liquid chromatograph/mass spectrometer interface, or capillary column gas chromatography. The N-trifluoroacetyl-O-tert-butyldimethylsilyl derivative, which has good stability towards hydrolysis, provides a spectrum well suited to gas chromatography/mass spectrometry (GC/MS) using selected ion recording. GC/MS data for two related compounds, kainic acid and dihydrokainic acid, are also reported. The latter is used as an internal standard for quantification of domoic acid, although the method reported is intended primarily for confirmation of the toxin and related compounds in shellfish tissue.
