Notes: Identifying complementary pairs of fragment ions, b-type vs y-type, is a key step in mass spectral sequencing of proteins without degradation prior to ionization. Observations that b-type ions tend to be less stable than y-type have been extended to carbonic anhydrase (29kDa) as a model protein. Infrared multiphoton dissociation (IRMPD) of 24+, 27+, 30+ molecular ions shows y-type ions from complementary pairs require, on average, 27% longer irradiation times for onset of abundance decrease, while in nozzle/skimmer dissociation of all charge states, y-type ions require a 20% higher potential difference. In some cases, b-ion dissociation appears to accompany its formation, as its rate of increase with increasing energy deposition is far less than that of its y-complement.

Notes: Orthoquinone diazides, which are widely used in microlithography, have attracted considerable attention, especially in terms of their application in the production of high-contrast resists. Previous electron impact mass spectrometric results have confirmed that the primary fragmentation process of these compounds is the elimination of N2 to form an indenoketene ion. This is analogous to the photodecomposition pathway which makes them effective in the lithographic process. Those results also revealed the occurrence of an alternative process, which involves a two-hydrogen reduction of the intermediate species formed prior to conversion to the ketene. The present study investigates the behaviour of the orthoquinone diazides when there is an abundance of protons available to form the reduction product. Several different types of diazonaphthalenone sulfonyl esters, ranging in complexity from the monosubstituted phenol esters to disubstituted dihydroxybenzophenones, were examined using fast atom bombardment mass spectrometry. Although reduction was the primary process in the hydrogen-rich matrices, the extent of reduction was characteristic of the particular isomer as well as the matrix used.

Notes: By dissociating noncovalent adducts with infrared radiation and minimizing ion charge density during measurement, Fourier-transform mass spectrometry with electrospray ionization yields 160 000 resolving power for porcine albumin. Its molecular weight calculated from the published sequence is 66 740, but values found for two different samples are MW = 66 736 and 66 886. The spectrum of their mixture confirms this mass difference. Similar measurements of two Protein A (45 kDa) samples also give molecular weights in substantial disagreement with literature sequences. Tandem mass spectrometry gives partial sequence information that demonstrates errors in some of the reported sequences.

Notes: Using electrospray ionization with a Fourier-transform mass spectrometer, MS/MS of molecular ions and seven fragment ions of carbonic anhydrase (259 amino acids) yields 74 identifiable fragment ions, including 23 amino acid spacings. Such sensitive and specific direct characterization of proteins appears to be complementary to conventional techniques including LC/MS/MS sequencing of oligopeptides from protein degradation.

Notes: Complete Raman data are reported for the ν1, ν6 and ν8 regions of the vibrational spectra of the C2ν deuterium-labeled pyridines and of their water complexes. Raman data are given for frequency shifts observed when some of these pyridines are dissolved in a selection of hydrogen-bonding solvents. The complicated ν8 region is afforded definitive assignment based on Fermi resonance between the A1 component of ν8 and the A1 combination tone ν1 + ν6a, as well as on the ways in which these resonances are made or broken when hydrogen-bonded complexes are formed with the deuterium-labeled pyridines.

Notes: Pyridine-3,5-d2 and pyridine-3,4,5-d3 have been prepared. Isotopic constitution and label specificity of each of several preparations were verified by mass spectrometry and proton NMR. Liquid phase Raman and both liquid and vapor phase infrared spectra of these molecules are reported. Vibrational assignments consistent with frequency sum and product rule constraints are proposed. The relative rates of base-catalyzed exchange of hydrogen with deuterium for the ring positions of pyridine-N-oxide were estimated during the course of the synthetic work. It is found that the 2 and 6 positions exchange very readily, that the 3 and 5 positions exchange slowly, and that the 4 position exchanges very slowly.

Notes: The Raman spectra of the trigonal LiCaAlF6 Crystal were recorded at room temperature and 10 K. Three A1g and eight Eg Raman lines are expected according to group theory. Based on the polarized Raman spectra, symmetry assignments were made for the 11 predicted Raman-active phonon modes. Throughout the temperature range studied, no drastic evolution of the Raman spectra was observed, which suggests that there is no structural phase transition in LiCaAlF6. Analysis of the Raman spectra was made by comparison with previous studies of several crystal fluorides such as Li2ZrF6.

Notes: Pyridine-4-d, pyridine-2,6-d2 and pyridine 2,4,6-d3 have been prepared. The isotopic composition and label specificity of each of several preparations were verified by mass spectroscopy and proton NMR. The liquid phase Raman and the liquid and vapor phase infrared spectra are reported for these materials and for pyridine at room temperature. A new vibrational assignment consistent with frequency sum and product rule constraints is proposed. Thermodynamic functions calculated from the pyridine assignment are found to be in good agreement with published experimental values.

Notes: Pyridine-2,3,5,6,-d4 has been prepared. The isotopic composition and label specificity of each of three preparations were verified by mass spectroscopy and proton NMR. The liquid phase Raman and the liquid and vapor phase infrared spectra of pyridine-2,3,5,6-d4 and pyridine-d5 at room temperature are reported. The vibrational spectra of these compounds are completely analyzed and a summary of the assignment for all eight C2ν deuterium labeled pyridines is given.
