    Notes: This study deals with some macroscopical, microscopical, and ultrastructural aspects of the spinal cord central canal of the German shepherd dog. The caudal end of the spinal cord is constituted by the conus medullaris, which may extend to the first sacral vertebra, the terminal ventricle, and the filum terminale. The latter structure is considered as internum (second to third sacral vertebrae) or externum (fifth caudal vertebra), according to its relation to the dura mater. Occasionally, there is a second anchorage which is close to the level of the sixth caudal vertebra. The central canal is surrounded by a ciliated ependymal epithelium, which differs depending upon the levels. The most caudal part of the filum terminale bears a columnar ciliated ependymal epithelium surrounded by two layers of glia and pia mater, which separate the central canal from the subarachnoid space. Microfil injections show a communication between the cavity and the subarachnoid space, as the plastic is able to pass through the ependymal epithelium. At the level of the terminal ventricle there are real separations of the ependymal epithelium, which seem to connect the lumen of the spinal canal with the subarachnoid space. These structures probably constitute one of the drainage pathways of the cerebrospinal fluid. The diameter of the central canal is related to the age of the animal. However, even in very old animals the spinal cord central canal reaches the tip of the filum terminale and remains patent until death. At the ultrastructural level the ependymal cells present villi, located on cytoplasmic projections, cilia, dense mitochondria, and oval nuclei. © 1995 Wiley-Liss, Inc.

    Notes: The intrinsic ASIS of the C-8, C-9 and C-10 methyl protons of camphor in 14 hydrocarbon solvents have been calculated using intrinsic ASIS values of the TMS protons. The intrinsic ASIS of the methyl groups show excellent correlation among themselves and, therefore, can be factored into independent solute and solvent factors UiVα. The solute factors, Ui, depend on the position of the methyl group with respect to the polar carbonyl group. The solvent factors, Vα, correlate with the benzene ring concentration of the aromatic solvents measured by d/M, which is in accordance with a general solvation mechanism but not with the formation of specific solute-solvent complexes.

    Notes: Recent studies with myosin heavy chain mutants in the slime mold Dictyostelium discoideum and the yeast Saccharoymyces cerevisiae indicate that the myosin heavy chain gene is not essential for cell survival under laboratory growth conditions. However, cells lacking a normal myosin heavy chain gene demonstrate substantial alterations in growth and cell division. In this study, we report that a disruption mutant in the rod portion of the yeast myosin heavy chain gene, MYOl, produces abnormal chitin distribution and cell wall organization at the mother-bud neck in a high proportion of dividing cells. It is suggested that this phenotype is the cause of the cell division defect and the osmotic sensitivity of yeast MYOl mutants. In the absence of a normal MYOl polypeptide, yeast cells alter their cell type specific budding pattern. It is concluded that an intact myosin heavy chain gene is required to maintain the cell type specific budding pattern and the correct localization and deposition of chitin and cell wall components during cell growth and division.

    Notes: The content of vinculin, a cytoplasmic protein found in focal contacts and cell-cell junctions, was increased in BALB/c 3T3 cells by gene transfection. The vinculin expressed from the full length chicken cDNA, incorporated into focal contacts and its pattern was identical to that of the endogenous protein. Cells stably expressing vinculin by 20% over the endogenous level had altered locomotory properties. In these cells, the ability to migrate into a wound formed in a confluent monolayer and the locomotion of individual cells were drastically reduced. The results provide direct evidence that cell locomotion can be regulated by modulating vinculin expression. © 1992 Wiley-Liss, Inc.

    Notes: Three selected peptidogycan monomers isolated from muramidase-digested Escherichia coli-sacculus were characterized by positive- and negative-ion 252Cf plasma-desorption mass spectrometry (PDMS). Dut to the high affinity of sugar moieties towards alkali cations multiple alkali attachment in PDMS can be a problem. Electrospray, nitrocellulose, nitrocellulose sandwich anthracene-9-carboxylic acid 3-(3-Pyridyl)acrylic acid deposition techniques were evaluated for the degree of (de)proltonation and cationization of the molecular species by sodium and/or potassium. Electrosprayed and nitrocellulose-deposited samples mainly exhibited intense cationized molecular ion species whereas anthracene-9-caboxylic acid-deposition sample show a similar pattern but with much lower intensity. An extension of the nitrocellulose deposition technique, the so-called nitrocellulose sandwich sample preparation reduced the aallkali attachment problem for these hydrophilic compounds. Application of the small molecule matrix 3-(3-pyridyl)acrylic acid yielded mass spectra with abundant [M+H]+ and [M—H]- ions in the positive- and in the negative-ion modes, respectively.

    Notes: Hepatocytes entrapped in collagen gel and cultured in serum-free conditions survived longer than cells cultured on plastic (5 days vs. 3 weeks), showed fewer signs of early cell senescence (no increase in c-fos oncoprotein expression), and maintained the expression of differentiated hepatic metabolic functions over a longer period of time. Cells cultured in collagen gels retained their ability to respond to hormones. The insulin-stimulated glycogen synthesis rate remained fairly constant during 18 days in culture (between 5.4 ± 0.37 and 9 ± 2.7 nmol glucose/h/μg DNA). Collagen-cultured hepatocytes recovered glycogen stores to levels similar to those found in liver, or in hepatocytes isolated from fed rats. Urea synthesis from ammonia remained stable for more than 2 weeks (average value, 23 ± 4 nmol urea/h/μg DNA). The rate of albumin synthesis in collagen-entrapped cells was maintained above the day-1 level during 18 days in culture. Cells showed high levels of glutathione (GSH) (1,278 ± 152 pmol/μg DNA). Biotransformation activities CYP4501A1, CYP4502A2, CYP4502B1, and CYP4503A1 remained fairly stable in collagen-cultured hepatocytes. CYP4502E1 and CYP4502C11 decreased but were still measurable after 18 days. After 4 days in culture, GST activity returned to levels observed in isolated hepatocytes. In contrast with plastic cultures, cells responded to CYP450 inducers (methylcholanthrene for CYP4501A1, CYP4501A2, and gluthatione-transferase, and ethanol for CYP4502E1) for more than 2 weeks. CYP4501A1, CYP4501A2, and glutathione-transferase A2 (GST A2) induction was preceded by an increase in specific mRNA, while the effects on CYP4502E1 seemed to be at a posttranslational level. Analysis of the expression of relevant hepatic genes by reverse Northern and semiquantitative reverse transcriptase-polymerase chain reaction (RT-PCR) revealed that culturing hepatocytes in collagen gels results in a sustained higher expression of key liver transcription factor genes DBP, C/EBP-α and -β, and HNF-1 and -4, as well as specific liver enzyme genes (phosphoenol pyryvate carboxykinase, and carbamoylphosphate-synthetase I).     Notes: The chemical shifts of the aldehydic proton in furfural, thiophenealdehyde and benzaldehyde have been measured in fourteen solvents. The correlation of the chemical shifts of thiophenealdehyde and benzaldehyde is excellent (r = 0·996) while it is lower for furfural-benzaldehyde (r = 0·956). The long range coupling constant Jα5 of furiurai has been measured in twelve solvents and the rotameric mole fractions determined. The chemical shifts of individual rotamers are calculated and shown to correlate with benzaldehyde, (r = 0·992; 0·993). Only one rotamer is predominant for thiophenaldehyde in all solvents. The intrinsic solvent effects of the three aldehydes are similar.

    Notes: The solvent shifts of haloformic protons, (Cl3CH, Br3CH, I3CH), have been measured in 24 n-electron donor solvents consisting of halogenated hydrocarbons, esters, ketones, ethers and amines. Deviations of ΔBr and Δ1 from linear dependence with ΔCl are indicative of the presence of halogen bond formation competitive with hydrogen bonding interactions. Bromoform interacts predominantly by hydrogen bonding, halogen bonding being detected to a small extent in chlorinated hydrocarbons and amines. Iodoform shows halogen bonding interactions which increase in relative importance to hydrogen bonding with solvent basicity. Halogen bonding is predominant for solutions of iodoform in amines.

    Notes: The solvent shifts of the methylenic protond of CH2Cl2, CH2Br2 and CH2I2 have been measured in amines, ethers and non-polar solvents. Correlation of the solvent shifts of the bromine and iodine compounds ΔCl, serves as a monitor of halogen bonding interactions which are competitive with hydrogen bonding association. Amines interact with CH2I2 via a halogen bond while in ethers hydrogen bonding predpminates. CH2Br2 shows solute-solvent interactions identical to CH2Cl2.
