    Notes: In a previous paper, the presence in human kidney vein tissue of a novel blood group A heptaglycosylceramide based on the type-3 carbohydrate chain GalNAcαl-3(Fucαl-2)Galα1-3GalNAcαl-3(Fucα1-2)Galβl-4Glcβl-1 Ceramide, was suggested based on thin-layer immunostaining and electron ionization mass Spectrometry. Ions corresponding lo a structure containing two deoxyhexoses, two hexosamines and three hexoses were identified, but no information was obtained from mass spectrometry concerning the carbohydrate sequence5. In the present paper, we report the identification of carbohydrate sequence ions corresponding to a type-3 chain A heptaglyco-sylceramide by electron ionization-tandem mass spectrometry of a permethylated-reduced glycosphingolipid mixture isolated from human kidney vein tissue. The use of a microchannel-plate-array detector increased the sensitivity for collision-induced dissociation spectra by a factor of at least ten over a conventional electron multiplier.

    Notes: Multiple clustering of ammonia molecules to triply protonated bovine insulin B chain has been observed in a novel collision cell incorporated as part of a triple quadrupole mass spectrometer. The nature of the phenomenon was examined as a function of both the collision energy and the collision gas thickness in the cell. Increased clustering was observed with higher collision-gas thickness and lower collision energy. Initial experiments indicate that the number of ammonia molecules clustering to the precursor ion may be dependent on the number of available acidic sites.

    Notes: Fluoresence resonance energy transfer (FRET) microscopy is used to observe structures of DNA aggregates formed during electrophoresis in ultradilute hydroxyethylcellulose (HEC). FRET gives evidence of aggregates with a tightly packed core and random entanglement of DNA in the outer portions. HEC concentrations also appear to play a role in aggregate formation. DNA aggregates take longer to assemble in higher HEC concentration solutions and assume a less compact form than those formed at lower HEC concentrations.

    Notes: Bacitracin is a mixture of polypeptide antibiotics produced by the action of Bacillus subtilis and Bacillus licheniformis. The mixture has previously been shown to contain at least ten components, of which only the major component, Bacitracin A, and its oxidized counterpart, Bacitracin F, have been fully structurally evaluated. Using fast atom bombardment ionization with tandem mass spectrometry, three isobaric components of one of the minor constituents of the mixture, Bacitracin B, have been structurally characterized, delineating the three single substitution sites within Bactitracin A where Ile has been replaced by Val.

    Notes: A combination of mass shifts arising from known structural variations of neutral precursors, and accurate mass measurements of key fragment tions, has led to a proposed systematization of the low-energy fragmentation mechanisms for MH+ ions of actinomycins. These rationalizations are used in a predictive manner to interpret fragment ion spectra, of unknown impurities in actinomycin standards, in terms of structure. Both the strengths and limitations of this approach are emphasized. This work has uncovered actinomycin variants in which one of the threonine residues is substituted by serine, a structural variation which does not appear to have been reported previously.

    Notes: The bacitracin antibiotic peptide complex has been analyzed using liquid chromatography in conjunction with ion spray tandem mass spectrometry and multiple reaction monitoring techniques. Fragmentation patterns were determined from the product ion spectrum of bacitracin A (mol. wt 1421.8 Da), a compound of well-established structure. Primary structural information has been determined for seven minor components of the bacitracin complex with regard to the extent of amino acid substitution taking place.

    Notes: The mass spectra of biological molecules, whose molecular mass exceeds 10 kDa, invariably contain multiply charged ions. For example, a survey scan of a small protein will produce singly, doubly and triply protonated molecules, the intensity of the doubly charged species often being greater than that of the singly charged entity. Although the spectra resulting from doubly charged peptides have not previously been studied, collisional activation of such doubly charged species may result in significant additional information pertaining to molecular structure. The techniques employed to study ions originating from multiply charged species were linked scanning of constant B/E and tandem mass spectrometry, namely low collision energy spectra acquired on a BEQQ hybrid instrument. The methodology was applied to model compounds whose tandem mass spectrometry characteristics are well known, e.g. gramicidin S and angiotensin I. The results for the product ions of the [M + 2H]2+ species of the models were obtained which highlight the methodology required for high-mass materials.

    Notes: Resonance inverse Raman spectroscopy was used to examine the interactions between the flavin and surounding protein in L-lactate oxidase (from Mycobacterium smegmatis) and Old Yellow Enzyme (from brewer's bottom yeast). Spectra were taken of the enzymes both free and bound to various ligands. For L-lactate oxidase, the ligands consisted of substrate analogs (acetate, propionate) and inorganic anions (phosphate, sulfate and nitrate). For the inorganic anions, the expected attenuation with detuning was not observed for several bands which are associated with flavin rings II and III. This effect appears to be due to a disruption of ring stacking between the uracil-pyrazine end of the flavin moiety and an aromatic amino acid. A shift in the 1232 cm-1 band and changes in the bandwidths of bands in the 1450-1600 cm-1 region indicate minor reorganization of the hydrogen bonding structure around the isoalloxazine on ligand binding.For Old Yellow Enzyme, the ligand was chloride. Chloride caused a slight change to the Raman spectrum, indicative of decreased hydrogen bonding to the flavin.

    Notes: The inverse Raman spectrum of bilirubin and bilirubin ditauride were measured under pre-resonant conditions. Spectra in chloroform and dimethyl sulfoxide were assigned by comparison with model compounds and on the basis of the known effects of the solvents on the internal hydrogen bonding in bilirubin. The band assignments were used to interpret the spectra of bilirubin ditauride in aqueous solution. Evidence is presented for strong hydrogen bonding to the pyrrole N—H bonds and to the lactam rings. It is proposed that hydrogen bonding to water is facilitated by photoactivation and, in turn, stabilizes photohiliruhin.
