    Beschreibung: The flipping of membrane-embedded lipids containing large, polar head groups is slow and energetically unfavourable, and is therefore catalysed by flippases, the mechanisms of which are unknown. A prominent example of a flipping reaction is the translocation of lipid-linked oligosaccharides that serve as donors in N-linked protein glycosylation. In Campylobacter jejuni, this process is catalysed by the ABC transporter PglK. Here we present a mechanism of PglK-catalysed lipid-linked oligosaccharide flipping based on crystal structures in distinct states, a newly devised in vitro flipping assay, and in vivo studies. PglK can adopt inward- and outward-facing conformations in vitro, but only outward-facing states are required for flipping. While the pyrophosphate-oligosaccharide head group of lipid-linked oligosaccharides enters the translocation cavity and interacts with positively charged side chains, the lipidic polyprenyl tail binds and activates the transporter but remains exposed to the lipid bilayer during the reaction.     Notizen: A single-step coating method for the preparation of glass capillary SCOT columns is described. It is reproducible and less time-consuming than the well-known two-step coating procedures. Other methods attempted are discussed briefly. Both the flame ionization and electron capture detectors could be used in conjunction with temperature programming. The separations achieved with an “activity mixture”, phenols and phenolic acids, illustrate the resolution obtained. The columns are suitable for quantitative determinations and a comparison is made with a conventional packed column.

    Notizen: Discrimination may occur when injecting samples onto gas chromatography capillary columns, whereby peak areas for higher boiling point compounds are smaller than they should be compared to lower boiling compounds. This problem is most important in quantitative work on solutes having a wide range of volatility. An all-glass inlet system was used. Injections were made with a 10-μl syringe onto a column without and injection point heater and with the column oven about 20°C below the boiling point of the solvent used for the solutes. Discrimination did not occur in a range of C12 to C36 n-alkanes.
