    Description: Genome sequencing projects are producing linear amino acid sequences, but full understanding of the biological role of these proteins will require knowledge of their structure and function. Although experimental structure determination methods are providing high-resolution structure information about a subset of the proteins, computational structure prediction methods will provide valuable information for the large fraction of sequences whose structures will not be determined experimentally. The first class of protein structure prediction methods, including threading and comparative modeling, rely on detectable similarity spanning most of the modeled sequence and at least one known structure. The second class of methods, de novo or ab initio methods, predict the structure from sequence alone, without relying on similarity at the fold level between the modeled sequence and any of the known structures. In this Viewpoint, we begin by describing the essential features of the methods, the accuracy of the models, and their application to the prediction and understanding of protein function, both for single proteins and on the scale of whole genomes.     Description: A major challenge of computational protein design is the creation of novel proteins with arbitrarily chosen three-dimensional structures. Here, we used a general computational strategy that iterates between sequence design and structure prediction to design a 93-residue alpha/beta protein called Top7 with a novel sequence and topology. Top7 was found experimentally to be folded and extremely stable, and the x-ray crystal structure of Top7 is similar (root mean square deviation equals 1.2 angstroms) to the design model.     Description: Development work on an arrangement using ailerons and spoilers for lateral control was carried out by the Vought-Sikorsky Aircraft Division of the United Aircraft Corporation on a small commercial airplane in flight and on an airfoil in a wind tunnel. Spoiler hinge moments were reduced by aerodynamic balance. The arrangement was then built into an experimental airplane and further improvements were adopted as the result of flight and tunnel tests. The use of ailerons for lateral control with flaps up, spoilers with flaps full down, and gradual transition as the flaps are lowered was found to provide lateral control under the flight conditions for which they were best suited. The ailerons were of short span, permitting the use of long-span flaps, and were drooped to a relatively large angle when the flaps were deflected. A high maximum lift coefficient was thus attained. With large control deflections in the intermediate flap-angle range and spoiler effectiveness near neutral improved by "ventilating" the spoiler, the lateral control was satisfactory for the experimental airplane and was a definite improvement over that of a conventional control arrangement.

    Description: A composite isogrid panel design for application to a rotorcraft fuselage is presented. An optimum panel design for the lower fuselage of the rotorcraft that is subjected to combined in-plane compression and shear loads was generated using a design tool that utilizes a smeared-stiffener theory in conjunction with a genetic algorithm. A design feature was introduced along the edges of the panel that facilitates introduction of loads into the isogrid panel without producing undesirable local bending gradients. A low-cost manufacturing method for the isogrid panel that incorporates these design details is also presented. Axial compression tests were conducted on the undamaged and low-speed impact damaged panels to demonstrate the damage tolerance of this isogrid panel. A combined loading test fixture was designed and utilized that allowed simultaneous application of compression and shear loads to the test specimen. Results from finite element analyses are presented for the isogrid panel designs and these results are compared with experimental results. This study illustrates the isogrid concept to be a viable candidate for application to the helicopter lower fuselage structure.

    Description: The results of an analytical and experimental investigation of the response of composite I-stiffener panels with extension-shear coupling are presented. This tailored concept, when used in the panel cover skins of a tiltrotor aircraft wing has the potential for increasing the aeroelastic stability margins and improving the aircraft productivity. The extension-shear coupling is achieved by using unbalanced +/- 45 deg. plies in the skin. Experimental and STAGS analysis results are compared for eight I-stiffener panel specimens. The results indicate that the tailored concept would be feasible to use in the wing skin of a tiltrotor aircraft. Evaluation of specimens impacted at energy level of 500 in.-lbs indicate a minimal loss in stiffness and less than 30 percent loss in strength. Evaluation of specimens with severed center stiffener and adjacent skin indicated a strength loss in excess of percent.
