    Beschreibung: The presence of lysylpyridinoline (LP) as a nonreducible cross-link in appreciable quantities has primarily been limited to the mineralized tissues, bone and dentin. However, the results reported here show that LP is not only present in the hypertrophic cartilage of the tibiotarsus isolated from newly hatched broiler chicks, but it is approx. 4-fold as concentrated as hydroxylysylpyridinoline (HP). Bone and articular cartilage surrounding the hypertrophic cartilage do not contain measurable quantities of LP. Purified LP has a fluorescent scan similar to purified HP and literature values, confirming that we indeed were measuring LP. Also, the cartilage lesion produced by immature chondrocytes from birds with tibial dyschondroplasia had LP but the HP:LP ratio was 〉 1. Thus, the low HP:LP ratio could be a marker for hypertrophic cartilage in avians.

    Beschreibung: The growth hormone (GH)-deficient dwarf rat was used to investigate recombinant human (rh) GH-induced bone formation and to determine whether rhGH facilitates simultaneous increases in bone formation and bone maturation during rapid growth. Twenty dwarf rats, 37 days of age, were randomly assigned to dwarf plus rhGH (GH; n = 10) and dwarf plus vehicle (n = 10) groups. The GH group received 1.25 mg rhGH/kg body wt two times daily for 14 days. Biochemical, morphological, and X-ray diffraction measurements were performed on the femur middiaphysis. rhGH stimulated new bone growth in the GH group, as demonstrated by significant increases (P 〈 0.05) in longitudinal bone length (6%), middiaphyseal cross-sectional area (20%), and the amount of newly accreted bone collagen (28%) in the total pool of middiaphyseal bone collagen. Cortical bone density, mean hydroxyapatite crystal size, and the calcium and collagen contents (microgram/mm3) were significantly smaller in the GH group (P 〈 0.05). Our findings suggest that the processes regulating new collagen accretion, bone collagen maturation, and mean hydroxyapatite crystal size may be independently regulated during rapid growth.

    Beschreibung: We investigated the hypothesis that hindlimb unloading inhibits healing in fibrous connective tissue such as ligament. Male rats were assigned to 3- and 7-wk treatment groups with three subgroups each: sham control, ambulatory healing, and hindlimb-suspended healing. Ambulatory and suspended animals underwent surgical rupture of their medial collateral ligaments, whereas sham surgeries were performed on control animals. After 3 or 7 wk, mechanical and/or morphological properties were measured in ligament, muscle, and bone. During mechanical testing, most suspended ligaments failed in the scar region, indicating the greatest impairment was to ligament and not to bone-ligament insertion. Ligament testing revealed significant reductions in maximum force, ultimate stress, elastic modulus, and low-load properties in suspended animals. In addition, femoral mineral density, femoral strength, gastrocnemius mass, and tibialis anterior mass were significantly reduced. Microscopy revealed abnormal scar formation and cell distribution in suspended ligaments with extracellular matrix discontinuities and voids between misaligned, but well-formed, collagen fiber bundles. Hence, stress levels from ambulation appear unnecessary for formation of fiber bundles yet required for collagen to form structurally competent continuous fibers. Results support our hypothesis that hindlimb unloading impairs healing of fibrous connective tissue. In addition, this study provides compelling morphological evidence explaining the altered structure-function relationship in load-deprived healing connective tissue.

    Beschreibung: A refinement of the current ultrasonic elasticity technique was used to measure the orthotropic elastic properties of rat cortical bone as well as to quantify changes in elastic properties, density, and porosity of the dwarf rat cortex after a treatment with recombinant human growth hormone (rhGH). The ultrasonic elasticity technique was refined via optimized signal management of high-frequency wave propagation through cubic cortical specimens. Twenty dwarf rats (37 days old) were randomly assigned to two groups (10 rats each). The dwarf rat model (5-10% of normal GH) was given subcutaneous injections of either rhGH or saline over a 14-day treatment period. Density was measured using Archimedes technique. Porosity and other microstructural characteristics were also explored via scanning electron microscopy and image analysis. Statistical tests verified significant decreases in cortical orthotropic Young's (-26.7%) and shear (-16.7%) moduli and density (-2.42%) concomitant with an increase in porosity (+125%) after rhGH treatments to the dwarf model (p 〈 0.05). A change in material symmetry from orthotropy toward planar isotropy within the radial-circumferential plane after GH treatments was also noted. These results demonstrate some alteration in bone properties at this time interval. Structural implications of these changes throughout physiological loading regimens should be explored.

    Beschreibung: We describe the scientific motivations, the mission concept and the instrumentation of SPACE, a class-M mission proposed for concept study at the first call of the ESA Cosmic-Vision 2015-2025 planning cycle. SPACE aims at producing the largest three-dimensional evolutionary map of the Universe over the past 10 billion years by taking near-IR spectra and measuring redshifts of more than half a billion galaxies at 0 〈 z 〈 2 down to AB approximately 23 over 37r sr of the sky. In addition, SPACE will also target a smaller sky field, performing a deep spectroscopic survey of millions of galaxies to AB approximately 26 and at 2 〈 z 〈 l0+. Owing to the depth, redshift range, volume coverage and quality of its spectra, SPACE will reveal with unique sensitivity most of the fundamental cosmological signatures, including the power spectrum of density fluctuations and its turnover, the baryonic acoustic oscillations imprinted when matter and radiation decoupled, the distance-luminosity relation of cosmological supernovae, the evolution of the cosmic expansion rate, the growth rate of cosmic large-scale structure, the large scale distribution of galaxies. The datasets from the SPACE mission will represent a long lasting legacy that will be data mined for many years to come.

    Beschreibung: Replacement of current turbojets by high-efficiency unducted propfans could have the unfortunate side effect of increasing cabin noise, essentially because unsteady-aerodynamic mechanisms are likely to be introduced whereby some of the energy saved may be lost again, to the production of propeller noise and to wing/pylon vibrations coupling to the cabin as a sounding board. The present study estimates theoretically associated harmonic aerodynamic forces for two candidate configurations: a pusher propeller which chops through the mean wake of the pylon supporting it, and in the process generates a blade-rate force driving the structure, and a tractor wing-mounted propeller, whose trailing rotating wake induces an unsteady downwash field generating unsteady wing airloads. Reported predictions of such propfan aerodynamic sources of structure-borne sound, or vibration, could be the basis for devising means for their mechanical isolation, and thus for the effective interruption of the structural noise path into the cabin. Both mechanisms are analyzed taking advantage of the high subsonic Mach number and high reduced frequency of the interaction between the impinging flow and the affected aerodynamic element.

    Beschreibung: This report develops and applies an aeroacoustic diffraction theory for a duct, or cowl, placed around modelled sources of propeller noise. The regime of flight speed is high subsonic. The modelled cowl's inner wall contains a liner with axially variable properties. Its exterior is rigid. The analysis replaces both sides with an unsteady lifting surface coupled to a dynamic thickness problem. The resulting pair of aeroacoustic governing equations for a lined 'ring wing' is valid both for a passive and for an active liner. Their numerical solution yields the effective dipole and monopole distributions of the shrouding system and thereby determines the cowl-diffracted component of the total radiated field. The sample calculations here include a preliminary parametric search for that liner layout which maximizes the cowl's shielding effectiveness. The main conclusion of the study is that a short cowl, passively lined, should provide moderate reductions in propeller noise.

    Beschreibung: Context. The nearby and young M star AU Mic is surrounded by a debris disk in which we previously identified a series of large-scale arch-like structures that have never been seen before in any other debris disk and that move outward at high velocities. Aims. We initiated a monitoring program with the following objectives: (1) track the location of the structures and better constrain their projected speeds, (2) search for new features emerging closer in, and ultimately (3) understand the mechanism responsible for the motion and production of the disk features. Methods. AU Mic was observed at 11 different epochs between August 2014 and October 2017 with the IR camera and spectrograph of SPHERE. These high-contrast imaging data were processed with a variety of angular, spectral, and polarimetric differential imaging techniques to reveal the faintest structures in the disk. We measured the projected separations of the features in a systematic way for all epochs. We also applied the very same measurements to older observations from the Hubble Space Telescope (HST) with the visible cameras STIS and ACS. Results. The main outcomes of this work are (1) the recovery of the five southeastern broad arch-like structures we identified in our first study, and confirmation of their fast motion (projected speed in the range 412 km/s); (2) the confirmation that the very first structures observed in 2004 with ACS are indeed connected to those observed later with STIS and now SPHERE; (3) the discovery of two new very compact structures at the northwest side of the disk (at 0.40 and 0.55 in May 2015) that move to the southeast at low speed; and (4) the identification of a new arch-like structure that might be emerging at the southeast side at about 0.4 from the star (as of May 2016). Conclusions. Although the exquisite sensitivity of SPHERE allows one to follow the evolution not only of the projected separation, but also of the specific morphology of each individual feature, it remains difficult to distinguish between possible dynamical scenarios that may explain the observations. Understanding the exact origin of these features, the way they are generated, and their evolution over time is certainly a significant challenge in the context of planetary system formation around M stars.

    Beschreibung: The project had two specific technical objectives: (1) to develop a realistic three-dimensional model of tonal noise due to rotor/stator interaction, as the input field for predictions of diffraction and dissipation by a lined cowl; and (2) to determine whether the generator curve of that cowl, or duct, could be 'steered' to yield substantially lower values of propulsor noise along the engine's fore and aft open sectors. The more general and important aim of their research is to provide the commercial aircraft industry with a useful predictive tool to help it meet its noise-reduction goals. The work has produced a tractable and yet realistic model of rotor/stator interaction noise. The blades in the fan stage are radially divergent, twisted, and of realistically wide chords to match the high frequencies and speeds of the sound-production process. The resulting three-dimensional acoustic nearfield insonifies the interior wall of the diffracting cowl, whose shape, incidentally, does not affect fore or aft noise significantly (but other factors do).

    Beschreibung: We continue a systematic study of chemical abundances of the Strontium Filament found in the ejecta of eta Carinae. To this end we interpret the emission spectrum of Sc II and Cr II using multilevel non-LTE models of these systems. Since the atomic data for these ions was previously unavailable, we carry out ab initio calculations of radiative transition rates and electron impact excitation rate coefficients. The observed spectrum is emitted from a mostly neutral region with electron density of the order of 10(exp 7) cm (exp -3) and a temperature between 6000 and 7000 K. These conditions are consistent with our previous diagnostics from [Ni II], [Ti II], amd [Sr II]. The observed spectrum indicates an abundance of Sc relative Ni that more than 40 times the solar values, while the Cr/Ni abundance ratio is roughly solar. Various scenarios of depletion and dust destruction are suggested to explain such abnormal abundances.
