    Notes: Abstract The Aspergillus nidulans hapC gene was expressed as a fusion protein with MalE or glutathione-S-transferase (GST) in Escherichia coli, and used for the purification of HapC and the preparation of anti-HapC antiserum. The CCAAT-binding factor AnCP/AnCF contains a component with an approximate molecular mass of 32 kDa that cross-reacts with the antibody. The MalE-HapC fusion protein was able to replace authentic HapC in AnCP when incubated under appropriate conditions. Furthermore, reconstitution experiments with recombinant HapC, yHAP2 and yHAP5 polypeptides showed that all three polypeptides were required for the assembly of a complex capable of binding to CCAAT-containing taaG2 promoter DNA. The relationship between AnCP/AnCF and the Saccharomyces cerevisiae HAP complex is discussed.

    Notes: Abstract The 5′ regulatory region of theamdS gene ofAspergillus nidulans, which encodes an acetamidase required for growth on acetamide as a carbon and nitrogen source, contains a CCAAT sequence which is required for setting the basal level ofamdS expression. Mobility shift studies have identified a factor inA. nidulans nuclear extracts which binds to this CCAAT sequence. InSaccharomyces cerevisiae theHAP3 gene encodes one component of a multisubunit complex that binds CCAAT sequences. A search of the EMBL and SwissProt databases has revealed anA. nidulans sequence with significant homology to theHAP3 gene adjacent to the previously cloned regulatory geneamdR. Sequencing of the remainder of this region has confirmed the presence of a gene, designatedhapC, with extensive homology toHAP3. The predicted amino acid sequence of HapC shows extensive identity to HAP3 in the central conserved domain, but shows little conservation in the flanking sequences. A haploid carrying ahapC deletion has been created and is viable, but grows poorly on all media tested. This null mutant grows especially slowly on acetamide as a sole carbon and nitrogen source, indicating thathapC plays a role inamdS expression. In agreement with this notion, it has been shown that thehapC deletion results in reduced levels of expression of anamdS::lacZ reporter gene and this effect is particularly evident under conditions of carbon limitation. Nuclear extracts prepared from thehapC deletion mutant show no CCAAT binding activity to theamdS orgatA promoters, indicating thathapC may encode a component of the complex binding at this sequence.

    Description: The detection of high-velocity H I in absorption against the nuclear continuum source in the giant elliptical galaxy NGC 315 is reported. The absorption line is found to be highly blueshifted in the reference frame of the galaxy with a projected velocity toward the galaxy of 490 km/s. The absorption line is very strong (244 + or - 4 mJy), consisting of two components with very narrow velocity widths of 2.5 km/s (FWHM) each and peak optical depths of 0.9 and 0.2. The kinetic temperature must be 150 K if the line widths are due entirely to thermal broadening, while the neutral gas column densities implied are 2-5 x 10 to the 20th hydrogen atoms/sq cm. No OH in absorption was detected in these clouds to a limiting peak optical depth of 0.04, although this result is consistent with either a galactic or extragalactic origin of the clouds. It is suggested that these clouds may have condensed out of a large unobserved galactic halo of gas, or they may reside in or have been captured from the intracluster medium.

    Description: The X-ray source 2A 0251 + 413 is identified with the AWM 7 group of galaxies. The source is extended, with a FWHM of roughly 9 arcmin if an isothermal sphere profile is assumed. The spectrum is best fitted by thermal bremsstrahlung at 4.0 + or - 0.5 keV, with a significant Fe line emission of equivalent width 0.63 keV centered at 6.6 keV. This temperature is consistent with a measured dispersion velocity of 600 km/s of the central galaxies.

    Description: A rapid sequence of highly polarized spikes was observed during the gradual rise of a longer lasting event in an Arecibo Observatory study of a radio burst at 1400 MHz from the main sequence radio star AD Leo. The maximum flux density of the spikes was 130 mJy, and they had rise times of less than about 200 msec. The rise times provide an upper limit to the linear size of the emitter of 6 billion cm, and an area of less than 0.03 of the star's surface area is inferred, for a spike brightness temperature of more than about 10 to the 13th K. The high brightness temperatures and high degrees of circular polarization are explained in terms of electron-cyclotron maser emission at the second harmonic of the gyrofrequency, in longitudinal magnetic fields of about 250 G.

    Description: The results of numerical simulations of nonlinear gravitational clustering in universes dominated by weakly interacting, 'cold' dark matter are presented. The numerical methods used and the way in which initial conditions were generated are described, and the simulations performed are catalogued. The evolution of the fundamental statistical properties of the models is described and their comparability with observation is discussed. Graphical comparisons of these open models with the observed galaxy distribution in a large redshift survey are made. It is concluded that a model with a cosmological density parameter omega equal to one is quite unacceptable if galaxies trace the mass distribution, and that models with omega of roughly two, while better, still do not provide a fully acceptable match with observation. Finally, a situation in which galaxy formation is suppressed except in sufficiently dense regions is modelled which leads to models which can agree with observation quite well even for omega equal to one.

    Description: Techniques for carrying out large N-body simulations of the gravitational evolution of clustering in the fundamental cube of an infinite periodic universe are described and compared. The accuracy of the forces derived from several commonly used particle mesh schemes is examined, showing how submesh resolution can be achieved by including short-range forces between particles by direct summation techniques. The time integration of the equations of motion is discussed, and the accuracy of the codes for various choices of 'time' variable and time step is tested by considering energy conservation as well as by direct analysis of particle trajectories. Methods for generating initial particle positions and velocities corresponding to a growing mode representation of a specified power spectrum of linear density fluctuations are described. The effects of force resolution are studied and different simulation schemes are compared. An algorithm is implemented for generating initial conditions by varying the number of particles, the initial amplitude of density fluctuations, and the initial peculiar velocity field.

    Description: Major advances in the art of redshift measurements have improved the obtainable accuracy to better than 30 km/s. It is now posible to obtain a redshift for almost any galaxy brighter than 15th magnitude on a 60-inch telescope in 60 minutes or less. These advances were utilized in an observation program initiated in the spring of 1978. This program represents a survey of radial velocities for all 2400 galaxies brighter than 14.5 at high galactic latitude in the northern hemisphere. The obtained data set has been employed as a basis for a derivation of a good measure of the local mean mass density. In addition, information was obtained concerning the overdensity and the dynamics of the local supercluster, and an analysis was conducted of the dynamics of groups and clusters of galaxies within the sample volume.

    Description: Quite soon after the first collapse of structure, almost half the matter in a neutrino-dominated universe is expected to reside in clusters. The masses and binding energies of these neutrino clusters are too large for them to be identified with observed galaxy clusters. Even if such objects were able to suppress all galaxy formation, their X-ray emission would, however, make them highly visible if more than 2.5 percent of their mass was in ordinary matter. Such a low baryon density leads to insuffient cooling for galaxies to form in pancakes. A neutrino-dominated universe appears to conflict with observation irrespective of the details of the processes which govern galaxy formation.

    Description: Evidence that the galaxy A0945-30 is a new extragalactic X-ray source was found from an analysis of rotating modulation collimator quick-look data collected from the SAS 3 X-ray observatory. A strong piece of evidence was the hydrogen column density of 3 plus or minus 1 times 10 to the 22nd power per sq cm for a power law spectrum extending to about 35 keV inferred from HEAO 1 data. Observations indicate that the galaxy is an early type spiral with a bright nucleus. A faint star alongside the galaxy within the 90% confidence X-ray source error circle is an F dwarf with no indication of emission features. Spectra of the galaxy in the red and blue show sharp, intermediate strength emission lines of forbidden O III, forbidden N II, and H-alpha together with strong Mg, Fe, Ca absorption features characteristic of a normal stellar continuum in the galactic nucleus.
