    Notes: By the radiation-copolymerization method with polyethylene glycoldimethacrylate (PGD) as a main polymerizable reagent, microbial cells of Brevibacterium ammoniagenes were immobilized with high specific activity of NAD kinase and high mechanical strength. The reagents used for the immobilization such as PGD, polyvinyl alcohol (PVA), and N,N′-methylenebisacrylamide (Bis) did not inversely affect the enzyme activity. Freezing and irradiation treatment of the cell-reagent solution did not inactivate the enzyme either, but longer freezing time or a lower irradiation dose (less than 400 krad) resulted in the unsatisfactory mechanical strength of the immobilized cells. Almost all of NAD and ATP consumed were converted into NADP within three hours reaction time. The drum reactor was found to be ideal for the reaction of immobilized cells, since it gave little mechanical stress to the immobilized cells for the effective mixing of the cells and the substrates. The immobilized cells were subjected to three hours reaction repeatedly for 30 times without any activity loss.

    Notes: The cell cycle dependency of rice α-amylase production in Saccharomyces cerevisiae was investigated using synchronous and arrested cultures. The results of two separate synchronous cultures, using α-mating factor and a cdc28 mutant, indicated that the rice α-amylase-specific production rate is not constant during the cell cycle. The specific production rates during G1, S, and M phases were then ascertained by inhibiting the progression of the cell cycle using α-mating factor, hydroxyurea, and nocodazole, respectively. The specific production rate was found to be maximal during the M phase. The increase in the specific production rate during the M phase was confirmed from the accumulation of M-phase cells using a cdc15 mutant. The intracellular content of rice α-amylase was also measured during the cell cycle. Like the specific production rate, the intracellular content was found to fluctuate throughout the cell cycle, and to reach a maximum during M phase. © 1997 John Wiley & Sons, Inc.     Notes: The glycerinated stalks of the peritrich ciliate, Vorticella, can contract helically and reversibly on the addition of not only Ca2+ but also other divalent or trivalent cations having ionic radii not far from 1 Å. In order to investigate the stalk contraction quantitatively in the absence of Ca2+-chelators, we developed a method to eliminate contaminating Ca2+ and other metal ions in KC1 and pHbuffer solutions by using a Ca2+-and heavy metal ion-specific ion exchange resin (Eporas MX-2) Thus, it was possible to measure the relationship between the fractional stalk length of Vorticella and the free concentration of alkaline earth metal, transition metal, and lanthanide ions in the 0.1 M KC1 and buffer (pH 6.8) solutions. Among these ions, Ca2+, Nd3+, and Eu3+ (having ionic radii of about 1 Å) had the highest affinity for the contractile element in the spasmoneme. As the concentration of lanthanide ions (except Nd3+ and Eu3+) is increased, the Vorticella stalk contracts abruptly at a threshold level; this means that the Hill's parameter is very high, probably more than 6. The results of these experiments and of the co-mixtures of Ca2+ and Tb3+ suggest that a contractile element in the spasmoneme contains both contractile Ca2+-binding and regulatory Ca2+-binding sites.

    Description / Table of Contents: Copolymers containing hydantoin- and imide-groups were prepared using purified materials and applied to the surface of MOSFET (Metal Oxide Semiconductor Field Effect Transistor) in order to amplify the influence of copolymers on semiconductor devices. The influence of the polymers on the residual current of the device was analyzed with respect to impurity content, softening temperature of the polymers, composition ratios etc. It was found that the residual current increases with increasing flowing temperature of the polymer containing only imide groups, and with increasing content of hydantoin groups.
    Notes: Copolymere, die Hydantoin- und Imidgruppen enthielten, wurden hergestellt und auf die Oberfläche von MOSFET (Metalloxid-Halbleitereffekt-Transistor) aufgetragen, um den Einfluß des Copolymeren auf Halbleiteranlagen darzustellen. Der Einfluß der Polymeren auf den Reststrom der Anordnung wurde hinsichtlich des Gehalts an Verunreinigungen, der Erweichungstemperatur, der Zusammensetzung etc. untersucht. Es wurde beobachtet, daß der Reststrom bei Polymeren, die nur Imidgruppen enthielten, mit steigender Erweichungstemperatur sowie bei den Copolymeren mit steigendem Hydantoingruppengehalt ansteigt.

    Notes: Polyester urethane network elastomers with incorporated hard segment oligomers have been prepared by poly(ethylene adipate)glycol (PEA), 2,4-tolylene diisocyanate (TDI), and 1,4-butanediol (BD). These hard segment oligomers were hydroxy-terminated oligomers ([BD-TDI]n-BD; n=1,3), obtained by reacting BD with TDI. Concentrations of allophanate as a cross-linking site were determined by the amine degradation method. Hard segment moieties were obtained by a novel selective hydrolysis of soft segments in the elastomers. Molecular weight distributions of hard segment were measured by means of GPC. Mechanical and thermal properties were measured. Dependence of rubber elasticity on physical cross-linking between normal elastomers and the elastomers with incorporated hard segment oligomers were discussed.

    Notes: Metallopolymers in which bis(2,2′-bipyridine)ruthenium(II) complexes are coordinated to imidazolyl residues on partially quaternized poly(1-vinylimidazole) having different degree of quaternization (RuQPIm) were synthesized. The luminescence quenching of the photoexcited RuQPIms with various alkylviologens having different length of alkyl group was investigated in MeOH. In quenching of RuQPIm with methylviologen (MV2+), the efficiency of quenching decreases with increasing degree of quaternization. This was attributed to the electrostatic repulsion between the MV2+ species and the quaternized imidazolium residue on the polymer backbone and steric hindrance of the alkyl side chain. Furthermore, the efficiency of quenching depends on the length of the alkyl group in viologen. These results suggest that the alkyl side chain on the polymer backbone exerts steric hindrance at low degree of quaternization and van der waals interaction at high degree of quaternization.

    Notes: The acetonitrile effects on the quenching and photosensitized charge separation using two partially quaternized poly(1-vinylimidazole)-bound ruthenium(II) complexes, RuQPIm-19 and RuQPIm-44, wherein the numbers represent the degree of quaternization in molar percentage, have been investigated. The systems consist of RuQPIm-19 and RuQPIm-44 as photosensitizers, 1,1′-dimethyl-4,4′-bipyridinium (MV2+) and 1,1′-didodecyl-4,4′-bipyridinium (C12V2+) dications as electron acceptors (quenchers), and triethanolamine (TEOA) as a sacrificial electron donor. The photosensitized charge separation reaction proceeds through two processes, an interactive process and a direct process, i.e., the viologen is undergoing an interaction with these polymers and no interaction, respectively. The charge separation through the interactive process is less effective than that through the direct process, because the reaction through the interactive process is effective for both the forward and the back reactions. With the addition of acetonitrile, the rates of viologen radical formation decrease for RuQPIm-19 and RuQPIm-44, while the quenching efficiency decreases for RuQPIm-19 and increases for RuQPIm-44. In MV2+, the acetonitrile effect contributes to the interactive and direct processes for RuQPIm-19, while it contributes to only the direct process for RuQPIm-44, particularly the forward reaction for RuQPIm-19 and the back reaction for RuQPIm-44. In C12V2+, the interactive process through which the C12V2+ increases, leading to a decrease in the rates for viologen radical formation. Furthermore, these results indicate that the conformational changes in these polymers play an important role in the charge separation reactions.

    Notes: A glucose sensor employing ferrocene-bound glucose oxidase was fabricated by the screen printing technique. In order to obtain high GOD activity, glucose oxidase (GOD) was modified with ferrocenecarboxylic acid (fc) as an electron mediator. Simultaneous spectroscopic and electrochemical analyses showed that ferrocene-bound GOD (fc-GOD) displayed an increase in catalytic current. The ferrocene acts as an electron transfer relay between the active center of the enzyme and the gold electrode. The screen printing process was used to print successfully the unique fc-GOD containing ink onto the gold electrode patterned on the glass plate. The fc-GOD containing ink was specially developed to be suitable for the printing apparatus. The printed glucose sensor was shown to offer a stable calibration profile and stable electrochemical properties.

    Notes: Transforming growth factor beta (TGF-β), a multifunctional polypeptide growth factor, regulates the expression of many genes critical to cell cycle progression, such as members of the jun gene family which encode components of the transcription factor complex AP-1. The transforming proteins encoded by the early region 1A of adenovirus12 (Ad. 12-E1A) abrogate some of the cellular responses to TGF-β as well as affecting, differentially, the expression of cellular jun genes. Our data demonstrate that expression of Ad. 12-E1A in rat 3Y1 fibroblast cells inhibits induction of junB by TGF-β1 while not altering the regulation of junB by phorbol ester or serum. Regulation of c-jun gene expression by TGF-β1, phorbol ester, and serum is not appreciably altered by the expression of Ad. 12-E1A. Inhibition of TGF-β induced junB expression is not due to a defect in TGF-β/receptor interaction on Ad. 12-E1A transformed cells and is not observed in other isotypic fibroblast cells transformed by SV40 or polyomavirus. These data suggest that multiple, independent, intracellular signal transduction pathways exist which mediate genomic responses to TGF-β. Cellular expression of Ad. 12-E1A-12S gene products results in selective disruption of some TGF-β1 signaling cascades and not those activated by phorbol ester or serum. These data further suggest that some cellular targets which mediate TGF-β1 action may also be unique targets of action for the E1A-12S transforming protein of adenovirus12. © 1994 Wiley-Liss, Inc.
