    Notes: This work provides examples of some of the imaging capabilities of environmental scanning electron microscopy applied to easily charged samples relevant to particle analysis. Environmental SEM (also referred to as high pressure or low vacuum SEM) can address uncoated samples that are known to be difficult to image. Most of these specimens are difficult to image by conventional SEM even when coated with a conductive layer. Another area where environmental SEM is particularly applicable is for specimens not compatible with high vacuum, such as volatile specimens. Samples from which images were obtained that otherwise may not have been possible by conventional methods included fly ash particles on an oiled plastic membrane impactor substrate, a one micrometer diameter fiber mounted on the end of a wire, uranium oxide particles embedded in oil-bearing cellulose nitrate, teflon and polycarbonate filter materials with collected air particulate matter, polystryene latex spheres on cellulosic filter paper, polystyrene latex spheres “loosely” sitting on a glass slide, and subsurface tracks in an etched nuclear track-etch detector. Surface charging problems experienced in high vacuum SEMs are virtually eliminated in the low vacuum SEM, extending imaging capabilities to samples previously difficult to use or incompatible with conventional methods. © 1993 Wiley-Liss, Inc.

    Notes: Modification of a standard differential scanning calorimeter to allow irradiation of the sample material with ultraviolet light allows one to follow the course of polymerization of photocurable polymer systems. The rate and heat of polymerization are readily determined by this technique which gives it a number of very useful applications. As a quality control tool, one can use the heat of polymerization to monitor the number of reactive sites present in the prepolymer thus achieving a degree of control on the ultimate properties of the cured film. Similarly, analysis of a cured film will indicate the presence of any uncured material, thus allowing one to adjust curing conditions accordingly. The technique has numerous other applications in the study of photocurable resins, such as evaluation of initiator efficiencies, optimization of curing parameters (initiator concentration, lamp design), and the sensitivity of prepolymers to oxygen and wavelength.

    Notes: Quantitative Differential, Thermal Analysis has been used in the wire and cable industry for determining the degree of oxidative stability of polyolefins for quality control purposes in lieu of a direct antioxidant analysis, However, it has never been shown whether the presence of each additive in a stabilizer package could be detected by this method. The results presented here indicate that the presence of a primary antioxidant and a copper inhibitor in combination can be detected separately by comparing the oxidative induction times in copper and aluminum pans. The addition of a thioester synergist such as distearyl thiodipropionate, DSTDP, is evidenced by an increased induction time in aluminum pans. However, the presence of DSTDP makes it impossible by this method to determine whether an effective level of copper inhibitor is present since the sample shows a reduced stability on copper compared to aluminum. One of two factors can be invoked to explain these results: a) DSTDP interferes with the complexing ability of the copper inhibitor or b) the copper-inhibitor complex retards the synergistic effect of the DSTDP. In either case, this factor is not important at lower temperatures since oxygen uptake data at 130°C indicate that DSTDP is an effective synergist in the presence of copper.

    Notes: Neovascularization was studied in the chorioallantoic memebrane of the chick embryo after implantation of bovine aortic endothelial and smooth muscule cells, Swiss and BALB/c 3T3 cells and human diploid fibroblasts cultured separately on microcarrier beads. Quantitative analysis of neovascularization indicated a 3 1/2-fold increase in the number of blood vessels responding to endothelial cells while smooth muscle cells induced a twofold increase when compared to the response of beads without cells. Skin fibroblasts and Swiss 3T3 cells did not elicit a comparable response. The marked angiogenic response induced by endothelial cells was characterized by a 137% increase in total vessel length and a 35% increase in average vessel area when compared to controls. Two of the properties required for an angiogenesis factor - stimulation of cellular migration and proliferation - can also be demonstrated using endothelial cell-conditioned medium in cell culture systems. Medium from cultured bovine aortic endothelium stimulates DNA synthesis, proliferation, and migration of smooth muscle cells. In adition, conditioned media from both endothelial cells and smooth muscle cells produced an angiogenic response in the chorioallantoic membrane assay, which was comparable to that produced by intact cells growing on microcarrier beads. Similar responses were not evident with medium conditioned by other cell types. These results indicate the potential importance of endothelial cells and endothelial cell products in regulating blood vessel growth.

    Notes: In response to externally applied shear stress, Cultured endothelial monolayers develop prominent, axially-aligned, microfilamentous bundles, termed “stress fibers” (Dewey: Journal of Biomechanical Engineering 106:31-35, 1984; Franke et al.: Nature 81:570-580, 1984; Franke et al.: Klin. Wochenschr 64:989-992, 1986; Wechezak et al.: Laboratory Investigation 53:639-647, 1985). It is unclear, however, whether similar stress fibers develop in noncontiguous endo-thelial cells and whether these structures are necessary for adherence of individual cells under shear stress. It also is unknown what alterations occur in microtubules, intermediate filaments, and focal contacts as a consequence of shear stress. In this study, endothelial cells, free of intercellular contact, were exposed to 93 dynes/cm2 for 2 hr. With the aid of specific labeling probes and interference reflection microscopy, the distributional patterns of microfilaments, microtubules, intermediate filaments, and focal contacts were examined. Following shear stress, microfilament bundles and their associated focal contacts were concentrated in the proximal (relative to flow direction) cell regions. In contrast, microtubules were distributed uniformly within cell contours. Intermediate filaments displayed only an occasional tendency for accumulation at proximal edges. When cells were shear-tested in the presence of cytochalasin B to inhibit microfilament assemly, considerable cell loss occurred. Following inhibition of tubulin polymerization, no increase was observed in the percentage of cells lost due to shear over nontreated controls. Nocodazole-treated cells, however were characterized by prominent stress fibers throughout the cell. These results indicate that stress fiber and focal contact reorganization represent major responses in isolated endothelial cells exposed to shear stress and that these cytoskeletal structures are necessary for adherence.

    Notes: A transmission-type attachment for energy-dispersive x-ray fluorescence analysis was developed. The attachment was designed for analysis to be carried out in a CAMEBAX microanalyzer, where the energy-dispersive spectrometer detector views the specimen through a hole in the objective lens and the space between the specimen and the pole piece of the objective lens is fairly small. Nevertheless, this type of attachment can be applied in other microanalyzers or scanning electron microscopes. Detection limits between 0.5 and 5 ppm on SRM 612 glass (NIST) and the applicability to the analysis of small particles (SRM 470 glass sphere 90 μm in diameter) are demonstrated.
