    Description / Table of Contents: Synthesis of 1,2,3,3,6,6-Hexamethyl-l-cyclohexene - A Cascade of C12H+ Carbenium Ion Rearrangements6-Chloro-2,6,7,7-tetramethyl-2-octene (4), prepared via prenyl chloride addition to 2,3,3-trimethyl-1-butene, at room temperature undergoes a zinc chloride/hydrogen chloride-catalysed cyclocondensation to yield 1,2,3,3,6,6-hexamethyl-1-cyclohexene (1). The reaction mechanism is elucidated by isolation of intermediate five-membered carbocycles and by force field calculations.

Der Reaktionsmechanismus wird durch die Isolierung intermediär auftretender fünfgliedriger Ringe und durch Kraftfeldrechnungen aufgeklärt.

    Description / Table of Contents: The ultraviolet absorption spectra of fractionated cellulose acetate of varying degrees of polymerisation were measured. In the region between 3000 and 3700 Ä the absorption coefficients were found nearly linear to the wave-length of the light absorbed.The change of the absorption coefficient with increasing wave-length is nearly the same for different degrees of polymerisation.
    Notes: Es wurde das UV-Absorptionsspektrum gereinigter Celluloseacetatfraktionen verschiedenen Polymerisationsgrades bestimmt. Im Bereich von 3000 bis etwa 3700 ÄE nimmt der Absorptionskoeffizient ungefähr linear mit der Wellenlänge des eingestrahlten Lichtes ab.Die Änderung des Absorptionskoeffizienten ist bei Fraktionen verschiedenen Polymerisationsgrades für gleiche Änderungen der Wellenlänge etwa gleich groß.

    Notes: In 1956 L. H. Tung has given a quite empirical and formal mathematical relationaship, by which - appropriate plotting of data provided - it is possible to represent the distribution of DP values, as obtained by a completed polymerization (polycondensation or polyaddition), by means of a straight line. In the course of the present publication it is shown that the authors - starting from a quite different problem - have found a relationship that proves (after an insignificant simplification and by suitable interchange of symbols) to be identical with Tung's equation. According to some special ideas about the chemism, the method of deducing this equation enables the authors to interpret Tung's equations in terms of kinetics. The basic equations obtained are \documentclass{article}\pagestyle{empty}\begin{document}$$ dN/dt = - 2kNt \ \ \ {\rm and} \ dN/dP = - 2AN \left({P - 1} \right)$$\end{document} These show that the reaction rate is merely proporional to the number N of monomers and to the time t. The chain lengths of the macromolecules are suggested to be only an appropriate measure for the time elapsed from the beginning of their formation. Interpreting Tung's equation kinetically it is possible to get information about the distribution of DP values that is to be expected at a given temperature. With regard to theoretical considerations one expects the integral number 2 to be used for the exponent b in the modified equation originally given by Tung: \documentclass{article}\pagestyle{empty}\begin{document}$$N = N_{0} e^{ - A \left( {P - 1} \right)^b}$$\end{document} This value is related to the fact that the chain growth takes place in two directions because of the bifunctionality of the monomers. Therefore, the termination of the reaction will also take place in two steps. The correctness of these suggestions is confirmed by the fact that (for all known fractionations of polycondensation products) the exponent b proves to be nearly 2. Using the value b = 2 the whole chain lengths distribution of normal polycondensation products can be easily calculated by means of one single number or weight-average value of the DP as determined with an unfractionated sample.

    Notes: It is hypothesized that nuclear DNA is organized in topologically constrained loop domains defining basic units of higher order chromatin structure. Our studies are performed in order to investigate the functional relevance of this structural subdivision of eukaryotic chromatin for the control of gene expression. We used the chicken lysozyme gene locus as a model to examine the relation between chromatin structure and gene function. Several structural features of the lysozyme locus are known: the extension of the region of general DNAasel sensitivity of the active gene, the location of DNA-sequences with high affinity for the nuclear matrix in vitro, and the position of DNAasel hypersensitive chromatin sites (DHSs). The pattern of DHSs changes depending on the transcriptional status of the gene. Functional studies demonstrated that DHSs mark the position of cis-acting regulatory elements. Additionally, we discovered a novel cis-activity of the border regions of the DNAasel sensitive domain (A-elements). By eliminating the position effect on gene expression usually observed when genes are randomly integrated into the genome after transfection, A-elements possibly serve as punctuation marks for a regulatory chromatin domain. Experiments using transgenic mice confirmed that the complete structurally defined lysozyme gene domain behaves as an independent regulatory unit, expressing the gene in a tissue specific and position independent manner. These expression features were lost in transgenic mice carrying a construct, in which the A-elements as well as an upstream enhancer region were deleted, indicating the lack of a locus activation function on this construct. Experiments are designed in order to uncover possible hierarchical relationships between the different cis-acting regulatory elements for stepwise gene activation during cell differentiation. We are aiming at the definition of the basic structural and functional requirements for position independent and high level gene expression. The result of these experiments will have important consequences for random gene transfer with predictable and reproducible expression of transgenes.
