    Notes: To investigate what role homeotic genes may play in morphological evolution, we are comparing homeotic gene expression in two very different insects, Drosophila (Diptera) and Schistocerca (Orthoptera). In this paper we describe a monoclonal antibody, FP6.87, that recognizes the products of both the Ultrabithorax (Ubx) and abdominal-A (abd-A) genes in Drosophila, via an epitope common to the carboxy terminal region of these two proteins. This antibody recognizes nuclear antigens present in the posterior thorax and abdomen of Schistocerca. We infer that it recognizes the Schistocerca homolog of UBX protein, and probably also of ABD-A. As the distribution of Schistocerca ABD-A protein is already known, we can use this reagent to map the expression of Schistocerca UBX in the thorax and anterior abdomen, where ABD-A is not expressed. Both the general domain, and many of the details, of UBX exp ression are remarkably conserved compared with Drosophila. Thus UBX expression extends back from T2 in the ectoderm (including the CNS), but only from A1 in the mesoderm. As noted for other bithorax complex genes in Schistocerca, expression begins in the abdomen, at or shortly before the time of segmentation. It only later spreads anteriorly to the thorax. For much of embryogene-sis, the expression of UBX in the thoracic epidermis is largely restricted to the T3 limb. Inthis limb, UBX is strikingly regulated, in a complex pattern that reflects limb segmentation.Reviewing these and earlier observations, we conclude that evolutionary changes affect both the precise regulation of homeotic genes within segments, and probably also the spectrum of downstream genes that respond to homeotic gene expression in a given tissue. Overall domains of homeotic gene expression appear to be well conserved between different insect groups, though a change in the extent and timing of homeotic gene expression may underlie the modification of the posterior abdomen in different insect groups. © 1994 Wiley-Liss, Inc.

    Notes: The glycoprotein hormone human chorionic gonadotropin (hCG) is synthesized in large quantities by the developing placenta, reaching peak concentrations in maternal blood during the late first trimester and early midtrimester of pregnancey. In general it is believed that the β-subunit of this dimeric hormone is expressed in pituitary gonadotropes, thyrotropes, and trophoblasts, while the β-subunit is expressed exclusively by trophoblasts. Studies from our laboratory and other laboratories have shown that some midtrimester human fetal tissues, in addition to the placenta, can synthesize proteins that appear to be very similar to the β-subunit of hCG. To define precisely the nature of this putative hCG-β-subunit in extraplacental fetal tissues, we have examined the mRNA from a variety of human fetal and adult tissues using nucleic acid hybridization and reverse transcription-polymerase chain reaction (PCR) methods. Our results demonstrate that midtrimester fetal kidney and adrenal tissues contain hCG-β mRNA transcripts at concentrations comparable to that of placenta, while fetal lung, brain, muscle, and adult adrenal contain only trace to undetectable levels of hCG-β mRNA. By restriction endonuclease mapping of PCR fragments from fetal tissue cDNAs, we show that the hCG-β transcript expressed in midtrimester human fetal organs is a bone fide copy of hCG-β gene No. 5 of the β-subunit gene family located on chromosome 19. © 1992 Wiley-Liss, Inc.

    Notes: More than 90% of epithelial ovarian cancers arise from single cells. Malignant transformation can be associated with a number of molecular alterations including upregulation of tyrosine kinases and phosphatases, physiologic activation of ras, mutation of p53, amplification of myc, and increased activity of matrix metalloproteinases 2 and 9. Proliferation of transformed epithelial cells can be enhanced through the persistence of autocrine growth stimulation by TGF-α, loss of autocrine growth inhibition by TGF-β, as well as paracrine growth stimulation by macrophage derived cytokines and OCAF, a novel lyso-phospholipid. Ascites tumor cells retain responsiveness to growth inhibition by TGF-β which induces apoptosis in malignant ovarian epithelial cells, but not in normal ovarian surface epithelium.Proliferation of surface epithelial cells following ovulation my contribute to the pathogenesis of ovarian cancer. Use of oral contraceptives that suppress evulation has been associated with reduced risk of ovarian cancer in later life. Retinoids also deserve further evaluation for chemoprevention. Treatement with fenretinide was associated with decreased incidence of ovarian cancer. Additive or synergistic inhibition of ovarian tumor cell proliferation has been observed with TGF-β in combination with all-trans-retinoic acid.Early detection of ovarian cancer could improve survival. Transvaginal sonography (TVS) and serum markers such as CA-125 have been evaluated in multiple clinical trials. The former lacks adequate specificity, whereas the latter is not sufficiently sensitive. Use of multiple serum markers can improve sensitivity. A combination of CA-125, M-CSF and OVX-1 has detected 〉 95% of Stage I ovarian cancers. If similar results are obtained with different data sets, multiple serum markers could be used to trigger the performance of TVS, providing a potentially cost effective screening strategy. Prospective trials will be required to demonstrate that screening for early stage ovarian actually impacts on survival.

    Notes: ABA block copolymers were prepared by the anionic polymerization of α-methyl-α-butyl-β-propiolactone, MBPL (B block), and pivalolactone, PL (A blocks). The MBPL block had a very low decree of crystallinity and a glass temperature of - 13°C, so phase separation with extensive crystallization of the PL blocks gave thermoplastic elastomers when the MBPL block constituted the principal and continuous phase. The observed crystallinity and melting point of 40-45°C in the MBPL homopolymer have not been previously reported. Measurements were obtained by electron microscopy of the initial size distribution of the PL domains as a function of copolymer composition and degree of polymerization, and on the effect of annealing on this parameter. Tensile strengths and elongations at break were both less than those previously observed for equivalent ABA block copolymers of PL and α-methyl-α-propyl-β-propiolactone.

    Notes: The central theme of this paper revolves about an attempt to provide information and ideas that might be useful to persons interested in cryogenic dental procedures. However, a broad spectrum of thermal properties is also considered. For convenience a summary table is given for pertinent thermal and mechanical properties of dental materials. A point demonstrated is that observations which are of direct empirical utility to dentists often can be enhanced in value if viewed in terms of simple analytical models. For example, a tooth can be expected to be somewhere between a sphere and a long rod in its cooling or heating characteristics. The importance of the thermal diffusivity rather than thermal conductivity in transient phenomena is discussed. Some graphical techniques that circumvent mathematical difficulties are developed in detail. An isolated extracted tooth in contact with a coolant approaches to within 98% of its final temperature in less than 1 min but in the jaw, in contact with a cold (or hot) probe, the equilibration will take much longer. Thermally generated stresses, either due to differences in expansivities of the dentin and enamel or to large temperature gradients across a single component, can be quite large (comparable to the reported tensile strength, 9.3 MN/m2 for enamel) at cryogenic temperatures. Our observations and analysis make us believe that the reported strength of 9.3 MN/m2 is too low by at least an order of magnitude.

    Notes: An analytical technique based on the method of least squares has been developed to fit experimental second virial coefficients to the functional form derived from the Lennard-Jones (6-12) potential. Both the functional form and the normal equations of the least squares fit are rigorously derived. Because of the implicit, nonlinear dependence upon one of the adjustable parameters, a computer solution of the normal equations is required. Second virial coefficient data for a large number (60) of materials, including hydrocarbons, halides, alcohols, and cyclic compounds, have been extracted from published sources, carefully evaluated for consistency, and fitted to the Lennard-Jones potential function.Force constants for these materials as determined from fitting the data in this uniform manner have been found to be related to structural parameters. The final correlation, with the potential well-depth related to critical temperature and the collision diameter calculated from a set of group contributions with appropriate corrections for polarity and/or association, produces average deviations between computed and experimental second virial coefficients of 10 to 15% or 100 cc./g.-mole, whichever is greater. In many cases the agreement is much better.

    Notes: Nonequilibrium diffusion behavior in nonionic and ionic surfactant microemulsion systems has been studied experimentally using the open-ended capillary method. Experimental results for these systems have been compared with a drop theory of diffusion for microemulsions under conditions where large concentration and electrostatic gradients exist. The results show good agreement in concentration profiles between theory and experiment for the microemulsion components - water, benzene, and phenol. Furthermore, under certain conditions the theory predicts that over a limited time interval phenol will diffuse from low-concentration regions to regions of higher concentration. This phenomenon has been observed.

    Notes: Solution rates of 1/2-, 3/4-, and 1-in. cast benzoic acid spheres were measured under natural-and forced-convection conditions. Upward-flowing streams of water and aqueous propylene glycol, in laminar and turbulent flow, were used to contact single spheres in either a 1.50- or 4.00-in.- diameter column. Sphere Reynolds numbers ranged from 10 to 16,920; Schmidt numbers from 987 to 69,680; sphere- to column-diameter ratios, ds/dc, from 0.123 to 0.497; and Grashof numbers from 5,130 to 125,200.The Sherwood number was found independent of the laminar-turbulent transition for pipe flow and the sphere- to column-diameter ratio when the Reynolds number is based on the average fluid velocity and sphere diameter.A correlation of the data based on the additivity of the natural and forced convection processes is proposed, and statistical analyses of the new experimental data result in equations which correlate other heat and mass transfer data for single spheres immersed in bounded and free-jet streams of gases and liquids for NRe, S from 1 to 30,000 and NSe from 0.6 to 3,000, within an average deviation of about 20%.
