    Notes: This work describes a Pd-catalyzed coupling of ferrocene alkyne derivatives to iodo amino acids. Ferrocene carboxylic acid propargyl amides were easily obtained in high yield. The crystal structures of the propargyl amine derivative 3 and the 1,1-diethylpropargylamine derivative 4 have been determined by X-ray diffraction. Pd-catalyzed coupling to p-iodoanilide amino acids gave the corresponding ferrocene-labeled amino acid derivatives, which were easily purified by diethyl ether extraction in the case of the 1,1-diethyl derivatives 8. The coupling reaction did not require anhydrous solvents and tolerated a variety of functional groups present in peptides such as alcohols (8a, Ser), thioethers (8d, Met), disulfide bonds (cystine, 12) esters (as in the N-labeled Leu derivative 10) and of course amides. A minor by-product of the coupling reaction, namely the homo-dimer bis(ferrocene carboxylic acid propargylamide) 9, was identified in the crude reaction mixtures by mass spectrometry and independently synthesized by oxidative coupling (Glaser and Eglington) of 3. All new compounds were completely characterized spectroscopically, including 15N- and 2D NMR spectroscopy, Mössbauer spectroscopy and electrochemistry. This work introduces a versatile procedure for a selective functionalization of amino acids with organometallics at the C-terminus which is expected to be of general applicability to peptide chemistry.

    Notes: A series of Fe4S4 cluster compounds, I, III, and V, in which the cuboidal cluster core is appended with four crown ether thiolate ligands, and II and IV, bearing thiolate ligands without crown ether parts, has been synthesized and characterized. The spectroscopic and electrochemical properties of these compounds are determined by the electronic nature of the thiolate ligands. Only in the case of III, where a very short α-thioacetyl linker was used to connect the crown ether ligands to the cluster core, was a restricted conformational freedom of the ligand observed. A detailed electrochemical study of the influence of alkali and earth alkali metal ions (Li+, Na+, K+, Mg2+, and Ba2+) on the reversible 2-/3- reduction of the cluster compounds was performed. In the case of the crown ether appended clusters I, III, and V, the addition of these metal ions resulted in an anodic shift, i.e. in positive direction, of the reduction potential (modulation effect) and to larger current responses (promotion effect). The magnitude of the modulation effects is determined by the binding affinity of the metal ions in the crown ether ligands, and by the distance between bound metal ions and the redox active cluster core. Variation of the linker between the cluster core and the metal ion binding site resulted in cluster compounds with almost inverse selectivities for e.g. K+ and Ba2+ in the case of I and III. For the large effects found for compound I a lariat binding mode is proposed.

    Notes: Skin tissue may be engineered in a variety of ways. Our cultured skin substitute (Graftskin, living skin equivalent or G-LSE), Apligraf™, is an organotypic culture of skin, containing both a “dermis” and “epidermis.” The epidermis is an important functional component of skin, responsible for biologic wound closure. The epidermis possesses a stratum corneum which develops with time in culture. The stratum corneum provides barrier function properties and gives the LSE improved strength and handling characteristics. Clinical experience indicated that the stratum corneum might play an important role in improving the clinical utility of the LSE. Handling and physical characteristics improved with time in culture. We examined the LSE at different stages of epidermal maturation for barrier function and ability to persist as a graft. LSE grafted onto athymic mice before significant development of barrier function did not withstand bandage removal at 7 days postgraft. LSE grafted after barrier function had been established in vitro were able to withstand bandage removal at day 7. Corneum lipid composition and structure are critical components for barrier function. Media modifications were used in an attempt to improve the fatty acid composition of the stratum corneum. The barrier developed more rapidly and was improved in a serum-free, lipid-supplemented condition. Lipid lamellar structure was improved with 10% of the stratum corneum exhibiting broad-narrow-broad lipid lamellar arrangements similar to human skin. Fatty acid metabolism was not appreciably altered. Barrier function in vitro was 4- to 10-fold more permeable than human skin. Epidermal differentiation does not compromise engraftment or the wound healing ability of the epidermis. The stratum corneum provides features beneficial for engraftment and clinical use. © 1996 John Wiley & Sons, Inc.

    Notes: The structure and physical properties of optically active, metal-free 2,3,9,10,16,17,23,24-octa (S-3,7-dimethyloctoxy)phthalocyanine ((S)-Pc(8,2)) are reported and compared with those of the phthalocyanine with (R,S) side chains (mixture of 43 stereoisomers). Unlike the latter compound, (S)-Pc(8,2) lacks a crystalline phase. A freshly prepared sample is in a distorted mesophase and reorganizes irreversibly to a more ordered phase above 65 °C. X-ray diffraction and circular dichroism studies indicate that the molecules are stacked in columns which have a hexagonal arrangement and a left-handed helical superstructure, that is, a novel chiral Dh* mesophase. Solid state NMR measurements reveal that the phthalocyanine units in the columns begin to vibrate laterally when the temperature is increased. At 111 °C (Dh* → Dr transition) they start to rotate around their columnar axes and at the same time the side chains become liquidlike. Energy migration is very efficient in the chiral Dh* phase and also in the frozen mesophase below 3 °C, as follows from luminescence spectroscopy. Intracolumnar charge transport, studied by the time-resolved microwave conductivity technique, turns out to be slower in the helically distorted columns than in linear columns. (S)-Pc(8,2) forms a very stable bilayer at the air-water interface, which can be transferred to give a high quality Langmuir-Blodgett film. The fact that this phthalocyanine is mesogenic at room temperature is thought to be responsible for this behavior.

    Notes: Selective NMR decoupling and nuclear Overhauser effect (NOE) experiments with phycocyanobilin (PCB) show proton-proton interactions between the terminal rings A and D, viz. the chiral C(2) methine center and the ethyl substituent at C(18), as a result of the helical conformation of this open-chain tetrapyrrole in solution. Quantitative NOE measurements and a combination of force-field and semiempirical calculations (FSC) afford inter-proton distances across the helical gap of 4.2-4.6 (NOE) and 3.2-4.2 A° (FSC). The NOE and FSC, in conjuction with a qualitative evaluation of the steric interactions in the two optimized helices, suggest furthermore that, in solution, the helix M is somewhat more stable than P. The coexistence of at least two diastereoisomers is corroborated also by the circular dichroism (CD) spectra of PCB in MeOH/EtOH which point to a temperature-dependent equilibrium in solution, and by a considerable increase of this CD upon changing the solvent from the achiral alcohols to ethyl (-)-(S)-lactate which reflects a selective solvent-induced CD differentiating between diastereoisomers.

Analogous Vilsmeier additions to 1-methoxy-1-penten-3-yne lead to 3-halogen-2-methyl-substituted unsymmetrical pentamethinium salts 13 and 14. Symmetrical 3-methoxy-and 3-methylthio-substituted vinamidinium salts 15 and 16 are observed by appropriate Vilsmeier additions to 1-(dimethylamino)-1-buten-3-yne (4). Further 3-substituted unsymmetrical and symmetrical pentamethinium salts are obtained by nucleophilic substitution at the C-3 position of these salts, leading to 19-27. The [C4 + C1] pathway to 3-substituted glutaconaldehyde derivatives represents a useful completion to their synthesis.

    Notes: A theoretical analysis was performed to quantify the π-electron-accepting ability of the boron atom in ethynylboranes. An expansion technique was employed which permits to obtain a set of localized bonding schemes and their weights from a delocalized molecular orbital determinantal wavefunction. The derived manifold of bonding schemes is close to the classical resonance hybrid used in organic chemistry (valence-bond description). We quantified the π-electron transfer into the empty π-orbital of the boron atom by investigating nine model compounds where substituents with π-electron-donating ability are adjacent to a boron atom. This led to an ordering of the substituents according to their electron-donating ability towards boron. The boron atom hesitates to accept π-electrons from the ethynyl group in ethynylboranes in particular when good π-donors like amino groups are present. The π-electron donation from the vinyl group to the adjacent boron centre is slightly stronger than from the ethynyl group. Nitrogen lone-pair electrons are easily transferred to a neighbouring boron centre. Bonding schemes and their weights are in line with computed bond lengths and rotational barriers. Moreover, our theoretical results rationalize previous NMR and X-ray experiments and are in line with the reactivity of related compounds. It is demonstrated that bond lengths alone do not necessarily correlate with the degree of π-bonding and should be discussed with caution. The analysis is substantiated by showing that weights for covalent bonding schemes, as obtained from the simple restricted closed-shell MO determinant, correlate with bond strengths. Furthermore, a correlation of bonding-scheme weights with quantities based on the fragment orbital approach is presented. This novel correlation elucidates molecular properties which determine the extent of the π-electron transfer to the boron atom and permits a quantitative interpretation and prediction of intramolecular π-bonding.

    Notes: Two novel phosphane ligands 3 and 4 based on the rigid diphenylglycoluril molecule have been synthesized and characterised. Binding studies with 3 and 4, using 1,3-dihydroxybenzene derivatives reveal that ligands 3 and 4 behave similarly to clip molecule 5, which has the same binding site as ligands 3 and 4. The size of the flexible spacers in the ligands has been varied and the effect of this variation on the association constant of resorcinol derivatives has been determined. These cavity-containing ligands are able to coordinate two transition metal centres, leading to bimetallic macrocycles. The metallamacrocycles formed from 4 containing platinum or rhodium bind the guest, olivetol (5-pentylbenzene-1,3-diol), almost four times as strongly as the free tetrapodant 4. Complexes of 4 having palladium centres display similar or reduced binding affinities for resorcinol derivatives, when compared to free 4. Metal complexes of ligand 3 do not form host-guest complexes, probably because of a too small a ring-size of the metallamacrocycle.

    Notes: Asymmetrically substituted, liquid-crystalline phthalocyanines were synthesized. They contain one methoxy, six dodecyloxy and one acryloyloxy- or methacryloyloxydodecyloxy groups. The latter functions were polymerized under free radical conditions to give polymers with molecular weights between 9000 and 47000. The aggregation behaviour of the phthalocyanines in the polymer side chains was studied in solution by absorption and fluorescence spectroscopy. An enhanced intramolecular interaction between the phthalocyanines was observed. In the solid state this interaction is reflected in the spontaneous formation of a columnar structure. The polymers, however, do not display liquid-crystalline behaviour. Temperature-dependent luminescence measurements (at 4,2 - 300 K) were performed and the results are explained by the presence of an efficient energy migration process through the stacked phthalocyanine units.
