    Notes: The molecular weight of peptides can be determined by fast-atom bombardment mass spectrometry (FAB-MS) following polyacrylamide gel electrophoresis (PAGE). Initial results combining the fast and efficient separation of peptides by PAGE and the unambiguous determination of molecular weights by FAB-MS have been demonstrated for the three peptides bradykinin, neurotensin and gramicidin S. This method has also been applied to the determination of the molecular weights of two fragments from the tryptic digest of horse-heart cytochrome c.

    Notes: The formation of non-covalently bound inclusion complexes between ß-cyclodextrin and peptides containing aromatic acid residues has been confirmed by elcctrospray ionization mass spectrometry. This preliminary study has indicated that phenylalanine and tryptophan residues can form 1:1 complexes with ß-cyclodextrin whereas no evidence for such a complex could be found for tyrosine residues. Bradykinin, which contains two phenylalanine residues, formed both 1:1 and 1:2 complexes.

    Notes: The aim of this work was to probe the stereochemistry of a series of monosaccharides. Firstly, the underivatized sugars were investigated using linked-scans and tandem mass spectrometry on a ‘hybrid’ instrument (hybrid MS/MS) and secondly, a comparison of linked-scan and hybrid MS/MS was carried out on the butyl and phenyl boronate di-esters. In situ derivatization of the sugar to the boronate di-ester allows a negative-ion fast-atom bombardment (FAB) investigation with greater sensitivity than for the underivatized sugar. The linked-scan data allow a great deal of information on the structure and chemistry of the sugars to be obtained, but the sensitivity at lower mass is limited. The hybrid MS/MS data give similar fragmentation information, plus improved sensitivity at lower molecular weight.

    Notes: Tandem mass spectrometric (MS/MS) techniques have been widely used for the differentiation of isomeric compounds, since their spectra may show differences sufficient to distinguish between them. There are several different ways by which the MS/MS data can be obtained depending on the energies of the ions and the collisions. In this paper MS/MS spectra have been obtained for a group of isomeric bile acids using: 1, low-energy ions and low-energy collisions in a triple quadrupole mass spectrometer by liquid chromatography/MS/MS; 2, high-energy ions and low-energy collisions in a hybrid mass spectrometer by fast-atom bombardment MS/MS. Liquid chromatography combined with tandem mass spectrometry (LC/MS/MS) has also been used to identify the bile acids present in biological matrices such as bile extracts.

    Notes: The oximes of monosaccharides have not been well characterized by mass spectrometry. Conventional electron ionization (EI) experiments have supported the structures of these compounds but the data produced are usually equivocal (Finch and Merchant, JCS Perkin 1, 1682 (1975)). The application of fast-atom bombardment mass spectrometry to monosaccharide oximes gives molecular weight information. However, little fragmentation is observed and it is not possible to distinguish between stereoisomers. Using fast-atom bombardment tandem mass spectrometry experiments, we have produced daughter-ion spectra which are sufficiently different to enable us to distinguish between the stereoisomers of the oximes of monosaccharides.

    Notes: There are numerous examples of the regular segregation of achiasmate chromosomes at meiosis I in Drosophila melanogaster females. Classically, the choice of achiasmate segregational partners has been thought to be independent of homology, but rather made on the basis of availability or similarities in size and shape. To the contrary, we show here that heterochromatic homology plays a primary role in ensuring the proper segregation of achiasmate homologs. We observe that the heterochromatin of chromosome 4 functions as, or contains, a meiotic pairing site. We show that free duplications carrying the 4th chromosome pericentric heterochromatin induce high frequencies of 4th chromosome nondisjunction regardless of their size. Moreover, a duplication from which some of the 4th chromosome heterochromatin has been removed is unable to induce 4th chromosome nondisjunction. Similarly, in the absence of either euchromatic homology or a size similarity, duplications bearing the X chromosome heterochromatin also disrupt the segregation of two achiasmate X chromosome centromeres. Although heterochromatic regions are sufficient to conjoin nonexchange homologues, we confirm that the segregation of heterologous chromosomes is determined by size, shape, and availability. The meiotic mutation Axs differentiates between these two processes of achiasmate centromere coorientation by disrupting only the homology-dependent mechanism. Thus there are two different mechanisms by which achiasmate segregational partners are chosen. We propose that the absence of diplotene-diakinesis during female meiosis allows heterochromatic pairings to persist until prometaphase and thus to co-orient homologous centromeres. We also propose that heterologous disjunctions result from a separate and homology-independent process that likely occurs during prometaphase. The latter process, which may not require the physical association of segregational partners, is similar to those observed in many insects, in Saccharomyces cerevisiae and in C. elegans males. We also suggest that the physical basis of this process may reflect known properties of the Drosophila meiotic spindle. © 1993 Wiley-Liss, Inc.

    Notes: A number of sulphonyl hydrazones and derivatives have been synthesized and tested for biological activity as pesticides during the crop protection research programme at the Hatfield Polytechnic. A recent comparative ionization study of some of these compounds using electron impact (EI), fast atom bombardment (FAB) and various chemical ionization methods showed FAB mass spectrometry to be the optimum technique to use in terms of molecular weight information obtained. This study compares the FAB mass spectral data in positive and negative ion mode using an alternating positive and negative ion detection system.

    Notes: The pKBH+ values of the conjugate acids of N-phenylhydroxylamines change drastically when the pKBH+ values are determined in dimethyl sulfoxide (DMSO) rather than methanol. The Hammett ρ values change from -5·69 to -1·20 on going from methanol to DMSO for protonated N-phenylhydroxylamines, in contrast to a shift of -4·70 to -4·83 for protonated N-methylanilines in the same two solvents. This large change in susceptibility indicates that the species from which the proton departs is not the same for protonated N-phenylhydroxylamines in the two solvents. Experimental and computational evidence supports ionization of the H+ from the O atom for the protonated N-phenylhydroxylamines in DMSO.
