    Notes: In a previous paper, the presence in human kidney vein tissue of a novel blood group A heptaglycosylceramide based on the type-3 carbohydrate chain GalNAcαl-3(Fucαl-2)Galα1-3GalNAcαl-3(Fucα1-2)Galβl-4Glcβl-1 Ceramide, was suggested based on thin-layer immunostaining and electron ionization mass Spectrometry. Ions corresponding lo a structure containing two deoxyhexoses, two hexosamines and three hexoses were identified, but no information was obtained from mass spectrometry concerning the carbohydrate sequence5. In the present paper, we report the identification of carbohydrate sequence ions corresponding to a type-3 chain A heptaglyco-sylceramide by electron ionization-tandem mass spectrometry of a permethylated-reduced glycosphingolipid mixture isolated from human kidney vein tissue. The use of a microchannel-plate-array detector increased the sensitivity for collision-induced dissociation spectra by a factor of at least ten over a conventional electron multiplier.

    Notes: Multiple clustering of ammonia molecules to triply protonated bovine insulin B chain has been observed in a novel collision cell incorporated as part of a triple quadrupole mass spectrometer. The nature of the phenomenon was examined as a function of both the collision energy and the collision gas thickness in the cell. Increased clustering was observed with higher collision-gas thickness and lower collision energy. Initial experiments indicate that the number of ammonia molecules clustering to the precursor ion may be dependent on the number of available acidic sites.

    Notes: Fluoresence resonance energy transfer (FRET) microscopy is used to observe structures of DNA aggregates formed during electrophoresis in ultradilute hydroxyethylcellulose (HEC). FRET gives evidence of aggregates with a tightly packed core and random entanglement of DNA in the outer portions. HEC concentrations also appear to play a role in aggregate formation. DNA aggregates take longer to assemble in higher HEC concentration solutions and assume a less compact form than those formed at lower HEC concentrations.

    Notes: Electric birefringence imaging (EBI) maps stresses in agarose electrophoresis gels. Both DNA and gel alignment can be monitored using this technique. Previously, we have reported birefringence images associated with resolved small (〈 48 kbp) and partially resolved large 48 kb - (2 Mbp) DNA fragments oriented with relatively short electric field pulses. In this paper, differences between images acquired using short and long (DC) voltage pulses are presented. It is shown that stress signals distribute over long distances when exposed to prolonged electric fields. Resolution also affects band shapes. Refinements in instrumental design and evaluation of limiting noise sources of the technique are also described.

    Notes: Bacitracin is a mixture of polypeptide antibiotics produced by the action of Bacillus subtilis and Bacillus licheniformis. The mixture has previously been shown to contain at least ten components, of which only the major component, Bacitracin A, and its oxidized counterpart, Bacitracin F, have been fully structurally evaluated. Using fast atom bombardment ionization with tandem mass spectrometry, three isobaric components of one of the minor constituents of the mixture, Bacitracin B, have been structurally characterized, delineating the three single substitution sites within Bactitracin A where Ile has been replaced by Val.

    Notes: A combination of mass shifts arising from known structural variations of neutral precursors, and accurate mass measurements of key fragment tions, has led to a proposed systematization of the low-energy fragmentation mechanisms for MH+ ions of actinomycins. These rationalizations are used in a predictive manner to interpret fragment ion spectra, of unknown impurities in actinomycin standards, in terms of structure. Both the strengths and limitations of this approach are emphasized. This work has uncovered actinomycin variants in which one of the threonine residues is substituted by serine, a structural variation which does not appear to have been reported previously.

    Notes: The bacitracin antibiotic peptide complex has been analyzed using liquid chromatography in conjunction with ion spray tandem mass spectrometry and multiple reaction monitoring techniques. Fragmentation patterns were determined from the product ion spectrum of bacitracin A (mol. wt 1421.8 Da), a compound of well-established structure. Primary structural information has been determined for seven minor components of the bacitracin complex with regard to the extent of amino acid substitution taking place.

    Notes: The vanadate-sensitive Mg2+-dependent ATPase activity of the human erythrocyte ghost is believed to be involved in the shape change events that convert echinocytic ghosts to smoothed forms (biconcave discs and stomatocytes). At physiological salt concentration, pH 7.4, 2 mMATP, 5 mM Mg2+ and 1 mM EGTA, the Mg2+-ATPase activity of ghosts was inhibited strongly by millimolar concentrations of sodium fluoride: I50 = 1.31 ± 0.2, mM (mean ± S.D., n = 12). The addition of aluminium chloride to 15 μM reduced the concentration of NaF required for 50% inhibition to 0.76 ± 0.21 mM (n = 10). Aluminium alone had only a small inhibitory effect of the ATPase activity (13 ± 9 %; n = 10). Desferrioxamine, a strong chalator of tervalent aluminium ion, failed to reverse the inbibition by fluoride and reversed the inhibition in the presence of aluminium and fluoride back to those values obtained with fluoride alone. Of several metal salts tested only beryllium sulfate was able to replace aluminium as an effective inhibitor in the presence of fluoride.Inhibition of the Mg2+-ATPase activity by fluoride and the aluminofluoride complexes correlated with an inhibition of the rate of MgATP-dependent change in red cell ghost shape from echinocytes to smoothed forms. All gross morphological changes of the smoothing process were affected, including the production of discocytes, stomatocytes and endocyctic vesicles.

    Notes: Time-resolved and steady-state electric birefringence imaging with a slow-scan video camera is used to study orientation during DNA agarose gel electrophoresis. The hydrodynamically induced gel distortion is shown to be the major source of birefringence under electrophoresis running conditions and to generate a birefringence image that approximates the image of the DNA concentration gradient in the electric field direction. A fluid kinematic model is presented to describe the spatial distribution of steady-state birefringence and is verified with fluorescence measurements of DNA distribution. The stress-optic coefficient of 1% agarose gel is measured by mechanical compression and used to evaluate the magnitude of the induced strain on the gel during electrophoresis.
