    Notes: Two types of calcareous spicules occur abundantly in Herdmania momus, a solitary pyurid ascidian with a worldwide warm water distribution. The large spindle-shaped body spicules are 1.5-2.5-mm long and are located primarily in the mantle, siphons, and branchial basket. Each body spicule possesses 100 or more rows of overlapping, unidirectional fringing spines. Numerous body spicules occur regularly spaced within a long common sheath of complex structure, and there are many sheaths per animal. Between neighboring body spicules and overlying the fringing spines are the tightly connected pseudopodial sclerocytes. Spine formation is hypothesized to occur within these cells. The body spicules apparently continue to increase in size throughout the animal's life.The tunic spicules are about one tenth the length of the body spicules. They have 20-40 rows of unidirectional nonoverlapping fringing spines and a mace-shaped spiny base that anchors them at the tunic surface. They form quickly in individual spicular envelopes inside the tunic blood vessels over a 4-5-day period. Each tunic spicule then leaves its surrounding envelope and blood vessel, passes into the tunic, and ultimately protrudes through the outer surface of the tunic. An organic covering inside the envelope closely adheres to the tunic spicules and stains with toluidine blue. Dissolution of the CaCO3 mineral phase by EDTA or EDTA-cetylpyridinium chloride-formaldehyde reveals an intricately patterned organic matrix within or upon which the spicules develop.

    Notes: The ascidian sperm reaction, Which involves swelling, migration, and loss of the single large mitochondrion, can be triggered in vitro by raising the seawater pH to 9.3 or lowering Na+ to 20 mM, but only if the sperm are allowed to attach to a suitable Substate. Mitochondrial translocation does not usually occur in the absence of sperm attachment. Extracellular Ca2+ is necessary for triggering the reaction with low Na+ but not high pH; however, the intrecellular Ca2+ blocker, TMB-8, inhibits high pH-induced mitochondrial movement in the absence of extracellular Ca2+. After swelling, the mitochondrion fluoresces in the presence of chlortetracycline, suggesting that Ca2+ becomes membranebound after activation. Elevated cAMP and theophylline both inhibit mitochondrial move ment but not sperm motility. The antiactin drug cytochalasin B(10μM) and the calmodulinblocking drugs TFP (1 μM) and W-13 (10 μM) block mitochondrial movement, suggesting roles for actin and calmodulin in mitochondrial movement. A model is proposed relating intracellular alkalinization, Ca2+ influx, actin, myosin, and calmodulin in mitochondrial translocation.

    Notes: The crystal structure of an infinite inorganic chain consisting of MnII and an N-centered tripodal ligand N,N-(2-pyridylmethyl)[(1-methylimidazol-2-yl)-methyl]glycinate is presented. It exhibits a chiral helical structure with a pitch of two monomeric units (each monomeric unit containing one Mn atom). Each manganese is connected to its neighbor through a carboxylate bridge in a syn-anti geometry. Around each manganese center, two carboxylates bind in a cis geometry. This peculiar bridging geometry (syn-anticis) provides a broken-line chain, running in a zig-zag manner along the b axis of the P21 space group. The magnetic properties have been investigated. They show a pseudo-2D magnetic structure, with one major pathway along the chain and an inter-chain minor one. The intrachain coupling is a weak antiferromagnetic interaction (J/k = -0.25). This low value is entirely consistent with the geometry of the bridge. The interchain coupling is a weaker antiferromagnetic coupling (J′/k = -0.11) and could be mediated through π-π interactions between pyridine and imidazole from two adjacent helixes.

    Notes: An experimental investigation of an impinging water jet freezing on a melting solid surface has been carried out. Attention was focused on the stagnation region of an axisymmetric jet. In the experiment, a water jet was directed upward against the lower end of a meltable rod, having a diameter about twice that of the jet orifice. Solid octane (m.p. - 56.5°C) and solid mercury (m.p. - 38.9°C) served as the meltable materials. A laminar-axisymmetric flow model was developed to describe melting heat transfer in the presence of jet solidification within the impingement region. Measurements of the melting rate and conditions for the onset of jet solidification were found to agree quite well with the values predicted with this model.

    Notes: The incompressible lattice-gas model by ten Brinke and Karasz is adopted to introduce the effect of specific interactions into a recently-presented Monte-Carlobased lattice expression for the Helmholtz energy of nonrandom mixing. While the lattice remains incompressible, intermolecular forces consist of two types: London dispersion forces and specific (chemical) forces. The specific interactions between similar components, as well as those between dissimilar components, are incorporated in a systematic manner. Closed-loop temperature-composition phase diagrams are obtained. The theory is compared with experimental data for several binary systems, including polymer solutions, which exhibit closed-loop coexistence curves. Theoretical and experimental results are in good agreement.

    Notes: The cyanobacterium Anabaena cylindrica B629 was suspended in small glass beds and incubated in a gas-tight glass vessel outdoors under a gas atmosphere comprising carbon monoxide (0.2%), acetylene (5%), oxygen (6.5%), and nitrogen. The solution phase initially contained sodium bicarbonate (10mM) at pH 7. Under these conditions the organism continuously produced hydrogen gas for over three weeks. The temperature of the culture was maintained below 30°C and minimum night temperatures were recorded. The vessel was covered by a shadecloth, which reduced the natural illumination by approximately 70%. The system is an alternative to those requiring the strict absence of oxygen and little nitrogen, and requires virtually no attention during the incubation period.

    Notes: BiP, GRP94 and PDI, three endoplasmic reticulum (ER) based proteins are involved in the maturation of secretory proteins and might represent a bottleneck in the secretory pathway of monoclonal antibodies (MAB). With the three hybridoma cell lines tested, MAB production kinetics were significantly increased for the batch cultures done in serum-free medium (SFM) with respect to those done in serum-containing medium (SCM). It could be established that there was a correlation between the cellular levels of PDI and GRP94 and the specific MAB production rate. With respect to BiP, no correlation with the MAB production rate was observed. The non-producing myeloma cell line X63, used as a reference, showed increased cellular PDI levels when cultivated in SFM. However, in this cell, the cellular GRP94 levels were not significantly influenced by the medium composition.It was concluded that SFM induced an increase of cellular PDI levels and this elevation seemed to be responsible for the increase in the specific MAB production rates. On the other hand, only MAB producing cells showed an increase in the cellular GRP94 levels which might be a result of increased MAB sythesis. Indeed, I.13.17 cultivated in SFM supplemented with serum showed a significantly reduced (about 50%) specific MAB production rate in comparison to I.13.17 cultivated in non-serum supplemented SFM. The cellular PDI and BiP levels did not vary between these conditions of culture, whereas the cellular GRP94 level was about two-fold lower in I.13.17 cultivated in SFM when supplemented with serum than in I.13.17 cultivated in SFM without futher supplementation. These results are discussed with respect to the medium composition as well as in the context of apparent and potential bottlenecks within the secretory pathway of MAB. © 1997 John Wiley & Sons, Inc.     Notes: We examined the effects of proinflammatory cytokines on the expression of two extracellular matrix proteins, e.g., thrombospondin (TSP) and fibronectin (FN) by cultured human umbilical vein endothelial cells (HUVECs). Treatment of HUVECs with human recombinant interleukin- 1β (IL-1β) or human tumor necrosis factor-α (TNF-α) caused a time- and dose-dependent decline in TSP production whereas FN production was not modified. At low concentrations, IL-1β and TNF-α in combination had a greater effect than either agent alone. Interferon-γ (IFN-γ) was without effect. The decline in TSP synthesis resulted in a decreased secretion of this glycoprotein into the extracellular matrix. Endothelial cell monolayers cultured on porous filters were used to study the polarity of TSP secretion. Approximately two thirds of the synthesized protein was secreted to the apical side medium and one third to the basal side medium and both types of secretion were inhibited to a similar extent by cytokine treatment. Immunoprecipitation experiments revealed no apparent degradation of secreted TSP, either in the apical or in the basal compartment. Treatment of HUVECs with IL-1β, either alone or in combination with TNF-α, had no significant effect on the steady-state TSP mRNA levels, suggesting a posttranscriptional regulation. Our results indicate that IL-1β and TNF-α can selectively modulate the composition of the extracellular matrix by decreasing TSP deposition and suggest different regulatory mechanisms for the expression of various secreted proteins by endothelial cells.
