    Notes: Spodoptera frugiperda insect cells were grown in Sf-900 serum-free medium and two kinds of serum-supplemented media (IPL -41 and Grace's). The specific growth rates of uninfected cells were found to be 0.024, 0.35, and 0.034 h-1 respectively, at 33°C. The IPL -41 medium supported to highest maximum cell density (10.6 × 106 cells/mL) compared to 3.5 × 106 and 8.7 × 106 cells/mL with the Grace's and serum-free media, respectively. In temperature shifdown experiments with a temperature-sensitive baculo-virus (acts10YM1CAT), virus titer and chloramphenicol acetyl transferase (CAT) expression were highest in the IPL -41 (5.1 × 107 PFU/mL and 20000 U/mL). Use of Grace's medium gave higher virus titers than the serum-free medium (4.4 × 106 vs 4.1 × 105 PFU/mL) as well as higher CAT titers (7050 vs 1980 U/mL). Interestingly, in the three media used, the highest virus and CAT titers were obtained at MOI (multiplicity of infection) of 0.02 At MOI of 2.0 virtually no increase in virus of CAT titer was observed. This result is contrary to those obtained at constant-temperature (27°C) infection and cell culture, in which higher virus titers and recombinant protein expression and obtained at higher MOI.

    Notes: Isotherms have been obtained for the absorption of water by proteins and polyamides and by simple amides closely related to the repeat chemical unit of the polymers. The differential heat of absorption of water at zero water content has also been determined for these substances, from isosteres and from heats of mixing. In addition the nature of the hydrogen bonding of water to these compounds has been investigated by infrared spectroscopic methods. The large differences found between the heats of absorption by the polymers and the simple model compounds are discussed.

    Notes: In synchronized V79 cells perturbed by serum, heat shock, or ionizing radiation at half-hour intervals through a modal 8.5-hour cell cycle, phase-response curves show a characteristic biphasic pattern of advances and delays in subsequent cell divisions. These observations, together with previous observations of quantizement of generation times in this and other cell lines have led us to consider a model incorporating, in the simplest case, a two-component oscillator with two threshold crossings required per cell cycle. By assuming that oscillator variables respond in a simple way to the experimental perturbations, for example, by first order destruction due to heat shock, a map of the qualitative features of the oscillator can be obtained by matching simulated with experimental phase response curves. Random fluctuations in oscillator variables about a fixed trajectory lead to subthreshold oscillations and result in a distribution of generation times which is roughly a negative exponential, but quantized within this exponential envelope. The extent of the random fluctuations can be determined from comparison with data on desynchronization of a cell population after mitotic selection. The same parameters which correctly simulate phase response and the desynchronization data also give good agreement with generation time distribution data.

    Notes: The synthesis of hexabenz-1,12;2,3;4,5;6,7;8,9;10,11-coronene, using two different reaction schemes is described. The cyclodehydrogenation of hexaphenylbenzene, and the reaction of dibenz-1,9;2,3-anthrone with Zn/ZnCl2 lead to identical products, the chemical and crystallographic properties of which demonstrate the structure of hexabenzcoronene. Furthermore, two other hydrocarbons have been isolated from the reaction of dibenzanthrone with Zn/ZnCl2. The UV. spectrum of the first, and its cyclization to hexabenzcoronene show that it is tetrabenz-4,5;6,7;11,12;13,14-peropyrene. The properties of the second indicate that it is tetrabenz-1,2;3,4,8;9; 10,11-bisanthene.

    Notes: The desire to increase cell density and product concentration has been the primary driving force for the development of better animal cell culture processes. In the technique used in our laboratory - microencapsulation - insect cells (Spodoptera frugiperda), infected with a temperature-sensitive mutant of the Autographa californica nuclear polyhedrosis virus (AcNPV), were cultured in multiple membrane alginate-polylysine (PLL) microcapsules which had a controlled membrane molecular-weight cutoff and an intracapsular alginate concentration which was ca. 16% lower than that obtained in the commercially available single-membrane system. Cell culture experiments indicated that the intracapsular alginate concentration appears to be a key factor in achieving good cell growth. It was possible to obtain intracapsular cell densities of 8 × 107 cells/mL capsules and virus concentrations to 109 IFU/mL capsules. The virus litre in the supernatant was ca. 300 times lower, indicating that virtually all of the virus was retained within the capsules.

    Notes: Numerical predictions of the immediate moment carrying capacity of a short pier foundation in saturated clay are presented. Three-dimensional finite element analyses are carried out using linear and non-linear programs and using a linear axi-symmetric program.Preliminary investigations are made to determine suitable boundary distances for analysis at full-scale and size of loading increment for non-linear analyses.Predictions of the behaviour of prototype pier and of conventional and centrifuge models of this pier are then made. It is shown that the axi-symmetric program yields significantly higher rotations per unit moment than the linear three-dimensional program and that, using both of these programs, elastic analyses of the conventional and centrifuge models and of the prototype yield very similar results. It is also shown that non-linear analyses of the conventional and centrifuge models yield significantly different moment/rotation relationship in accordance with the behaviour actually observed in the model tests. The relationship obtained for the centrifuge model is shown to differ only slightly from that obtained for the prototype, due to the boundary restrictions in the model, and to be of the same order as the centrifuge test result at working condition but not at ultimate capacity.
