Notes: The chromatographic performance of the deuterated solvents, CD3OD and D2O, has been investigated in reversed-phase micro high performance liquid chromatography. The chromatographic performance of CD3OD is only slightly superior to that of CH3OH. However, the performance of D2 is significantly superior to that of H2O, separation of aromatics being improved by about 30%. D2 is a particularly powerful solvent for the separation iof deuterated and non-deuterated compounds.

Notes: The low temperature effect has been investigated in reversed phase micro high performance liquid chromatography with various aqueous mobile phase systems by an approach based on enthalpy-entropy compensation. The compensation temperatures, Tc, were determined for these systems, and the results show that the retention mechanism in a lower temperature column is similar to that in a normal temperature column in the reversed phase mode at mobile phase water contents above 9.8%. At lower water contents, the separation mechanism is different from that in the reversed phase mode, but otherwise very similar to that in the normal phase mode.

Notes: Identification problems often encountered in high performance liquid chromatography (HPLC)/Fourier transform infrared spectrometry (FTIR) can be circumvented through the use of a thin-layer chromatographic (TLC) plate as deposition and infra-red sampling medium. The combination of complementary separation modes is shown to demonstrate increased resolution of the components of complex mixtures. In this particular work, the effluent from a reversed-phase microcolumn is continuously deposited on a TLC plate with alumina stationary phase. The solute remains on the plate as a continuous record of the HPLC separation, which is then analyzed by diffuse reflectance FTIR. When the HPLC separation is inadequate for full separation of the components, the immobilized HPLC chromatogram serves as a starting point for subsequent TLC separation. A number of FTIR reconstructed chromatograms and spectra which are derived from the TLC plate aid in the interpretation.

Notes: The structure-retention relationship in reversed phase micro high performance liquid chromatography has been investigated for substituted alkylbenzene derivatives. The correlation between van der Waals volume and retention data was completely linear and any deviation from this relation might becaused from the overlap volume of substituent alkyl chains. Standard enthalpies of transfer from the mobile to the stationary phase are also proportional to the van der Waals volume of each solute; the results indicate that the mechanism of reversed phase liquid chromatography might involve a solute-induced volume change in the mobile phase.

Notes: An existing commercial refractive index detector was modified for use with slurry-packed capillary column LC systems. The cell gasket of the original detector cell was replaced by a micro cell gasket giving a volume of 0.8 μL. The relationship between detector response and the amount of sample injected was examined. The limit of detection was 2-3 ng for di-n-pentyl phthalate.

Notes: The retention characteristics of alkylbenzenes and polycyclic aromatic hydrocarbons (PAHs) have been examined in liquid chromatography on a microcapillary column packed with cellulose acetate. Particulate and fibrous cellulose acetate were used as the stationary phase. Fibrous cellulose diacetate was found to be of use as an alternative stationary phase for liquid chromatography, although the separation efficiency was low. The retention characteristics for planar PAHs were linearly correlated with the size of the molecules, but the retention behavior for alkylbenzenes and nonplanar PAHs was affected by exclusion. The retention order was strongly influenced by the addition of dimethylformamide or water to methanol in the mobile phase. The results suggested that the retention behavior is influenced by a slight change in the polymer matrix of cellulose acetate.
