    Notes: Hydroxylaminoglutethimide (3-ethyl-3-(4-hydroxylaminophenyl)-2,6-piperidinedione) has been identified as a novel metabolite of aminoglutethimide (3-(4-aminophenyl)-3-ethyl-2,6-piperidinedione) in the urine of patients treated chronically with this drug. The metabolite was isolated by reverse-phase thin-layer chromatography, and characterized by comparison of its mass spectrum and chromatographic properties with those of the synthetic compound. Hydroxylaminoglutethimide is unstable; it is readily oxidized to nitroso-glutethimide and disproportionates in the mass spectrometer into this compound and aminoglutethimide. In none of four patients studied was the metabolite detected in the urine after the first dose of the drug. In one patient it appeared after the second dose and in two more within seven to eight days suggesting that its formation is drug-induced, and that it may be the metabolite responsible for the diminished half-life of aminoglutethimide during chronic therapy. The profile of metabolites from one patient, examined by high-performance liquid chromatography after the first dose and again after six weeks of therapy afforded evidence that the formation of hydroxylaminoglutethimide was at the expense of a major metabolite N-acetylamino-glutethimide. Hydroxylaminoglutethimide was not an induced metabolite in the rat.

    Notes: The fundamental processes of protonation and ethylation, occurring in a methane chemical ionization source, have been investigated for a variety of aromatic amines. The positions of protonation and ethylation on the substrate amines were determined by generating isomeric ions either by protonation of neutral ethyl substituted amines or by ethylation of the amines themselves. The product ions were investigated for structural differences via collision induced dissociation and subsequent analysis via mass analysed ion kinetic energy spectrometry. Similarities and differences between mass analysed ion kinetic energy/collision induced dissociation spectra of these isomeric ions were used to determine protonation and ethylation sites for imidazole, benzimidazole, indazole, pyrrole, pyridine and aniline.

    Notes: The effect of the addition of argon and other gases upon the intensities of negative ion species formed in an electron impact source has been investigated. The negative ion current generated for a series of aromatic compounds has been investigated as a function both of sample and argon pressure in the ion source of a ZAB-2F mass spectrometer. For all compounds studied, a striking enhancement of molecular negative ion current occurred on increasing either the presure of the sample or of argor. The results are consistent with thermalization of the 50 eV electrons by collisions with neutral molecules in the high pressure ion source and collisional stabilization of the negative ions initially formed. Analytical applications of the technique are discussed.

    Notes: The sequence of amino acids in a series of underivatized di- and tripeptides has been determined via collision induced dissociation of [M - H]- ions produced by chemical ionization using [OH]-. The collision induced dissociation spectra of individual [M - H]- ions are determined via mass analysed ion kinetic energy spectrometry. Isomeric peptides can be readily distinguished by this method and characteristic sidechain reactions permit the distinction between leucine and isoleucine. The capability for selection of ions of specific m/z value enables mixtures of non-isomeric peptides to be handled by the above technique. Dipeptides containing certain aromatic amino acids (e. g. tryptophan and histidine) eliminate water thermally but this does not necessarily preclude the identification of their constituent amino acids.
