    Notes: The detection limits of a series of small peptides derivatized with 4-aminonaphthalenesulphonic acid have been determined using a magnetic sector mass spectrometer operated in the negative-ion mode. The instrument (AutoSpec-QFPD) is equipped with an electrospray ion source and both array and point detectors. Detection limits when using the array detector were on the sub-femtomole level at a mass resolution of 2000 (full width half maximum height). Cone-voltage and linked-scan collision-induced dissociation spectra were recorded using both the array and point detectors. When using the array detector, amino acid sequence information could be obtained from as little as 50 fmol of sample consumed.

    Notes: Negative-ion fast-atom bombardment mass spectra of bile acids show prominent [M - H]- pseudomolecular ions, and few fragment ions. Collision-induced dissociation of such pseudomolecular ions gives charge-remote fragmentations which are useful in structural identifications. It has been shown that bile acids conjugated with taurine (2-aminoethanesulphonic acid) give more intense and informative collision-induced dissociation spectra than unconjugated bile acids. In this laboratory, a procedure for the aminosulphation of biological samples, for analysis by tandem mass spectrometry, has been developed. In the present study, a comparison has been made between the collision-induced dissociation spectra of a series of sulphated and glucuronidated bile acids and their 2-aminoethanesulphonic acid derivatives.

    Notes: A series of small peptides has been studied by negative-ion fast-atom bombardment mass spectrometry with collision-induced dissociation. It has been found that by derivatizing peptides with 4-aminonaphthalenesulphonic acid in a peptide linkage at the C-terminus, negative-ion formation can be enhanced and fragmentation in collision-induced dissociation reactions controlled. The peptide-naphthalenesulphonates show charge-remote fragmentations and the resultant spectra give sequence information.

    Notes: Substantial evidence has suggested that a nonsterol product of mevalonic acid (MVA) is essential for the initiation of DNA synthesis in mammalian cells. Several possible isoprenoid candidates have been suggested, but the identity of this compound still remains unknown. In this study we have isolated and purified MVA products from SV40-transformed human fibroblasts and identified fractions with a growth-stimulatory effect. The cells were labelled with [14C]MVA in the presence of inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase. After lipid extraction, the [14C]MVA-labelled lipids were subjected to high performance liquid chromatography and size-exclusion chromatography, and the effect of the fractionated eluate on the DNA synthesis of arrested MVA-depleted target cells was tested. Thereby we found a fraction of [14C]MVA-labelled lipids with a substantial stimulatory effect on DNA synthesis. The chromatographic behavior suggested that the growth-stimulating fractions contained dolichol-20. This was confirmed by mass spectrometric analysis. Similar results were obtained when lipids from hepatocellular carcinoma cells and a sample from breast tumor were isolated and analyzed by the same procedure. The mechanisms by which these compounds induce DNA synthesis are unknown. Recent data obtained in our laboratory have provided evidence that dolichyl groups are covalently linked to tumor cell proteins, which implicates a new biological function for long-chain polyisoprenoid alcohols (Hjertman et al. [1997] FEBS Lett 416:235-238). In this study we demonstrate that tumor cells containing dolichol-like growth-stimulatory lipids also contained dolichylated proteins. This raises the question whether the growth-stimulatory dolichol-like lipids serve as substrates for the dolichylation reaction. J. Cell. Biochem. 71:502-514, 1998. © 1998 Wiley-Liss, Inc.
