    Notes: An automated, 96-well parallel array synthesizer for solid-phase organic synthesis has been designed and constructed. The instrument employs a unique reagent array delivery format, in which each reagent utilized has a dedicated plumbing system. An inert atmosphere is maintained during all phases of a synthesis, and temperature can be controlled via a thermal transfer plate which holds the injection molded reaction block. The reaction plate assembly slides in the X-axis direction, while eight nozzle blocks holding the reagent lines slide in the Y-axis direction, allowing for the extremely rapid delivery of any of 64 reagents to 96 wells. In addition, there are six banks of fixed nozzle blocks, which deliver the same reagent or solvent to eight wells at once, for a total of 72 possible reagents. The instrument is controlled by software which allows the straightforward programming of the synthesis of a larger number of compounds. This is accomplished by supplying a general synthetic procedure in the form of a command file, which calls upon certain reagents to be added to specific wells via lookup in a sequence file. The bottle position, flow rate, and concentration of each reagent is stored in a separate reagent table file. To demonstrate the utility of the parallel array synthesizer, a small combinatorial library of hydroxamic acids was prepared in high throughput mode for biological screening. Approximately 1300 compounds were prepared on a 10 μmole scale (3-5 mg) in a few weeks. The resulting crude compounds were generally 〈80% pure, and were utilized directly for high throughput screening in antibacterial assays. Several active wells were found, and the activity was verified by solution-phase synthesis of analytically pure material, indicating that the system described herein is an efficient means for the parallel synthesis of compounds for lead discovery. © 1998 John Wiley & Sons, Inc.     Notes: MgBr2·OEt2 (1), which is commercially available and has been examined by different methods in the past, was obtained in crystalline form as a by-product of the reaction of amine-stabilised A1C1 solution with mesitylene magnesium bromide. The following unexpected structure was obtained by X-ray techniques: five-coordinate magnesium centres form trigonal bipyramids, which are connected by two common bromine atoms into chains. Structural details and especially the relationship with solid MgBr2, MgBr2·(OEt2)2 and MgI2·(OEt2)2 (2) are discussed in order to illuminate the unusual bonding in 1.

    Notes: An improved synthesis of the enantiomers of the new benzofurane-type antiarrhythmic compound 1 is described, which makes use of the enantiomerically pure mbe-lactol. Thus, acylation of the benzofurane 4 with acetic anhydride and subsequent bromination gave the bromoacetyl-derivative 6, which, after reduction with LiAlH4, was protected with mbe-lactol to give a mixture of the diastereomers 8A and 8b. After separation via column chromatography assignment of absolute configuration was carried out using a well-established NMR- method. Reaction with propylamine and cleavage of the protective group gave (R)-1 and (S)-1, respectively. Enantiomeric purity was confirmed using a direct HPLC method with rsp-cyclodextrine as stationary phase. Pharmacological investigations on isolated guinea pig heart muscle preparations showed that GE 68 and its two enantiomers did not significantly differ from each other with regard to their negative inotropic, negative chronotropic, and lack of β-adrenoceptor blocking action. In contrast, the reference drug propafenone was equally potent in its negative inotropic and chronotropic activity as GE 68, but additionally showed a weak β-adrenoceptor blocking activity. © 1994 Wiley-Liss, Inc.

Addition and Insertion Reactions of Thermically Generated Fragments from PentaphenylcyclopentaphosphineAddition and insertion reactions of phenylphosphinidene (PhPl), generated by heating pentaphenylcyclopentaphosphine (1), are investigated. At 160°C 1 reacts with tolane to form a complex mixture, from which tetraphenyl-Δ3-1,2-diphosphetene (4) has been isolated. After reaction of the mixture with sulfur or oxygen pentaphenyl-Δ4-1,2,3-triphospholene 1,3-disulfide (9) and 1,2,3,4,5-pentaphenyl-3-phospholene 1-oxide (10) have been found.  -  With allyl ethyl sulfide 1 forms the product of direct insertion of phenylphosphinidene into the C—S bound. After reaction with sulfur the insertion product has been determined as ethyl allylphenyldithiophosphinate (13).
    Notes: Additions- und Einlagerungs-Reaktionen des beim Erhitzen von Pentaphenylcyclopentaphosphin (1) gebildeten Phenylphosphinidens (PhPl) werden untersucht: Bei 160°C reagiert 1 mit Tolan zu einer komplexen Mischung, aus der Tetraphenyl-ΔO3-1,2-diphospheten (4) isoliert wurde. Nach Umsetzung der Reaktionsmischung mit Schwefel oder Sauerstoff wurden das 1,3-Disulfid von Pentaphenyl-Δ4-1,2,3-triphospholen (9) und das 1-Oxid des 1,2,3,4,5-Pentaphenyl-3-phospholens (10) isoliert.  -  Bei der Reaktion von 1 mit Äthylallyl-sulfid wurde in großer Konzentration das Produkt der Einschiebung von Phenylphosphiniden in die C—S-Bindung nachgewiesen und nach Reaktion mit Schwefel als Allylphenyldithio-phosphinsäure-äthylester (13) bestimmt.

    Notes: In this paper we present an extension of Reusken's Lemma about the smoothing property of a multigrid method for solving non-symmetric linear systems of equations. One of the consequences of this extended lemma is the verification of the smoothing property for all damping factors οε(0, 1). Additionally, a semi-iterative smoother is constructed which gives, in some sense, optimal smoothing rate estimates.
