    Notes: The effects of the uncouplers CCCP and DNP on photokinesis, phototaxis, and the photophobic response in the desmid Cosmarium have been studied both in population systems and by videomicrographic, single-cell analysis. Light-dependent motility is specifically inhibited by both uncouplers, indicating that photokinesis is driven by photophosphorylation. In population experiments, phototaxis and accumulations in light traps due to photophobic responses are inhibited by drug concentrations comparable to those that inhibit photokinesis. Analysis of single-cell behavior demonstrated, however, that neither photophobic responses elicited by an increase in light intensity (step-up response) nor by a decrease (stepdown response) are inhibited, as long as the reduced motility allows the organisms to cross a light--dark border. Phototactic orientation is not impaired by DNP in the single cell analysis, but CCCP significantly reduced the degree of orientation. The results indicate that, although chlorophyll is the photoreceptor for all three photoresponses, at least the photophobic response is independent of both the photosynthetic electron transport chain and photophosphorylation.

    Notes: Photoreceptor proteins for photoorientation in microorganisms are usually membrane bound and can be isolated by standard biochemical methods. Three examples are shown: the flagellates Euglena gracilis, Peridinium gatunense and the slime mold Dictyostelium discoideum. The photoreceptor of Euglena is attached to the basis of the flagellum and is composed of at least four chromoproteins which can be separated by gradient sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) fast protein liquid chromatography (FPLC) and isoelectric focusing (IEF); it contains pterins and a flavin as chromophoric groups. The photoreceptor of Peridinium absorbs in the red wavelength band. Though not yet identified in detail, multiple receptors are probably involved, as indicated by fluorescence spectroscopy. Dictyostelium shows positive and negative phototaxis in its amoebal form and exclusively positive phototaxis in its pseudoplasmodial form. It is still open to discussion whether the two stages use separate photoreceptors. From amoebae two photoreceptor pigments have been isolated, showing an absorption which resembles the action spectrum, one membrane bound with a molecular mass of 45 kDa and one cytoplasmic fraction with a molecular mass of 27 kDa.

    Notes: Techniques for automatic analysis of two-dimensional electrophoresis gels by computer-aided image analysis are described. Original gels or photographic films are scanned using a laser scanner and the files are transferred to a microcomputer. The program package first performs a compression and preevaluation of the files. Spot identification and quantification is performed by the chain code algorithm after appropriate zooming and cutting. Labeling facilitates spot identification and quantification in numerical and graphical (pseudocolor) representation on peripheral devices for camera ready output. Interpolation between measured basepoints is performed by cubic spline algorithms which are automatically switched on and off, depending on the need by the program. High speed analysis and graphic representation is achieved using fast Assembler language routines rather than high level languages. One-dimensional gels can be analyzed using the same software. Spot matching between parallel two-dimensional gels has not yet been implemented.

    Notes: Equipment made of special materials. Errors in the planning phase of capital expenditure and major repairs often lead to particularly high costs in remedying them. The investigations that were necessary to ensure reliable manufacture are described with reference to a heat exchanger made of zirconium. In addition to design details, the testing of the explosiveclad tube sheets and the welding of the heat exchanger tubes are explained at length. In the repair sector a report is given on damage to an evaporating vessel made of nickel-clad sheet. Particular attention is paid to the build-up of tension in the cladding in relation to the support material. A second example of damage is used to show the influence of design details on the damage mechanism. In this case Monel 400 piping is involved.

Abschließend wird der Einsatz von Berstsicherungen zum Zwecke der Explosionsdruckentlastung behandelt und weiterhin die Konzeption eines „druckstoßfesten Behälters“ erläutert.

    Notes: Preventive and constructional measures against gas and dust explosions. Even during the planning of plant for processing potentially explosive products, process and constructional aspects should be accompanied by investigations on the products conducted with a view to obtaining as complete a picture as possible of the danger of explosion of the materials. These studies provide the basis for estimates of explosive risks in the plant and for decisions concerning the extent to which safety measures against the occurrence of explosions or the effects of explosions can be implemented. An explosion-proof plant can be accomplished by preventive measures hindering an explosion, or by constructional measures suppressing the effects of an explosion. This paper is intended to be of assistance in the choice of explosion-proofing equipment.

    Notes: Die aquatischen Ökosysteme produzieren etwa genauso viel Biomasse wie alle terrestrischen Ökosysteme zusammengenommen. Neben den Makroalgen, welche die Küsten der Weltmeere besiedeln, sind vor allem die Phytoplanktonorganismen (meist einzellige Mikroalgen) die wichtigsten Primärproduzenten. Obwohl die Einzelorganismen mikroskopisch klein sind (Abbildung 1), stellen sie die entscheidende erste Stufe des weitverzweigten Nahrungsnetzes dar. Eine meßbare Reduktion der Produktivität des Phytoplanktons durch erhöhte solare UV-Strahlung hat sicherlich drastische Folgen für die nachgeschalteten Nahrungsketten sowie das gesamte Ökosystem. Außerdem muß mit dramatischen Folgen für unser Klima gerechnet werden [9].
