    Notes: Conventional sources of protein cannot meet the present or projected needs for human consumption. Single cell proteins from fermentation of petroleum and cellulosic wastes are likely sources of additional protein. The volume of cellulosic wastes is sufficient to supply all additional protein needs on a continuing basis for cellulose is a renewable resource. Both mesophilic and thermophilic microorganisms utilize cellulose at reasonable rates. Biodegradation of lignin and lignin-cellulose complexes constitutes a major obstacle to commercial utilization of cellulosic wastes. Thermophilic actinomyces appear to be the most effective organisms for single cell protein production from cellulosic wastes.

    Notes: Lipid synthesis and secretion was measured in primary rat mammary epithelial cells cultured on basement matrix in medium supplemented with lactogenic hormones. The cells grew and differentiated to form alveolar-like structures reminiscent of lactating mammary gland. They synthesized abundant triacylglycerol, containing fatty acids characteristic of rat milk (C10:O-C14:0), using 14C-glucose, 14C-oleic acid or 14C-glycerol as precursors. Basal levels of triacylglycerol secretion were measured using 14C-oleic acid labeling; 1.3±0.3% of the labeled cellular triacylglycerol was secreted into the medium in 24 hours. Secreted lipid droplets were surrounded by a bilayer membrane with an electron-dense inner coat characteristic of fat globules secreted by the mammary gland. The rate of triglycerol secretion was increased to 998±98% of control (P〈0.01) by the addition of phorbol 12-myristate 13-acetate (PMA) in combination with staurosporine, a protein kinase inhibitcn. Several other protein kinase inhibitors, when combined with PMA, also markedly stimulated secretion. Effective protein kinase inhibitors included sphingosine (has diverse cellular effects including the inhibition of protein kinase C; 13-fold increase in secretion), and KT5823 (a cGMP dependent protein kinase inhibitor; 5-fold increase). KT5720 (a cAMP-dependent protein kinase inhibitor) did not alter secretion. Kinase inhibitors were effective only in the presence of a phorbol ester. 4α-phorbol-12,13-didecanoate, a phorbol ester which does not activate protein kinase C (PKC), could substitute for PMA. Lipid release was not mediated by disruption of cell-cell tight junctions, as EGTA did not release lipid. Based on these observations we suggest that two signals are needed to enable or stimulate lipid secretion in cultured rat mammary epithelial cells: (1) inhibition of a protein kinase and (2) a PKC-independent effect of phorbol ester. We have, for the first time, characterized a cell culture model suitable for studying lipid synthesis and secretion by mammary epithelial cells. © 1993 Wiley-Liss, Inc.

    Notes: The authors have developed a statistical model for a branched polymer built from a monomeric unit (such as glucose) which has two types of reactive groups (aldehydic and alcoholic). The treatment differs from that of Flory's in that the different B groups in the A-R-Bf-1 monomer have differing reactivities instead of equal reactivities. From the above, x̄n, the number-average DP, x̄w, the weight-average DP, x̄z, the Z-average DP, and wx, the weight fraction of an x-mer, have been obtained. It is shown that the limit of x̄z/x̄w is 3 and the limit of x̄w/x̄n is infinity as the degree of polymerization increases for all cases where “f” is greater than two. These results were used for the randomly branched polymers, amylopectin and glycogen, where the functionality of the monomer in both cases is three. p4 and p6 were used to represent the probabilities that any glucose unit is linked at the 4 or 6 position. Using values of p6 = 0.05 and p4 = 0.94933, x̄n = 1500, M̄n = 243,000, M̄w=35,100,000, and M̄z = 104,500,000, giving M̄w/M̄n = 140 and M̄z/M̄w = 2.99. Literature values for molecular weights of amylopectin and glycogen are compared with the above statistical model and with less random statistical models.

Methods are also now available for obtaining pure populations of haemopoietic cells. The use of such cells, in combination with pure growth factors, has provided intriguing information about the biological activities and mode of action of the factors in faciliating survival, proliferation and differentiation of the haemopoietic cells.

    Notes: An intelligent closed-loop expert control system has been developed for automated control of the resin transfer molding process of a graphite fiber preform using an epoxy resin, E905L. The sensor model system has been developed to make intelligent decisions based on the achievement of landmarks in the cure process, such as full preform impregnation, the viscosity, and the degree of cure of the resin rather than time or temperature. In-situ frequency dependent electromagnetic sensor (FDEMS) and the Loos resin transfer model are used to monitor and control the processing properties of the epoxy resin during RTM impregnation and cure of an advanced fiber architecture stitched preform. Once correlated with viscosity (η) and degree of cure (α), the FDEMS sensor monitors and the RTM processing model predicts the reaction advancement of the resin, viscosity and the impregnation of the fabric. This provides a direct means for monitoring, evaluating, and controlling intelligently the progress of the RTM process in situ in the mold throughout the fabrication process and for verification of the quality of the composites.

    Notes: New adenine (A) and uracil (U) π → π* transition-moment parameters have been derived from a recently developed semiempirical procedure. Using conformational energy probabilities based on the Boltzmann equation, the new parameters were assigned by optimizing the calculated CD of cyclic nucleotides against measured CD. The derived A-and U-parameters (along with guanine and cytosine parameters derived previously by the same procedure) have been assessed in CD spectral calculations of some polyribonucleic acid sequences, in assumed A-class geometries. Comparisons have been made between CD spectra calculated from the newly derived parameters and those calculated from parameters obtained from a combination of crystal optical measurements and quantum-mechanical calculations. Although some spectral differences do occur, for the RNA sequences considered, no major disagreements were found in CD spectral signs and shapes, between measurements and calculations. Overall, the results indicate that the newly derived A-, U-, G-, and C-parameters show better agreement between theory and experiment than those used in previous nucleic acid CD calculations.

    Notes: In long-term marrow cultures haemopoiesis can be maintained in vitro for up to 6 months. Critical analysis of the cell populations produced has shown that the stem cells and their committed progeny have characteristics in common with the corresponding cell types in vivo. The maintenance of haemopoiesis in vitro is associated with the development of an appropriate inductive environment provided by bone marrow derived adherent cells. Analysis of the interactions between environmental and haemopoietic cells has been facilitated by the development of in vitro systems reproducing the naturally occurring genetic environmental defects and other systems where the development of a competent inductive environment shows a dependency upon corticosteroid hormones. Investigations have shown that stem cell proliferation may be controlled by production of opposing activities, one stimulatory for DNA synthesis, the other inhibitory. A model is proposed whereby modulation in the production of these factors is determined by the physical presence of stem cells in a proposed cellular milieu, within the adherent layer. The adherent layer, apart from acting at the level of stem cell proliferation, can also modify the response of differentiating cells (eg, GM-CFC) to exogenous stimulatory activities. Addition of GM-CSF or of CSF-antiserum has no effect on haemopoiesis in long-term cultures.
