    Notes: Immobilization of amyloglucosidase (EC 3.2.1.3) from Endomycopsis bispora and Aspergillus niger was followed with carriers containing epoxide, aldehyde, and primary amino groups. Determinations of stability of bound enzyme showed that the most active and stable preparations were obtained by application of the carrier with amino groups activated with glutaraldehyde (activity half-life 96.6 days). Optimum pH and and temperature were found for cleavage of starch solution and KM(app) and Vmax(app) values were determined for all prepared samples.

    Notes: Two types of bead-form macroporous carriers based on glycidyl methacrylate with ethylene dimethacrylate copolymers were used for the immobilization of penicillin amidase either directly or after chemical modificaton. Direct binding through oxirane groups, which is equally efficient at pH 4.2 and 7, is relatively slow and brings about an activity loss at low enzyme concentrations. The most efficient immobilization was achieved on glutaraldehyde-activated amino carier, irrespective of whether the amino groups were formed by ammonia or 1,6-diaminohexane treatment of the original oxirane carrier. Hydrazine treatment gave lower immobilization yields. The same is true of the azide method independent of the length of the spacer. Most enzyme activity was preserved by coupling the carbodiimide-activated enzyme to the carrier with alkyl or arylamino groups at the end of a longer substituent. Immobilization on diazo-modified carrier gave average results. Rapid immobilization by a lysine-modified phosgene-treated carrier resulted in an activity loss. It is suggested that multipoint and very tight attachment of the enzyme molecule to the matrix decreased the activity. The immobilized activity is quite stable in solution and very stable upon lyophilization with sucrose.

    Notes: Glucose oxidase was immobilized by covalent bond to two basic types of sorbents - glycidylmethacrylate copolymers and bead cellulose. These two types of carries were chemically modified, if needed, by the employing various procedures and subsequently used in the immobilization of native and oxidized glucose oxidase. The samples thus obtained were compared with those of immobilized glucose oxidase bound onto some common carriers. Samples which possessed not only a high absolute activity but also adequate mechanical and flow properties were characterized in greater detail with respect to the immobilization efficiency and kinetic properties of bound glucose oxidase.

    Notes: Alcohol dehydrogenase has been immobilized to the basic copolymer and its several derivatives using various techniques. Enzyme coupling to the supports with amino groups by means of glutaraldehyde was found the most suitable. Activity of alcohol dehydrogenase coupled to these amino supports was comparable to that of the enzyme bound to Sepharose. Thermal and pH stability of alcohol dehydrogenase increased essentially upon immobilization. Kinetic properties of the immobilized enzyme differed from those of free alcohol dehydrogenase, pH optimum shifted to alkaline range, and apparent Michaelis constants for substrates and coenzymes increased. Curvatures observed in Lineweaver-Burk plots for coenzymes suggest an involvement of diffusion effects in the reaction catalyzed by alcohol dehydrogenase linked to these polymers.

    Description / Table of Contents: Makroporöse reaktive Copolymere auf der Basis Glycidylmethacrylat und Ethylendi-methacrylat wurden so modifiziert, daß sie schließlich über ein mmol g-1 an Sulfonyl-gruppen enthielten. Zunächst wurden dazu die Epoxygruppen zu Hydroxylgruppen hydrolysiert, dann wurden diese durch Behandlung mit Propansulton in Gegenwart von Natronlauge verethert. Der Einfluß unterschiedlicher Reaktionsbedingungen auf das Ausmaß der chemischen Modifikation und die Porenstruktur wurden genauer untersucht. Die Ausbeute der Sulfopropylierung hängt hauptsächlich von der Konzentration an NaOH und von der spezifischen Oberfläche der makropörosen polymeren Matrix ab.
    Notes: Macroporus reactive polymers based on glycidylmethacrylate and ethylenedimethacrylate were modified so as to contain more than one mmol g-1 of sulfonic acid groups. This modification consists in the introduction of hydroxyl groups, by hydrolysis of the epoxy groups of the reactive polymers, and in its subsequent etherification by treating with propane sultone in aqueous sodium hydroxide medium. The effect of changing reaction conditions on the extent of chemical modification and on pore structure was studied in detail. The yield of sulfopropylation depends mainly on the sodium hydroxide concentration and on the surface area of the macroporous polymeric matrix.

    Description / Table of Contents: Durch Reaktion des Copolymeren aus Glycidylmethacrylat und Ethylendimethacrylat mit 8-Aminochinolin wird ein Harz hergestellt und damit die Sorption der Ionen Fe(III), Al(III), Mn(II), Co(II), Ni(II), Cu(II), Zn(II), Cd(II), Au(III), Pd(II), Pt(IV) und Hg(II) untersucht. Aus Salzsäure werden Gold und Palladium stark sorbiert. Aus den Ergebnissen von statischen und dynamischen Versuchen folgt die Möglichkeit, Au(III) und Pd(II) von Pt(IV) und anderen Metallionen zu trennen. In salpetersauren Lösungen sorbiert das Harz stark Hg(II)-Ionen.
    Notes: The sorption of Fe(III), Al(III), Mn(II), Co(II), Ni(II), Cu(II), Zn(II), Au(III), Pd(II), Pt(IV), and Hg(II) ions on a sorbent prepared by reacting the copolymer of glycidyl methacrylate and ethylene dimethacrylate with 8-aminoquinoline was investigated. Gold and palladium are strongly absorbed in hydrochloric acid. Results of static and dynamic tests indicate the possibility of separation of Au(III) and Pd(II) from Pt(IV) and from the other metal ions investigated in the study. The sorption of Hg(II) by the polymer increases in nitric acid.

    Description / Table of Contents: Das Verhalten von makroporösen Methacrylat-Copolymeren wurde in einem Temperaturbereich von 20-270°C untersucht. Dabei wurde in der Retension von Methanol und n-Dekan eine Diskontinuität bei 40°C beobachtet, die durch eine steigende Kettenbeweglichkeit der die innere Oberfläche der Polymerteilchen bildenden Makromoleküle interpretiert werden kann. Die Temperatur stimmt gut mit dem veröffentlichten Wert für Tg des Glycidylmethacrylats überein und ist unabhängig von dem Grad der Substituierung der Ketten mit ionogenen Gruppen. Bei einer Temperatur von 210-250°C läuft eine Depolymerisation ab; im Fall von Methacrylsäure-Polymeren entstehen zyklische Anhydride, und die Ester spalten Alkyl-Gruppen ab. Durch ionogene Gruppen modifizierte 2,3-Dihydroxypropylmethacrylat-Copolymeresind stabiler, die Depolymerisation ist bei gleichen Bedingungen schwächer. Die Einwirkung der Temperatur verursacht auch Änderungen sowohl in der Morphologie der Partikel und der Poren, als auch in der Polarität der Polymeren, die sich im Verhaltnis der Retensionszeiten für Alkohole und Alkane und in der Lösungswärme äußiert.
    Notes: The behaviour of macroporous methacrylate copolymers was investigated in the temperature range between 20 and 270°C. On the basis of retention of methyl alcohol and n-decane a discontinuity at 40°C was recorded in all cases, interpreted by the increasing mobility of chains of surface layers of the polymer globules. This temperature agrees with the reported Tg value of poly(glycidyl methacrylate) and is independent of further substitution of the polymer chain with ionogenic groups. At 210-250°C depolymerization takes place; in the case of methacrylic acid copolymers cyclic anhydrides are formed, while in the case of esters alkyls are eliminated. Modified copolymers of 2,3-dihydroxypropyl methacrylate with ionogenic groups are more stable thermally  -  the depolymerization proceeds under the same conditions to a smaller degree. Heating is connected with changes in the morphology of particles and pores, and also in the polarity of the polymers noticeable by the ratio of retentions of alcohols and paraffins and by the heat of dissolution.
