    Notizen: The in vivo pathway of folding and subunit assembly of a trimeric bacteriophage protein has been studied by characterizing precursors to the native protein and by analyzing temperature-sensitive mutations that kinetically block the pathway. The native trimer is formed via an intermediate composed of three partially folded chains, the protrimer. At 39°C, temperature-sensitive mutations prevent the formation of both the native trimer and the protrimer, possibly by destabilizing earlier intermediates. However, the mutations do not affect the stability of the native protein, formed at 30°C. Thus, these mutations identify amino acid residues involved in interactions that determine the folding pathway.

    Notizen: The elastic modulus (G′) of factor XIIIa induced fibrinogen gels was found to be substantially lower than the G′ of fibrin gels that were formed by clotting fibrinogen with thrombin. The addition of fibronectin and/or the reducing reagent dithiothreitol (DTT) to the factor XIIIa coagulation mixture led to the formation of a weaker gel structure, while the rigidity of thrombin induced clots was not appreciably affected by the inclusion of the DTT but increased somewhat in the presence of fibronectin. The reasons for the differing clot rigidities are discussed in terms of biochemical mechanisms.

    Notizen: We report the temperature dependence of the H2 and H8 purine ring proton resonances of oligoriboadenylates up to chain length 11, with or without a single guanosine residue at the 5′-end, second position, or 3′-end. The results suggest the following generalizations: (1) Stacking of the bases in a right-handed single-stranded helix is more extensive in the interior of the chain than at the chain ends. (2) The tendency of the terminal base to unstack is greater at the 5′-end than at the 3′-terminus. (3) G stacks more weakly than A, as evidenced by weak stacking of 3′-terminal G. Anomalies were also observed in the unstacking profile of G at the second position in the chain, indicating a conformational anomaly such as looping out of G, thereby allowing adjacent A's to stack together, or adoption by G of some other alternative structure. (4) The results imply that the environment at a given base is influenced by effects of longer range than nearest- or next-nearest-neighbor. Increasing ion condensation as chain length increases may be responsible for the slow approach of oligomer behavior to the properties of the high polymer.

    Notizen: Dynamic light scattering has been used to characterize a variety of lathyritic rat skin collagen solutions. The technique was used to monitor the onset of fibril assembly in vitro and to investigate the thermal memory effect. Although the incorporation of thermal memory was demonstrated by reheating the sample and subsequently observing a shortened turbidimetric lag phase, no significant differences between naive solutions and ones exhibiting thermal memory could be detected using photon correlation spectroscopy. This suggests that subtle changes in the state of the collagen molecules rather than extensive changes in the degree of aggregation are responsible for the thermal memory effect. During fibrillogenesis, no large-scale changes in the distribution of monomers or aggregates occur until near the end of the lag phase.

    Notizen: Fourier transform 1H-decoupled 13C NMR spectra were obtained on seven poly(vinyl chloride), (PVC), samples in solution in o-dichlorobenzene. Four of the polymers were of the commercial type and the others were prepared by suspension polymerisation at +90°C, +5°C, and -30°C. Measurements of the spectra were used to assess the precision and accuracy with which information about the tacticity of PVC can be obtained from such spectra. The results show that the well-resolved α-carbon triad and β-carbon tetrad spectra obtained may be reliably analysed to give values of the probability of syndiotactic placement with a precision of about 0,005 for measurements on a single run on a single sample. The simplest and probably the most reliable method of measuring the areas under the peaks which correspond to specific triads ot tetrads is to use the spectrometer integrator, although results of slightly lower precision may be obtained by the use of a curve resolver. Consideration of possible systematic errors due to different T1 relaxation times, unequal nuclear Overhauser enhancement (NOE) factors, and incomplete dissolution of the polymer has shown that the only one of these which is possibly significant is that due to NOE, which is almost certainly less than 0,008, so that the absolute accuracies of the Pr values should be better than that. A more detailed study of the data shows that the polymers made at 5°C and -30°C are almost certainly non-Bernoullian and that firstoder Markov statistics can adequately describe the observed departures from Bernoullian statistics. These departures may have significant consequences for the physical properties of the polymers.

    Notizen: Various poly(alkyl 2-cyanoacrylates) and poly(allyl 2-cyanoacrylates) having different molecular weights have been prepared. Dynamic mechanical analysis was carried out over the temperature range 23°Cto 230°Cat a fixed frequency of 110 Hz, in order to study the effect of temperature on the storage modules, loss modulus and loss tangent of the films. From these data, the glass transition temperature of the polymers could be determined and the effect of the alkyl group on the stiffness of the polymers could be evaluated. It is shown that crosslinking occurs in the case of poly(allyl 2-cyanoacrylates) when it is subjected to temperatures above its glass transition temperature. This crosslinking is seen as a rapid increase in the storage modulus. This increase is eliminated if the polymer samples are thermally aged prior to dynamic mechanical analysis.

    Notizen: Block-copoly(oxypropylene/oxyethylene/oxypropylene)s, with central-block lengths of 39 and 75 oxyethylene units and end-block lengths in the range 1 to 13 oxypropylene units, were investigated in their solid states by small-angle X-ray diffraction and low-frequency Raman spectroscopy, supplemented by differential scanning calorimetry. Low-frequency Raman transitions were assigned to longitudinal modes (LAM) of either unfolded or once-folded chains. The solid state comprised stacked monolayer lamellae in which the chains were usually tilted with respect to the lamellar end plane.

    Notizen: The flow behavior of α,ω-dihydroxypolydimethylsiloxanes, having a weight-average number-average molecular weight ratio of 1.1-1.2, was studied with a Cannon-Manning viscometer and an Instron rheometer. Comparison of the flow behavior of samples with narrow and broad molecular weight distributions indicated that the onset of non-Newtonian behavior occurred at a much higher shear rate for narrow-distribution polydimethylsiloxanes than for polydisperse polydimethylsiloxanes. A plot of reduced viscosity versus \documentclass{article}\pagestyle{empty}\begin{document}$ \dot \gamma \eta _0 {M \mathord{\left/ {\vphantom {M T}} \right. \kern-\nulldelimiterspace} T} $\end{document} gave two experimental master curves, one for polymer of narrow distribution and the other for polydisperse polymer. The experimental master curve obtained from the narrow-distribution polymer was found to fit the theoretical master curve derived from Graessley's entanglement theory. The viscosity-molecular weight relationship for the higher molecular weight polydimethylsiloxanes was found to be the same for both hydroxydimethylsilyl- and trimethylsilyl-endblocked polymers. However, at low molecular weight, the viscosity-molecular weight curve deviated from linearity because of the association of polydimethylsiloxanols, which apparently is not significant at higher molecular weights. The critical molecular weight of entanglement, Mc, was found to be about 30,000.

    Notizen: Heterogeneous propionylation of cotton cellulose, in the form of yarn, was carried out by reaction with propionyl chloride, in a medium of pyridine and dimethylformamide (DMF). The product was a mixed propionate-α-propionylpropionate ester of DS varying from 0.24 to 2.94. The supramolecular structure of these esters was studied by kinetic analysis as well as by the measurements of density, refractive index, and x-ray diffraction. The Sakurada plot of DS against time of reaction showed a discontinuity at a DS of about 2.0, where the x-ray diffraction pattern shows almost complete loss of crystalline structure. The interpretation, based on the assumption that the Sakurada curve actually represents the sum of two simultaneous rate processes acting in the amorphous phase and at the crystalline surfaces, seems to explain the data adequately. The density showed a continuous decrease, consistent with the idea of continuous destruction of crystalline structure with progressive substitution. Refractive indices showed a continuous decrease with substitution and birefringence was almost absent at complete substitution. The calculated value of molar refraction for the sample of DS 2.94 agreed closely with the experimentally observed value. The x-ray diffraction intensity traces gave convincing evidence of the progressive decrystallization of cellulose with the degree of substitution. Decrystallization seems to be more or less complete at about a DS of 2.0.
