Notizen: Two series of 5-furylmethylenehydantoins and 5-thienylmethylenehydantoins were prepared and their 1H and 13C NMR spectra studied in comparison with those of compounds in the analogous 5-aryl- and 5-pyridyl- series. Differences in the effects of the aromatic, six- or five-membered heteroaromatic rings are discussed. Spectral analysis enables Z/E configurations to be assigned with confidence and conformational preferences to be qualitatively deduced. Some interesting solvent effects were also observed.

Notizen: The Z/E photoisomerization of a series of 5-arylmethylenehydantoins and a series of 5-pyridylmethylenehydantoins by direct irradiation in methanol or ethanol solvent was investigated. The isomeric ratios at the photostationary state are dependent on the excitation wavelength and are related to the relative absorption coefficients of the two isomers. Measurements of quantum yields show the absence of quenching effects by oxygen or azulene, suggesting that triplet states either are not involved or are too short-lived for quenching to be observed. Preparative photoisomerization provides a route to the isomer which is not isolated or obtained only as a minor product by the usual method of synthesis.

Notizen: The ionization constants of nine amphoteric 5-pyridylmethylenehydantoins were measured in 80% (w/w) dimethyl sulphoxide-water at 25 °C. The effects of structure and Z/E configuration on both pKaI and pKaII values are discussed. The especially low basicity and acidity of the Z-isomers of the N-unsubstituted and 3-methyl-substituted 5-(2-pyridylmethylene)hydantoin are attributed to the formation of intramolecular N(1)—H…N hydrogen bond in the s-cis conformation of these compounds. The existence of N(1)—CH3…N attraction is also postulated for the Z-isomer of 1-methyl-5-(2-pyridylmethylene)hydantoin.

Notizen: Nine 5-pyridylmethylenehydantoins were prepared. Each of the 1-methyl-substitute compounds was obtained in two stereoisomeric forms. Only one form of each of the 3-methyl-substituted and N-unsubstituted compounds was obtained directly from synthesis but could be partially converted into the other stereoisomer photochemically. The Z/E configurations and the conformational relationship between the pyridine and hydantoin rings were studied by 1H and 13C NMR spectroscopy, including variable-temperature 1H NMR. The existence of N—H…N or C—H…N interactions and the possibility of tautomerism are suggested for some of the compounds. The Z-isomers of compounds with 2- or 3-pyridyl rings prefer an s-cis conformation whereas the E-isomers prefer an s-trans conformation.

Notizen: The patterns of the NH stretching vibrations in the solid-state IR spectra of a series of 5-pyridylmethylenehydantoins revealed the presence of various modes of hydrogen bonding: intermolecular NH&4nldr; &4nldr;OC and intermolecular or intramolecular NH&4nldr; ‥‥N(py). These variations are related to the orientation of the pyridyl nitrogen and to stereochemistry. A distinction between intramolecularly and intermolecularly hydrogen-bonded compounds was also provided by comparison of the 1H NMR spectra in (CD3)2SO and in CDCl3.

Notizen: A method of evaluating the in vitro viscoelastic properties of microfibrous Biomer poly(ether-urethane-urea) vascular prostheses is outlined. Quasi-static and dynamic tests were carried out on Biomer grafts of diameter between 3.4 mm and 3.8 mm and wall thickness between 0.25 mm and 0.55 mm. It is shown that the quasi-static compliance of a Biomer graft may be determined from an equation relating transmural pressure, radius, and longitudinal strains in terms of the graft dimensions and material constants. The dynamic compliance spectra were evaluated as a function of the longitudinal and circumferential strains and temperature. Although the ratio of dynamic compliance to quasi-static compliance was linearly related to the logarithm of frequency it was not significantly affected by strains or temperature over the relevant ranges studied. Employing the usual assumptions of linear isotropic incremental elastic theory the dynamic elastic and viscous moduli were calculated as a function of frequency. Biomer grafts were more viscous than canine carotid and femoral arteries, especially at the higher frequencies. The variation in the ratio of dynamic to static incremental modulus with frequency was similar to that observed in the femoral arteries by Bergel (J. Physiol., 156, 458-469 (1961)).

Notizen: This study identified the effects of various manufacturing processes on the crystalline microstructure, mechanical properties, and biocompatibility of a polyvinylidene fluoride (PVDF) suture. To achieve this, changes in the crystalline microstructure and the tensile behavior of PVDF monofilaments were monitored in vitro after different thermal processing, coloration, and sterilization treatments. In addition, the in vivo biocompatibility of the manufactured and sterilized PVDF suture was assessed by using it to anastomose a preclotted polyester vascular prosthesis as a thoracoabdominal bypass in a series of dogs. The tissue response was followed by histologic and scanning electron microscopy over implantation periods ranging from 4 h to 6 months. Differential scanning calorimetry and infrared spectroscopy (FTIR-ATR) showed that thermal processing and the addition of a coloring agent had a direct effect on modifying the crystalline microstructure and hence changing the mechanical properties. For example, thermal processing converted some of the α phase into the β and γ polymorphs, whereas coloration led only to a major increase in the β-to-α ratio. The tensile properties were found to be optimized when the relative proportion of the β and γ phases combined compared to the α form gave rise to an FTIR A509/A532 absorption ratio between 4.0 and 4.5. Sterilization was found to cause some modifications to the crystalline microstructure near the surface of the monofilaments, but it did not change their mechanical properties. Pathologic examination of the anastomotic regions after different periods of implantation revealed a minimal cellular response, with no mineralization, intimal hyperplasia, or excessive fibrous tissue reaction. This good biocompatibility, together with other desirable characteristics such as ease of manipulation and satisfactory mechanical strength, makes PVDF an attractive alternative monofilament suture material for cardiovascular surgery. © 1995 John Wiley & Sons, Inc.
