    Notes: A method of analyzing volatile organic sampling train (VOST) samples by megabore capillary gas chromatography/mass spectrometry has been developed to enhance compound resolution and to improve the characterization of products of incomplete combustion in stack emissions. Several analytical systems were evaluated. The combination of components which gave the best overall performance consisted of a clamshell oven used to desorb the Tenax® and/or Tenax®/charcoal VOST cartridges onto an analytical trap in a commercial purge and trap unit. The purge and trap unit thermally desorbed the contents of the analytical trap onto a megabore capillary column installed in a gas chromatograph where the VOST target compounds were characterized and quantified by a computerized mass spectrometer. All VOST target compounds showed response factors with coefficients of variation of less than 25% for triplicate analyses at four concentrations. The time of analysis using the capillary column was 30% faster than the present VOST protocol that requires packed-column gas chromatography.

    Notes: Polarized Raman spectra have been obtained from single microcrystals of the duplex of the decamer d(A5T5)2 using a Raman microscope. This is the first report of Raman spectra from a crystal of a deoxyoligomer that contains only long, nonalternating sequences of adenine and thymine. Sequences containing d(A)n and d(T)n are of interest in view of recent suggestions that they induce bends in DNA and that they might exist in a nonstandard B-conformation. Polarized Raman spectra of a crystal of d(pTpT) have also been obtained. Both crystals display Raman bands whose intensities are very sensitive to the orientation of the crystal with respect to the direction of polarization of the incident laser beam. These spectra indicate that the helical axes of the oligonucleotides are parallel to the long axes of the crystals and that the d(A5T5)2 is not appreciably bent in the crystal. The Raman spectrum from the d(pTpT) crystal indicates that all of the furanose ring puckers are in a C2′-endo configuration since only the C2′-endo marker band at 835 ± 5 cm-1 is present. Crystals of d(A5T5)2 show measurable Raman intensities in both the 838- and 816-cm-1 bands. This indicates the presence of both the C2′-endo and C3′-endo, or possibly other non-C2′-endo, furanose conformations. The 816-cm-1 band is weak so that only a small fraction of the residues are estimated to be in the non-C2′-endo conformation. In both the d(pTpT) and d(A5T5)2 crystals the intensity of the bands due to vibrations of the backbone show only a small dependence on orientation of the crystals. This result is explained by the low symmetry of the puckered sugar rings. It is concluded that Raman spectra obtained from oligonucleotide crystals in which the orientation of the crystal axes to the laser polarization is not carefully controlled may contain intensity artifacts that are due to polarization effects.

Acylation of Trimethylsilyl Enol Ethers with Malonyl Dichloride  -  Synthesis of 4-Hydroxy-2H-pyran-2-onesTrimethylsilyl enol ethers of aldehydes 1, acyclic and cyclic ketones 4 and 10, respectively, are acylated by malonyl dichloride (2) at room temperature or even at -60°C. After aqueous workup, the 6-unsubstituted 4-hydroxy-2H-pyran-2-ones 3 are obtained from 1-(trimethyl-siloxy)alkenes 1, whereas the 5,6-alkylene-bridged 4-hydroxy-2H-pyran-2-ones 11 are formed from 1-(trimethylsiloxy)cycloalkenes 10. Acylation of 2-(trimethylsiloxy)-1-propene (4a) and 3-(trimethylsiloxy)-2-pentene (4b) results in mixtures of 4-(trimethylsiloxy)-2H-pyran-2-ones 5 (main products) and phloroglucinol tris(trimethylsilyl) ethers 6 (by-products).
    Notes: (Trimethylsilyl)enolether von Aldehyden 1 und nichtcyclischen bzw. cyclischen Ketonen 4 bzw. 10 lassen sich mit Malonyldichlorid bei Raumtemperatur bzw. bereits bei -60°C acylieren. Hierbei entstehen nach wäßriger Aufarbeitung aus den 1-(Trimethylsiloxy)alkenen 1 die bislang nur schwer zugänglichen 6-unsubstituierten 4-Hydroxy-2H-pyran-2-one 3 und aus den 1-(Trimethylsiloxy)cycloalkenen 10 die 5,6-Alkylen-überbrückten 4-Hydroxy-2H-pyran-2-one 11. Die Acylierung von 2-(Trimethylsiloxy)-1-propen (4a) und 3-(Trimethyl-siloxy)-2-penten (4b) führt zu Gemischen aus 4-(Trimethylsiloxy)-2H-pyran-2-onen 5 (Hauptprodukte) und Phloroglucin-tris(trimethylsilylethern) 6 (Nebenprodukte).

Aus diesen Daten läßt sich eine experimentelle Energiehyperfläche konstruieren, die folgende Merkmale aufweist: 1) Die Grundzustandsenergie von endo-1 ist um 8 kcal mol-1 höher als die von exo-4. 2) Der wichtigste Reaktionsweg ausgehend von endo-1 ist die formal verbotene [2σ + 2π]-Cycloaddition zu 3, deren Mechanismus diskutiert wird. 3) Obwohl die Aktivierungsenergien der Wege 4→6 und 4→3 gleich sind, verläft die Reaktion zu 6 aufgrund des größeren A-Faktors rascher. 4) Der Tetracyclus 3 thermolysiert gemäß dem Prinzip der mikroskopischen Reversibilität über exo-4 zu dem Dien 6.
    Notes: The C8H10 hydrocarbons 1, 3, 4, and 6 have been thermolyzed in a static system and the Arrhenius parameters have been obtained. Calorimetric measurements have been carried out to determine the heats of formation. From these data an experimental energy hypersurface is constructed which shows the following remarkable features: 1) The ground-state energy of endo-1 is higher than that of exo-4 by 8 kcal mol-1. 2) The predominant reaction pathway of endo-1 is the formally forbidden [2σ + 2π] cycloaddition to 3, the mechanism of which is discussed. 3) Although the activation energies of the routes 4→6 and 4→3 are the same, the reaction yielding 6 is faster due to a sizeably higher A factor. 4) The tetracycle 3 chooses the microscopic reverse pathway, i.e. its thermolysis proceeds via exo-4 to give the diene 6.

    Notes: The inhibitory effect of phorbol dibutyrate (PDB) on B-cell stimulation was evaluated using a model in which activation is induced by modest doses of antiimmunoglobulin antibody (anti-lg) and progression to DNA synthesis is induced by cytochalasin. PDB preferentially inhibited anti-lg-induced activation and did so during brief (2 hr) preincubation with anti-lg. Activation was inhibited whether PDB was added before or shortly after anti-lg. Since activation for cytochalasin responsiveness appears to be mediated by Ca2+, the effect of PDB on the anti-lg-induced rise in intracellular Ca2+ was evaluated. PDB (and other phorbol esters that activate protein kinase C) inhibited the rise in Ca2+ normally associated with anti-lg treatment; moreover, PDB reversed an established anti-lg-induced Ca2+ response. These data suggest that phorbol esters inhibit B-cell activation by interfering with the elevated levels of intracellular Ca2+ produced by cross-linking of surface immunoglobulin by anti-lg. This could represent a “feedback inhibition” type of response, but it remains to be seen if this occurs under physiological conditions of protein kinase C activation.
