    Notes: Using an adaptive strategy, Chinese hamster ovary (CHO) cell lines were developed that are capable of robust growth in serum-free suspension culture. These preadapted derivatives of the commonly used strain of CHO cells (CHO DUKX), termed PA-DUKX, were used for the introduction and stable expression of several heterologous human genes. A significant advantage of recombinant PA-DUKX cells was their ability to readily resume growth in serum-free suspension culture after transfection and amplification of heterologous genes. Expression of recombinant human proteins in PA-DUKX cells was quantitatively similar to that of lineages generated using conventional CHO DUKX cells. In addition, recombinant human proteins expressed by transfected PA-DUKX lineages were shown to be biochemically and structurally similar to those expressed in CHO DUKX cells, PA-DUKX host cell technology provides an opportunity for reducing the time and resources required to develop large-scale, suspension culture-based manufacturing processes employing serum-free medium. © 1996 John Wiley & Sons, Inc.

    Notes: The dielectric and optical characteristics of a sample of cellulose acetate (DS = 2.45) in dioxane solutions were studied at 10-50°C of concentration 10-50 wt% to include both isotropic and anisotropic phases. The study showed that the loss maximum, εmax″, magnitude of polarization, (ε0 - ε∞), static dielectric constant, ε0, time of relation, (2πfm)-1, and refractive index, nD, steadily increase with concentration up to the critical concentration (41 wt%) and then decrease. The mean-square dipole moment, 〈gμ2〉, decreases steadily up to the critical concentration then remains nearly constant, indicating that the isotropic solution changes to anisotropic, with smaller mean-square dipole moment. Comparison between the results of cellulose acetate (CA) and those of hydroxypropyl cellulose (HPC) reveals that, at the critical concentration in dioxane, the cholesteric structure of HPC possesses a greater mean-square dipole moment with higher temperature coefficient than does CA. The activation energy of the relaxation process for hydroxypropyl cellulose is higher, indicating a greater intrachain interaction compared with cellulose acetate.

    Notes: In this work, optical properties of samples of nylon 66 fibers were quenched in coarse-grained ice after annealing in the temperature range 80-180°C for 1-10 h. Two independent techniques were used to study optical parameters in these fibers. The first technique is the application of the diffraction technique to study the effect of quenching on the swelling factors of these fibers for some different liquids at room temperature of 25 ± 1°C. The second method dealt with the application of multiple-beam Fizeau fringes in transmission to estimate the refractive indices and birefringence of the skin and core of these fibers. The obtained results were utilized to calculate the polarizability per unit volume, isotropic refractive indices, and the Cauchy's dispersion constants. The relationship between n∥,⊥s, n∥,⊥c, n∥,⊥a, Δns, Δnc, Δna, n(iso)s, n(iso)c, and n(iso)a different annealing times and temperatures is given for these fibers. Microinterferograms and curves are also given. © 1996 John Wiley & Sons, Inc.

    Notes: Electroblotting proteins separated by gel electrophoresis provides a suitable support for further manipulations and analysis of small amounts of relatively pure samples. On-membrane digestion, peptide mapping by mass spectrometry, and database searching offer sensitive and fast tools to identify the analyte. By providing sequence information, tandem mass spectrometry can go a step further, confirming the database identification, solving problems connected with post-translational modifications and sequence variations, or supplying the stretches of internal sequence necessary to synthesize an oligonucleotide probe for gene isolation. The viability of this approach was successfully evaluated using different tandem mass spectrometric techniques: metastable decomposition in a matrix-assisted laser desorption/ionization (MALDI) time-of-flight instrument with a curved-field reflectron; low energy collision-induced dissociation in a MALDI quadrupole ion trap mass spectrometer; and high energy collision-induced dissociation in a high-performance four-sector mass spectrometer with massive cluster-impact ionization.

    Notes: Nylon 66(ICI Polyamide 66, Carded & Combed Top) fibers were annealed in the temperature range 80-180°C for 1-10 h. Refractive indices and birefringence were measured interfer-ometrically. Two independent techniques were used to study the optical anisotropy in these fibers. The first technique was to study the effect of annealing on the swelling properties of fibers from its diffraction pattern using a He - Ne laser beam. The second was the application of multiple-beam Fizeau fringes in transmission to determine the skin and core refractive indices and double refraction of annealing samples. The application was carried out using multiple-beam Fizeau fringes in transmission to determine Cauchy's constants and the dispersive coefficient for the fiber layers. The resulting data were used to calculate the mean polarizability per unit volume and the isotropic refractive indices. Behavior of optical properties at different annealing conditions is discussed. The results obtained clarify the new reorientation, and each layer change occurred due to annealing at different conditions.     Notes: Plasma coating (deposition of polymer under the influence of plasma) is utilized to modify the surface properties of corneal contact lens. An ultrathin layer (thickness of roughly 200 Å) of plasma polymer of acetylene/H2O/N2 is applied to poly(methyl methacrylate) (PMMA) corneal contact lenses. The surface becomes highly wettable with water compared to uncoated lens. When coated and uncoated contact lenses are placed on rabbit eyes, a remarkable difference in accumulation of mucous matter is observed. With the control lenses, the accumulation of mucous matter in a week is sufficient to affect the optical clarity of the lenses, whereas the coated lenses show no change after three months continuous wearing. The comparative degree of adhesion of the corneal epithelium cells onto glass, modified glass, PMMA, and coated PMMA surfaces is studied using tissue cultures and phase contrast microscopy. The coated PMMA surface exhibits a degree of tissue adhesion lower than that of control PMMA and higher than that of glass surface, and no sign of toxicity of the coated surface is observed by the tissue cultures.

    Notes: Over a six-year period, 530 fire fatalities resulting from 398 fires were studied in the State of Maryland. The study had two major objectives: (1) to determine the specific cause of death by a detailed autopsy stud of fire victims, and (2) to determine the specific cause of fatality-producing fire by an on-the-scene fire investigation. The fire fatality study was limited to residential fires and to fatalities that occurred within 6 h of the fire. The results of the toxicological analysis show that (1) 60% of the victims had a carboxyhemoglobin value greater than or equal to 50% carbon monoxide saturation, (2) an additional 20% had elevated carboxyhemoglobin with preexisting cardiovascular disease, (3) 11% of the victims had severe burns, (4) 9% were unexplained and (5) 40% of the victims had positive blood alcohol levels with 30% of these meeting the legal definition of intoxication (blood alcohol ≥0.1%). The fire investigations confirmed that the predominant fatal scenario is the cigarette ignition of upholstered furniture or bedding. This scenario accounted for 47% of the fires and 44% of the victims. Alcohol also appears to be significant factor in this scenario.

    Notes: Two-dimensional numerical simulations of laminar natural convection in a partially cooled, differentially heated inclined cavities are performed. One of the cavity walls is entirely heated to a uniformly high temperature (heat source) while the opposite wall is partially cooled to a lower temperature (heat sink). The remaining walls are adiabatic. The tilt angle of the cavity is varied from 0° (heated from left) to -90° (heated from top). The fast false implicit transient scheme (FITS) algorithm, developed earlier by the same authors, is modified to solve the derived variables vorticity-streamfunction formulation. The effects of aspect ratio (AR), sink-source ratio and tilt angle on the average Nusselt number are examined through a parametric study; solutions are obtained for two Grashof numbers, 105 and 107. Flow patterns and isotherms are used to investigate the heat transfer and fluid flow mechanisms inside the cavity. © 1998 John Wiley & Sons, Ltd.

    Notes: The model systems of molecular nitrogen fixation [N2 + H]⋅, [N2 + H]+, [N2 + H]-, [N2 + H2], [N2 + H2]+, and [N2 + H2]- were studied by the semiempirical INDO method. The study was based on the formal analogy between the catalytic reactions and the photochemical, radical, and ionic reactions on the other side. Symmetrical and donor-acceptor properties of necessary catalytic systems were proposed using the dependence of energy characteristics and electron structure on reaction coordinate. On the basis of this MO study we have proposed the appropriate symmetry types of catalysts for each of acceptable models of nitrogen fixation. For one of the proposed systems there was realised a model MO computation with explicit inclusion of atoms of transition metals (Fe, V).
