Notizen: Abstract Reduced fertility for a disease can greatly influence the estimation of morbid risk in various classes of relatives. The strongest effect is on parents of probands. Essen-Möller [(1955).Acta Genet. 5:334–342] derived an approximate correction for the morbid risk to parents and applied it to a sample of schizophrenics. That reduced fertility can also diminish the observed morbid risk to other classes of relative, such as siblings, grandchildren, niece/nephew, etc., seems to have been unappreciated in the literature. This report gives a simple method for correcting the morbid risk estimate to parents under reduced fertility which requires fewer assumptions than the method of Essen-Möller (1955). In addition, a method is derived for determining the potential effect of reduced fertility on morbidity risk estimates for other classes of relatives.

Notizen: Abstract Segregation analysis was performed on a large body of data on hand preference in nuclear families. The results suggest that the pattern of inheritance is consistent with Mendelian transmission, with different penetrance parameters for males and females. Although a single-locus model was more likely, a polygenic alternative could not be rejected. The parsimonious model specified a recessive right-shift gene of frequency 0.748. All females with the right-shift genotype become right-handed, while a small percentage (4.2%) of males with the right-shift genotype becomes left-handed (sporadic left-handers). Goodness-of-fit tests of simple genetic models were performed on the current data set as well as six others from the literature. Results suggest an adequate fit of both single-locus and polygenic models in nearly all data sets, with reasonable consistency among the parameter estimates. Furthermore, the observed generational effect in hand preference (reduction in left-handedness in parents compared with offspring) is found to be consistent with the relaxation of cultural pressure over time. Also, the simple genetic models of hand preference provide an acceptable alternative to maternal transmission, which has previously been suggested.

Notizen: Summary Assuming a four strand model and no chromatid interference, lack of chiasma interference is known to be equivalent to the assumption that the formation of chiasmata follows a Poisson process. We prove that lack of chiasma interference is also equivalent to the assumption that a random gamete shows recombination on any given interval of a chromosome independently of recombination on all disjoint intervals. Both assumptions are sufficient, but not necessary, for Haldane's formula relating recombination to map distance to be true, as we demonstrate by specific counterexamples. These issues are discussed in the context of the theory of stochastic point processes.
