    Notes: Summary Mechanically skinned fibre preparations from the extensor digitorum longus muscle of the rat were used to test whether a rise in myoplasmic [NH4 +] in the range 2–10 mm interferes with the mechanism of excitation-contraction coupling in fast-twitch mammalian muscle. Under our conditions (pH 7.10, Mg2+ 1 mm, temperature 23° C), [NH4 +] up to 10 mm had little effect on the Ca+-activated force and on the peak of the t-system depolarization-induced force response. However, the duration of the depolarization-induced force response was decreased significantly at [NH4 +] ≥2 mm. From these data we conclude that the intracellular accumulation of NH4 + is not likely to play a major role in fatigue. Nevertheless, the build up of NH4 + during fatigue, may have a significant inhibitory effect on the force output by decreasing the duration of the t-system depolarization-induced activation of the contractile apparatus.

    Notes: Abstract The main objective of this study was to analyse glycogen in single muscle fibres, using a recently developed microfluorometric method which detects subpicomol amounts of NADPH, glucose and glycogen (as glucosyl units) (detection limit 0.16–0.17pmol in a 25nl sample) without fluorochrome amplification. The fibres were freshly dissected from the twitch region of the iliofibularis muscle of the cane toad (Bufo marinus), and were mechanically skinned under paraffin oil to gain access to the intracellular compartments. The results show that: (1) glycogen concentrations in toad skeletal muscle fibres range between 25.8 and 369mmol glucosyl units/litre fibre volume; (2) there is a large variation in glycogen content between individual fibres from the iliofibularis muscle of one animal; (3) there are seasonal differences in the glycogen content of toad single muscle fibres; (4) the total amount of glycogen in single muscle fibres of the toad does not decrease significantly when storing the tissue, under paraffin oil, at 20–25°C for up to 6h or at 4°C for up to 24h; and (5) 15–26% of fibre glycogen can be washed in an aqueous solution at pH 5–7, within 5min, while 74–85% of fibre glycogen remains associated with the washed skinned fibre, even after 40min exposure of the skinned fibre preparation to the aqueous environment. The retention of most glycogen in the fibre preparation after mechanical removal of the plasma membrane and extensive washing indicates that in toad skeletal muscle fibres the largest proportion of glycogen is tightly bound to intracellular structures. The results also show that the skinned muscle fibre preparation is well suited for microfluorometric glycogen determination, since low molecular weight non-glycogen contributors to the fluorescence signal can be removed from the myoplasmic space prior to the glycogen hydrolysis step.

    Notes: An organism diversity index for use with mixed liquor or wastewater samples was developed to assess the species richness and diversity of activated sludge. The index was used in several studies and was found to be fast and simple to perform using basic laboratory equipment. Two bench-scale and two pilot-scale studies found that the resulting ‘organism diversity index’value was a good indicator of process performance and was not influenced by the total concentration of the mixed-liquor suspended solids but by their nature alone. The technique rapidly yielded pertinent information about the health of the sludge and could be used instead of genetic investigations to obtain population information quickly enough for wastewater-treatment plant process control.

    Notes: Tax Increment Financing (TIF) is a popular yet controversial tool that allows local governments to use property tax revenue to fund the public costs of economic development. Since TIF gives one local government the power to affect the tax bases of the overlapping jurisdictions, there is uncertainty and argument on the part of government officials and taxpayers as to who really finances the program. To evaluate the alternative contentions, this paper presents a general methodology that identifies which taxpayers in which locations fund the TIF's expenditures, and sets forward the conditions under which such a local economic development policy can be beneficial to taxpayers. The paper applies the model to study the TIF program currently active in downtown Des Moines, Iowa. The evidence indicates that the taxpayers in the entire metropolitan area subsidized the downtown activities in the early years, but now pay lower property tax rates due to the city's TIF-financed urban revitalization program.

    Notes: A simplified model of domain rearrangement in multidomain particles following the application of an alternating (AF) or direct (DF) field is used to quantify the dependence of anisotropy of magnetic susceptibility on magnetic history. To be able to account for changes in the average susceptibility (i.e. The average of three mutually perpendicular susceptibility measurements) of the sample following field treatment, the model has to be refined by introducing interaction between domains. By fitting theoretical curves of anisotropy versus peak AF to experimental results for samples containing different size fractions of magnetite particles, of average grain size ranging from 0.7 to 58 μm, four relevant parameters can be computed (for each sample), which enables a good fit to be obtained. By allowing two of these parameters to change systematically when a DF is applied (and an IRM is acquired), the anisotropy dependence on direct field strength can also be modelled. This includes the sign change observed as the field-impressed ellipsoid changes from prolate to oblate, together with increases in the average susceptibility as the field increases. However, some details, such as discrepancies between predicted and actual increases in the average susceptibility at high fields, suggest that further refinement of the model is required.

    Notes: Many interesting inverse problems in geophysics are non-linear and multimodal. Parametrization of these problems leads to an objective function, or measure of agreement between data and model predictions, that has a complex topography with many local minima. Optimization algorithms that rely on local gradients in the objective function or that search the model space locally may become trapped in these local minima. By combining simulated annealing with the downhill simplex method, a hybrid global search algorithm is presented in this paper for non-linear, multimodal, inverse problems. The hybrid algorithm shares the advantages of both local search methods that perform well if the local model is suitable, and global methods that are able to explore efficiently the full model space. The hybrid algorithm also utilizes a larger and more complex memory to store information on the objective function than simulated annealing algorithms. The effectiveness of this new scheme is evaluated in three problems: minimization of the multidimensional Rosenbrock function, non-linear, 1-D, acoustic waveform inversion, and residual statics. The performance of the hybrid algorithm is compared with simulated annealing and genetic algorithms and is shown to converge more rapidly and to have a higher success rate of locating the global minimum for the cases investigated.

    Notes: The effects of soil fertility (two levels of soil nitrogen and two levels of soil phosphorus) and mycorrhizal infection on pollen production and pollen grain size were studied in two cultivars of the common zucchini (Cucurbita pepo). Overall, soil fertility and mycorrhizal infection had significant effects on traits affecting the male function of plants (staminate flower production, pollen production per flower and pollen grain size). There were also differences between the cultivars for these male traits in all three experiments. In addition, pollen grain size decreased toward the end of the growing season. In the mycorrhiza experiment, both phosphate concentration per pollen grain and total phosphate content per anther were greater but not significantly greater in the mycorrhizal plants than in the non-mycorrhizal plants. A significant negative relationship between pollen production and pollen grain size was found in the mycorrhiza and soil phosphorus experiments, indicating that there was a trade-off between pollen production and pollen size. This study is the first to show that mycorrhizal infection has an effect on male function (pollen production and size) in addition to the well-documented effects on female function (fruit/seed production and size).

    Notes: Broccoli is well recognized as a source of glucosinolates and their isothiocyanate breakdown products. Glucoraphanin is one of the most abundant glucosinolates present in broccoli and its cognate isothiocyanate is sulphoraphane, a potent inducer of mammalian detoxication (phase 2) enzyme activity and anti-cancer agent. This study was designed to measure: glucosinolate levels in broccoli florets from an array of genotypes grown in several environments; the elevation of a key phase 2 enzyme, quinone reductase, in mammalian cells exposed to floret extracts; and total broccoli head content. There were significant environmental and genotype-by-environment effects on levels of glucoraphanin and quinone reductase induction potential of broccoli heads; however, the effect of genotype was greater than that of environmental factors. The relative rankings among genotypes for glucoraphanin and quinone reductase induction potential changed, when expressed on a per head basis, rather than on a concentration basis. Correlations of trait means in one environment vs. means from a second were stronger for glucoraphanin and quinone reductase induction potential on a per head basis than on a fresh weight concentration basis. Results of this study indicate that development of a broccoli phenotype with a dense head and a high concentration of glucoraphanin to deliver maximum chemoprotective potential (high enzyme induction potential/glucoraphanin content) is a feasible goal.

    Notes: The Devonian of East Greenland comprises a thick sequence of continental clastic sediments infilling an extensional basin. West of the main basin bounding fault (Western Fault Zone) are scattered outliers of Devonian conglomerate which accumulated in small basins such as found on the island, Ella Ø. The Ella Ø Basin formed by extensional movement along the Narhval Sund Fault accompanied by the formation of a prominent bedding parallel detachment surface which was subsequently modified by sub-aerial exposure to become the unconformity surface. Mapping of this unconformity surface shows major vertical relief. A thick sequence of conglomerate occurs on Ella Ø, which, close to its exposed basal unconformity, has three lacustrine beds intercalated within it. Detailed analysis of one lake unit shows it to have significant lateral variation. At proximal localities it largely comprises lacustrine turbidites, whereas more distal locations were within a stratified lake. The interpreted sequence of events on Ella Ø is an interval of fluvial sedimentation followed by rapid drowning of the topography with lacustrine sediments onlapping onto basement. After an interval of deeper lacustrine sedimentation including laminites, the lake shallowed, the lithology changed to limestone and the lake dried out. Conglomerate deposition then recommenced. Maximum lake water depth of 100 m is estimated following correction (7°) for post-Devonian rotation, both determined using the distribution of lacustrine sediments on the unconformity surface. The preferred mechanism for flooding of the Ella Ø Basin is episodic flooding of the entire basin system. Lacustrine sediment preservation results purely from its deposition amongst topography at the edge of the active fluvial system. In such fluvial systems lacustrine sediments may be part of the normal sedimentary cycle but were almost always removed by reworking.
