    Notes: Abstract Horse serum is shown to contain a soluble class I molecule analogous to the secreted Q10 molecule in the mouse. This molecule has several similarities to the recently described mouse Q10 molecule: (1) it is smaller than membrane-bound equine class I molecules; (2) it occurs in a high molecular mass complex of 200–300 kd in serum; and (3) the serum levels of the equine molecule are similar to that of the Q 10 molecule (about 30 μg/ml). A soluble molecule is also detected in the sera of species related to the horse; it has in fact been found in all the wild members of the order Perissodactyla so far tested. However, it was not detected in the serum of members of the orders Carnivora, Sirenia, Proboscidea, Artiodactyla, and Primates that were tested, nor in the serum of members of the order Rodentia other than in that of the genus Mus.

    Notes: Abstract The complete amino acid sequence of the CNBr fragment comprising residues 229–284 of the murine major histocompatibility complex antigen H-2Db has been determined using radiochemical methodology. The sequence was determined by N-terminal sequence analysis of the intact CNBr fragment and by sequence determinations of peptides derived from this fragment by trypsin and staphylococcal V8 protease cleavage. In addition to the amino acid assignments for H-2Db, it was possible to assign the linkage position of the third N-linked glycosyl unit to the asparagine at residue 256. Additional amino acid sequence assignments have also been made for three other CNBr fragments that span residues 99–138, 139–228, and 308–331 of the H-2Db molecule. The total protein sequence information available (222 of 338 residues) agrees in every comparable position with the protein sequence derived from the cDNA clone (pH203) isolated by Reyes and co-workers (1982b), which strongly suggests that this clone encodes H-2Db. Combination of the protein sequence with that deduced from the cDNA clone provides the complete H-2Db protein sequence. Comparison of this sequence with other available protein sequence information for murine class I molecules has revealed protein sequences that may be unique to either K or D region molecules.

    Notes: Abstract Monofluoro- and monochlorobenzoates did not support the growth of Pseudomonas PN-1, either aerobically or anaerobically (nitrate respiration), when supplied as sole sources of carbon and energy. Anaerobic growth yields on nonfluorinated substrates were increased by p-fluorobenzoate (pFBz) with a utilization of pFBz and release of F-. Cell suspensions grown on p-hydroxybenzoate (pOHBz), either aerobically or anaerobically, only degraded o-fluorobenzoate (oFBz) and pFBz of the monohalogenated benzoates tested. Both compounds were catabolized anaerobically, but not aerobically, with a release of F-. oFBz was immediately attacked, by cells grown anaerobically on pOHBz, whereas pFBz was only degraded after a lag phase; chloramphenicol inhibited the breakdown of pFBz, but not oFBz, thereby indicating the need for additional enzyme(s) to attack pFBz. o-Chlorobenzoate (oClBz) inhibited the anaerobic, but not aerobic, oxidation of pOHBz and stopped anaerobic growth on pOHBz. A mutant was isolated which metabolized pOHBz in the presence of oClBz but it was defective in its anaerobic metabolism of benzoate (Bz). Comparative studies, of the mutant and Pseudomonas PN-1, indicated that the mutation involved a metabolic site common to Bz, oClBz and the monofluorobenzoates. The dependence of the oxidation rate of Bz and oFBz on their concentrations at a millimolar level, in the mutant but not Pseudomonas PN-1, suggested a defect at the permease level: the uptake of 14C-labelled Bz by the mutant was also concentration-dependent. The response of the organism to the inhibitory effect of oClBz on pOHBz catabolism is discussed with respect to its significance in the perturbation of natural degradative processes by unnatural chemicals in the environment.

    Notes: Abstract Natural soils on steeply sloping landscapes in the Appalachian coal fields of Virginia. West Virginia. Kentucky, and Tennessee are often thin, rocky, acidic and infertile, making the topsoiling of surface mined sites impractical in many cases. Topsoil substitutes composed of blasted rock fragments are commonly used in this region. The proper selection and placement of designated topsoil substitutes is therefore critical to long term reclamation success. These mine soil surfaces are not in equilibrium and with the surface environment, and it is quite difficult to diferentiate among dissolution, adsorption, desorption and precipitation reactions as these surfaces weather with time. Severe compaction limits the productivity of many otherwise suitable topsoil substitutes. A minimum non-compacted thickness of 1 m is desirable to insure long run mine soil productivity for a variety of post-mining land uses. Significant changes in the physical, chemical, and mineralogical properties of mine soils occur within one year after placement. Mine soils high in silt content often form hard vesicular surface crusts, particularly when left unvegetated. The long term survival of plant communities on these mine soils is dependent upon mine soil organic matter accumulation and N and P cycling. Little is currently known about N and P dynamics in these mine soils, but P-fixation is a profound problem in high Fe3- spoils. Revegetation practices that were designed to meet 2-year bond release requirements may not he sufficient to meet new 5-year release standards. Hard rock derived mine soils can often equal or exceed native topsoil in productivity and post mining land use potential.

    Notes: Abstract Factor analysis of questionnaire responses produced by over 6,000 individuals revealed parental reactions to good and bad grades grouped themselves into five categories: (1) Grades are important to me; bad ones lead to negative consequences; (2) Good grades are important to me, but I will support you regardless of grade; (3) Bad grades make me mad; (4) Do your best, but there is more to life than grades; and (5) Grades are important; you will know how I feel on the basis of a joking comment. When these patterns were related to reports of specific student actions in college, such as getting good grades, dropping a course, or cheating on an examination, results indicated only a few significant negative correlations, and only for students whose parents reacted to bad grades in a negative emotional way; i.e., in terms of categories 1 and 3. More benign reactions - categories 2, 4, and 5 - did not correlate either positively or negatively with student actions or grades. Results were taken to suggest that parental reactions to grades are not only attempts at changing student behavior but also may be viewed as emotional responses expressing personal needs and values.

    Notes: Abstract This paper describes a recent effort to infuse the Total Quality Improvement (TQI) approach, popularized by Deming and others, into an upper-division, junior-senior economics course at the University of Wisconsin-Madison. The process of infusing TQI into instruction has received relatively little attention. Most efforts to bring TQI into higher education focus on improving administrative operations and establishing courses and programs for students to learn how to apply TQI in their future jobs. The challenge is in using TQI to help students realize their potential for learning in traditional courses. The TQI instruction approach developed for use in the course had three major elements. Customer Focus is represented by the proficiencies emphasized in the course and expected of graduating economics majors. Student Involvement is represented by team-centered research projects whose purpose is to enhance the proficiency of creating new knowledge. Continuous Improvement is represented by ongoing student evaluations of the course and instructor, carried out by a student team. After discussing the motivation for adopting this approach and the independent development of the concept of proficiencies in the economics major, the paper moves on to discuss the planning, implementation, and execution of the course. Particular attention is given to discussing the operation of the teams and the development of a multi-faceted ongoing evaluation process to assess the effectiveness of the TQI Instructional Approach. The evaluation results indicate strong student satisfaction with the TQI orientation of the course, while at the same time pointing out ways of improving what was done.

    Notes: Abstract Reduction of the e-isoprene unit of polyprenols to form dolichols was studied in vivo using3H-polyprenol derivatives as substrates and liposomes as carriers. Liposomes containing labeled polyprenol, polyprenyl phosphate, or polyprenyl pyrophosphate were injected through the portal vein into the livers of rats under anesthesia. Uptake and conversion of the labeled compounds to dolichol derivatives was studied at different intervals. The greatest conversion to dolichol derivatives was found with polyprenyl pyrophosphate and polyprenyl monophosphate, with 31% and 8% of the absorbed dose converted respectively. Less than 0.2% of the absorbed polyprenol was converted to dolichol derivatives. These results suggest that the substrate for the α-isoprene reductase involved in dolichol biosynthesis is either polyprenyl monophosphate or polyprenyl pyrophosphate, or both.
