    Notes: Abstract Limited feeding of nitrate during culture of Nannochloris sp. UTEX LB1999 for intracellular lipid and triglyceride accumulation was investigated with the aim of obtaining cells superior for liquefaction into a fuel oil. The intracellular lipid contents and the percentage of triglycerides in the lipids of cells grown in a nitrogen-limited medium (0.9 mM KNO3) were 1.3 times as high as those grown in a modified NORO medium containing 2.0–9.9 mM KNO3. However, the cell concentration was too low for the practical production of fuel oil by high-pressure liquefaction of the cell mass. A single feeding of 0.9 mM nitrate after nitrate depletion during cultivation in a nitrate-limited medium increased the cell concentration to twice that obtained without such feeding, and the lipid content was maintained at a high level. The timing of nitrate feeding, i.e., whether it was given during the log phase (before nitrate depletion), the constant growth phase (just after the depletion), or the stationary phase (after the depletion), had negligible effect on the intracellular lipid content and percentage of triglycerides in the lipids. When 0.9 mM nitrate was intermittently fed ten times during the log phase in addition to the initial nitrate feed (0.9 mM), the cell concentration reached almost the same (2.16 g/l) and the intracellular lipid content and the percentage of triglycerides in the lipids increased from 31.0 to 50.9% and 26.0 to 47.6%, respectively, compared with those of cells cultured in a modified NORO medium containing 9.9 mM KNO3 without additional nitrate feeding.

    Notes: Abstract A CH3OH-utilizing bacterium that has the ability to produce extracellular polysaccharide (EPS) was isolated from a soil sample, and was identified as the obligate methylotroph Methylobacillus sp. strain 12S on the basis of its 16S rDNA sequence and growth-substrate specificity. The EPS produced by strain 12S was purified and the sugar composition was analysed by GC-MS and HPLC to reveal that the EPS was a heteropolymer composed of glucosyl, galactosyl, and mannosyl residues in the molar ratio 3:1:1. In order to produce mono- and/or oligosaccharides by single-step fermentation from CH3OH, stain 12S was mutagenized by transposon 5. Among eleven EPS-deficient mutants, three strains were found to accumulate significant amounts of reducing sugars in the media. The amounts of the reducing sugars produced by the mutants (〉ca. 700 mg glucose equivalent/l) were 〉11–22 times higher than those produced by the wild-type strain (〈ca. 60 mg glucose equivalent/l). The GC-MS analysis showed that all the mutants accumulated glucose, erythrose, threose and a disaccharide-like compound in the media.

    Notes: Abstract Electric spatial patterns of bands formed along the cell wall of the characean internode were studied using a multi-electrode measuring system. The electric potential near the surface of the cell was measured by arranging about 25 electrodes along the cell at approximately 1.6 mm intervals. Since the time required for one scan over the cell length is only 1 s, the temporal change in the spatial pattern of surface electric potential can be readily observed. Oscillations were sometimes found as the electric pattern started to appear after the cell was illuminated. Fourier analysis shows that a single spatial mode arises gradually and then becomes stabilized in an oscillatory manner. A simple electric circuit model comprising three variables, i.e., a membrane potential, an electric current across the membrane and an electromotive force, can simulate well the oscillatory rise of bands. These results imply that the electric spatial pattern observed in characean internodes is a self-organized structure emerging far from equilibrium, known as a dissipative structure. Biophysical mechanisms of band formation are discussed.

    Notes: Abstract Cell cultures of Perilla frutescens, growing on Linsmaier and Skoog medium, released more anthocyanin with 40 to 50 g sucrose/l compared with the control of 30 g sucrose/l of medium. More proteins were also released into the medium with the higher sucrose concentration and the cell volume was smaller, suggesting that the higher osmotic pressure, caused by the high sucrose concentration, may cause these releases. The capacitance of the cultured cells indicated a difference in membrane structure between the cells cultivated with different sucrose concentrations, supporting the hypothesis that cell permeabilization is increased at higher sucrose concentrations.

    Notes: Abstract A truncated Man9-mannosidase gene and a full-length β-1,4-galactosyltransferase gene were isolated from human kidney and placenta cDNAs, respectively. Both genes were cloned in plasmid pMAL-c2 to produce fusions with maltose-binding protein. Fusion products were purified by affinity chromatography. Purified enzymes were assayed using pyridyl-amino labelled oligosaccharides as substrates and analysed by HPLC.

    Description / Table of Contents: Résumé L'évolution chronologique au cours du compostage des facteurs importants (quantité de CO2 produite, température, poids, et teneur en eau) a été simulée par une analyse de regression linéaire. D'autre part, la variation de température pendant la première phase du compostage a pu être éstimée de façon précise d'après le bilan thermique de la production microbienne de chaleur, déterminé par analyse de regression linéaire. En combinant cette simulation avec la méthode Simplex, on a déterminé un taux optimum d'aération réduisant à 16,7 h le temps nécessaire pour que la température s'élève à 65°C, c'est à dire au niveau où la plupart des bactéries pathogènes du compost sont tuées.
    Notes: Summary The time courses for the important factors in the composting process for activated sludge, that is the amount of CO2 generated, the temperature, weight change and water content of the composting materials, were simulated by means of regression analysis. Furthermore, we could accurately estimate the temperature change of the composting materials in the early phase of composting from the heat balance including microbial heat generation estimated by regression analysis. Combining this simulation method with the Simplex method, we determined the optimum aeration regimen that minimized the time required for the temperature to increase to 65°C, at which most pathogens in compost materials are killed, to be 16.7 h.

    Notes: Abstract The effects of surfactants, adecanol LG-294 and silicone A, on anthocyanin accumulation and the growth ofPerilla frutescens cells in suspension cultures were studied. Production of the red pigment was remarkably reduced from about 1.9 g/l to 0.4 g/l by adecanol LG-294 at 0.06 ml/l but not by silicone A up to 0.4 ml/l. Several repeated shake-flask cultures also demonstrated no adverse effects of silicone A on the metabolite accumulation by the suspended cells. Furthermore, the addition of silicone A to a culture in a stirred bioreactor produced a three-fold higher growth rate and a seven-fold increase in anthocyanin compared with surfactant-free cultures. The improvement was due to the substantial reduction or prevention of foaming and of cell adhesion to the bioreactor wall.

    Notes: Abstract Streptomyces D3, derived from protoplast fusion between Streptomyces cyaneus 190-1 and Streptomyces griseoruber 42-9, has the ability to produce high levels of xylose isomerase when grown on hemicellulosic materials such as xylan as the carbon source. Comparison between the partial nucleotide sequences of the 16S ribosomal RNA genes from S. cyaneus 190-1, S. griseoruber 42-9, and fusant D3 showed that the 16S rRNA gene of fusant D3 was identical to that of S. cyaneus 190-1. Partial sequence analysis of the xylose isomerase genes also indicated that the gene of fusant D3 was identical to that of S. cyaneus 190-1. The partial DNA fragments for the xylanase genes (xlnA and xlnB) of fusant D3 were amplified by PCR, and subjected to Southern hybridization analysis. The results revealed that the xlnB gene of fusant D3 was similar to that of S. cyaneus 190-1, but that the xlnA gene of fusant D3 was similar to that of S. griseoruber 42-9. These results suggest that the majority of the genome of fusant D3 may be derived from S. cyaneus 190-1.

    Notes: Abstract A locally isolated strain, Bacillus subtilis NM-39, was selected as an active mannan-utilizing bacterium based on high saccharifying activities on coconut residue and locust bean gum galactomannan. The optimal pH and temperature ranges for activity of the crude enzyme were 5.0 to 6.0 and 50 to 60°C, respectively. The organism gave maximum mannanase activity when grown in liquid mineral salts medium containing 1% (w/v) each of coconut residue and soybean flour, as carbon and nitrogen sources, respectively, at pH 7.0 and in aerobic growth for 28 h at 37°C. High saccharifying activity on coconut mannan was also observed.

    Notes: Abstract We present an overview and instrumental details of the solar XUV Doppler Telescope (XDT) launched in January 1998 with the S520CN-22 sounding rocket of the Institute of Space and Astronautical Science. The XDT observes nearly single-temperature solar corona at 1.8 MK with angular resolution of ≈ 5′′ pixel size, together with the ability to detect the coronal velocity field with a full-Sun field of view. By use of normal incidence optics whose primary and secondary mirrors are coated with multilayer materials in two sectors, the XDT takes images of the Sun in a set of shorter and longer wavelength bands around the Fe xiv 211.3 Å emission line. Summation of a pair of images in the two bands provides an image of the 1.8 MK-corona while the difference between the two provides velocity images of the Fe xiv-emitting plasma. A brief description on the observation sequence together with the flight result is also given.
