    Notes: Summary The consumption of benzodiazepine drugs (BDZ) in Chile in the period 1982–1986 has been studied by the DDD method. National use was assessed using drug import forms as a source of information, assuming that the total amount imported was manufactured and consumed in the same year. The utilization did not show a constant pattern, being 32.7, 33.6, 50.2, 34.9, and 31.3 DDD 1000 per inhabitants per day in 1982, 1983, 1984, 1985 and 1986, respectively. During the study period, diazepam was the most commonly used agent amongst anxiolytic BDZ, reaching a peak of 27.1 DDD per 1000 inhabitants per day in 1984, and flunitrazepam was the most popular hypnotic, attaining its maximum in 1986 (6.4 DDD per 1000 inhabitants per day). The use of BDZ at community pharmacy level was also evaluated, employing the International Marketing System (IMS) as the source of information. At that level the pattern of utilization showed a constant increase during the study period, being 14.9 and 25.8 DDD/1000 inhabitants/per day in 1982 and 1986, respectively. In community pharmacies the anxiolytic BDZ most often consumed was diazepam (maximum 9.1 DDD per 1000 inhabitants per day in 1985), and the commonest hypnotic was flunitrazepam (peak 6.0 DDD per 1000 inhabitants per day in 1986). National consumption of BDZ appeared higher and more variable than use at the community pharmacy level, but in both there was greater usage of anxiolytic than of hypnotic BDZ, and diazepam and flunitrazepam were the most popular agents.

    Notes: Abstract Specimens of O. americanus collected in eight localities from Rio Grande do Sul and four from Paraná (Brasil) were studied. Patterns of chromosomal localization and activity of nucleolar organizer regions (NORs) were established using the silver banding technique. It was shown that diploid and tetraploid specimens from all localities have homologues of group 4 bearing active NORs. In addition, some populations showed NOR activity on only one homologue number 5 (Uruguaiana, Prudentópolis), 8 (Itaqui) or 2 (Pato Branco). Intrapopulation polymorphism was found, some specimens showing the extra band and others not. Most cells analysed from the intestinal epithelium of an individual showed the same particular staining pattern of NORs, which were unequally stained, suggesting differences in their activity. The tetraploid genomes showed a maximum of five and a minimum of two active NORs. Diploid genomes showed two active NORs and four in the case of one specimen from Pato Branco. Four diploid populations and sympatry of 2n and 4n animals in Santa Bárbara do Sul (RS) were detected for the first time in South Brasil. Karyological evolution, as well as the meaning of number and unequal activity of NORs on tetraploid genomes are discussed.

    Notes: Abstract Eucheuma uncinatum (Setch. & Gard.) Daw. from the Gulf of California was cultured in 1001 tanks in outdoor conditions. The cultures consisted of a 3 × 3 factorial design of 10, 19 and 33 % incident light and three different conditions of nutrients during winter (0; 20 µM N03; and 20:2 µM NO3:PO4) and summer (0; 40:4; and 80:8 µM of NO3:PO4). Best growth and carrageenan yield conditions were determined under each experiment. Carrageenan extracts were analyzed for total carbohydrates, 3,6-anhydrogalactose anhydrogalactose and sulphates. Higher carrageenan yields were obtained from cultures with fertilized plants under higher light conditions from winter and summer experiments although summer cultures provided the highest yields (48%). During summer a direct relationship between nutrients and sulfate content was observed. This effect was not observed in the winter experiment. An inverse carbohydrate: light relationship was observed in winter but not in summer. Maximum values of 3,6-anhydrogalactose in winter and summer were 23.9% and 18%, respectively. Results indicate that there is the potential to increase yield and quality of Eucheuma uncinatum carrageenan cultured under natural conditions.

    Notes: Abstract The concept of genetic conformity lies at the basis of the definition of essential derivation, or the process of using a protected variety (or `initial variety', IV) as the base to develop another similar variety (the essentially derived variety, EDV). Methods based on morphology, biochemistry or on molecular markers can be used to estimate genetic conformity. In this study, the capability of AFLP®1 markers to provide a reliable and meaningful estimate of genetic conformity of different varieties was investigated in diploid perennial ryegrass (Lolium perenne spp.), an allogamous species whose varieties are genetically heterogeneous. Twelve accessions with known breeding lineage, comprising five closely related groups, were included in the study. For the set of test accessions analysed, the AFLP protocol accurately reproduced the same relationships as were evident from examining their morphology and both these results were consistent with the relationships known to exist within the different test groups. Principal components analysis as well as cluster analysis associated unambiguously the IV and the EDV accessions. It was concluded that the methodology developed in this study could be used as a model from which to create a protocol for evaluating putative cases of essential derivation.

    Notes: Abstract In this study the morphological and protein diversity of twelve diploid perennial ryegrass accessions (Lolium perenne L.) was examined. These accessions comprised five closely related groups, each containing an `initial variety' (IV) and one or more declared `essentially derived varieties' (EDV), with differing degrees of relatedness. `Essential derivation' is a legal concept relating to intellectual property in plant varieties and is additional to Plant Breeders Rights (PBR). An EDV is defined as clearly distinct from, but conforming in its expression of the essential characteristics of an IV, from which it is found to have been predominantly derived. Where an essential derivation has been confirmed, the breeder of the IV may be entitled to some royalty sharing of the EDV. Clearly, therefore, in any successful EDV claim, evidence of a high degree of conformity in either the phenotype or genotype would be required. Examination of plant morphology indicated that all the EDVs were morphologically distinct from their corresponding IV in one or more morphological characteristic. using a principal co-ordinates analysis to give an overall measure of morphological difference, all twelve accessions were correctly clustered into their related groups and the magnitude of the differences within groups reflected their known breeding histories. Examining protein diversity by methods that targeted single- and multiple-locus genes also clustered the accessions into their correct groups, in most cases. However, only by examining diversity in seed storage proteins were within-group relationships accurately represented. The methods used provided no consistent representation of between-group relationships. It was concluded that the morphological method provided a creditable measure of genetic conformity, but to avoid incurring excessive time, work and cost, results from existing national PBR trials would need to be openly available. Within the limits of the genetic material examined, seed storage protein diversity appeared to provide a suitable combination of accuracy and efficiency on which to base a routine test. However, given more complex breeding relationships than those in this study, methods such as AFLP1 markers that sample more genetic diversity, may be necessary to maintain this level of accuracy.

    Notes: Abstract An evaluation was performed of the potential use of AFLP markers to reveal polymorphisms among Lolium perenne plants with different degrees of kinship. Radioactive and fluorescent detection techniques were applied. The use of a fluorescent detection approach contributed greatly to the speed and ease of conducting and interpreting the AFLP patterns. The great discriminative power of AFLP markers and their capacity to represent genetic relationships among ryegrass plants was shown. Despite the high polymorphic value of the AFLP markers, standard statistical tests could not differentiate between two gene pools derived from different breeding programmes. It proved also impossible to correlate fodder and turf phenotypes with AFLP distance data. A very important point revealed by our data is the high degree of genetic diversity within commercial ryegrass varieties. Our findings are relevant to any outcrossing crop with a breeding strategy based on the production of synthetic populations.
