    Notes: Abstract The rapid formation (in less than 45 min) of internal septa and the apparent subdivisionin situ of mitochondria from cardiac and skeletal muscle are described following a variety of experimental treatments. For example, the ionophore A23187, caffeine, DNP, ruthenium red, and the insecticide lindane have been applied to intact, glycerinated, and chemically skinned skeletal muscle fibers and to cardiac muscle strips from both amphibians and mammals. In some mitochondria, the two compartments are in the same configuration; in others they are different. The significance of these mitochondrial septa is discussed, and it is suggested that the findings are consistent with the hypothesis that a variety of experimental procedures can promote rapid mitochondrial division.

    Notes: Abstract Britain is renowned for low indoor winter temperatures and a high rate of excess winter deaths and a causal association has been drawn between the two. A lack of priority given to energy efficiency here is often justified in terms of the mild climate. However, it can be shown that British climatic features may be related to poor health as long as poor standards of construction and insulation prevail. Legislation should therefore be implemented to improve standards both for new housing and refurbishment of much existing housing stock, not only for reasons of energy conservation but also for community health.

    Notes: Summary Experimentally-induced rises in intracellular calcium ([Ca2+]i) promote rapid myofilament degradation in amphibian and mammalian cardiac muscle strips.     Notes: Abstract Water-soluble components of a nephrotoxic isolate of Penicillium aurantiogriseum have been fractionated by sequential ion-exchange, size-exclusion gel filtration, reverse-phase silica chromatography and HPLC. Nephrotoxicity in the rat was confined to a size-exclusion fraction approximating to 1500 daltons, which also inhibited DNA synthesis in cultured kidney cells. The more sensitive in vitro assay allowed toxicity to be followed to a sub-fraction from gradient-elution HPLC which in further HPLC resolved into a small group of glycopeptides. Recent Yugoslavian P. aurantiogriseum isolates, from a village in which the idiopathic human disease Balkan Nephropathy is hyperendemic, elicited a similar nephropathology and were acutely cytotoxic, reinforcing a need to regard this novel Penicillium nephrotoxin as a potential factor in human nephropathy.

    Notes: Abstract Three unselected cell lines of C. roseus maintained on a growth-associated alkaloid production medium were studied over a period of 2 to 5.5 years for the stability of alkaloid production (serpentine and ajmalicine). Large fluctuations in the total alkaloid content of 20-day-old cells were found for all three cell lines at each subculture over a two-year period. Growth rates increased during prolonged subculture and one cell line became unproductive after five years culture. By selection of small autofluorescent aggregates, high alkaloid production was restored in this cell line, while the parent line was found to be unresponsive to alkaloid induction treatments. The instability in both alkaloid production and spectrum and the loss of alkaloid productivity are discussed in relation to the selection pressures present during long-term maintenance of cell suspension cultures.

    Notes: Abstract Measurements of 137Cs, 239/240Pu and 241Am contamination in substrate and vegetation on a tide washed pasture in the Mersey Estuary have enabled the dynamic response of vegetation contamination resulting from individual inundation events to be observed. Vegetation contamination is almost entirely due to external contamination by sediment, although there is some evidence for a small contribution from root uptake for 137Cs, with a transfer factor from substrate to internal plant of about 0.003. Marked temporal variation is consistent with contamination by deposition of suspended sediments during tidal inundations, subsequently decreasing with a halflife of 20–30 days. Overall accumulation factors (total plant:substrate) are lower than those observed for the few pastures and saltmarshes studies further north in Cumbria, and consistent with the lower inundation frequency of the Mersey Estuary site.

    Notes: Abstract In this paper stress intensity factors are presented for a bimaterial wedge in which plasticity effects are allowed for. Results are presented for a range of materials and for wedges in which each material occupies a sector of angle equal to one half that of the wedge. The calculations are an extension of previous work in which only a 180° wedge was considered and an entirely different numerical scheme is used that leads to analytic fits to the stress parameters.

    Notes: Abstract The testis is a complex organ in which local control is achieved by signalling between its constituent cells. Herein we describe the responses of cultured rat testicular cells and a mouse Sertoli cell-line to stimulation by endothelin and ATP, and elsewhere we have shown that rat peritubular myoid cells possess phosphoinositidase C-coupled Vla-vasopressin receptors identical to those of liver (Howl, J.et al, 1995, Endocrinology 136: 2206–2213). 1. Peritubular myoid cells from pre-pubertal rats responded through ETA receptors with PtdIns(4,5)P 2 hydrolysis [EC50 for endothelin-1 (ET-1)∼0.4 nM], elevation of intracellular [Ca2+], and tyrosine phosphorylation of a variety of cellular proteins. They also showed enhanced adenylate cyclase activity, with an EC50 for ET-1 of∼3 nM, also through ETA receptors. Pharmacological elevation of [cAMP] did not immediately change the ET-1-stimulated formation of inositol phosphates, but attenuated the response after several hours. 2. Pre-pubertal rat Sertoli cells showed no detectable responses to ET-1, but responded to FSH with elevated [cAMP] and to ATP with PtdIns(4,5)P 2 hydrolysis. PtdIns(4,5)P 2 hydrolysis was equally responsive to ATP and UTP, and so appears to be activated by P2U-purinergic receptors. This response was enhanced by protein kinase C inhibition and attenuated by PKC activation. 3. Despite its lack of effect on rat Sertoli cells in primary culture, ET-1 provoked PtdIns(4,5)P 2 hydrolysis in the TM4 murine Sertoli cell line (EC50∼0.6 nM), and this response was negatively regulated by protein kinase C activation. 5. No receptorstimulated activation of phosphoinositase C was detected in ‘germ cell’ populations, but the non-specific G protein activator AIF 4 − provoked inositol phosphate accumulation in these cells, so demonstrating their potential to respond through yet to be identified G protein-coupled receptors with phosphoinositidase C activation. 6. Immunoblotting studies showed the presence in rat testis of phosphoinositidase C-β1 and the α-subunits(s) of the G-protein(s) Gq and/or Gll. These studies show that testicular myoid and Sertoli cells use at least three G protein-coupled receptors (Vla-vasopressins, ETA-endothelin and P2U-purinergic) to signal through phosphoinositidase C activation, that ET-1 can activate multiple signalling pathways in myoid cells, and that the ET-1-stimulated phosphoinositidase C responses of myoid and Sertoli cells have different regulatory characteristics.
