Notes: Abstract three forms of cell adhesion determine the life cycle ofDictyostelium: i) adhesion of bacteria to the surface of the growing amoebae, as the prerequisite for phagocytosis;ii) cell-substrate adhesion, necessary for both locomotion of the amoebae and migration of the slug;iii) cell-cell adhesion, essential for transition from the unicellular to the multicellular stage. Intercellular adhesion has received the most attention, and fruitful approaches have been developed over the past 25 years to identify, purify and characterize cell adhesion molecules. The csA glycoprotein, in particular, which mediates adhesion during the aggregation stage, is one of the best defined cell adhesion molecules. The molecular components involved in phagocytosis and cell-substratum adhesion are less well understood, but the basis has been laid for a systematic investigation of both topics in the near future.

Notes: Abstract The H1° gene has a long 3′ untranslated region (3′UTR) of 1,125 nucleotides in the rat and 1,310 in humans. Analysis of the sequences shows that they have features of simple DNA that suggest involvement of replication slippage in their evolution. These features include the length imbalance between the rat and human sequences; the abundance of single-base repeats, two-base runs and other simple motifs clustered along the sequence; and the presence of single-base repeat length polymorphisms in the rat and mouse sequences. Pairwise comparisons show numerous short insertions/deletions, often flanked by direct repeats. In addition, a proportion of short insertions/deletions results from length differences in conserved single-base repeats. Quantification of the sequence simplicity shows that simple sequences have been more actively incorporated in the human lineage than in the rodent lineage. The combination of insertions/deletions and nucleotide substitutions along the sequence gives rise to three main regions of homology: a highly variable central region flanked by more conserved regions nearest the coding region and the polyA addition site.

Notes: Summary The complete 3′ untranslated region (3′UTR) sequence of the human skeletal-actin gene has been compared with the corresponding regions of the rat and chicken skeletal-actin genes. This comparison reveals that the skeletal-actin 3′UTR is composed of conserved and nonconserved segments. By using genomic Southern transfer blots and thermal stability (Tm) measurements, we found that the cardiac-actin gene 3′UTR also consists of conserved and nonconserved segments. Comparison of human andXenopus laevis cardiac-actin mRNA sequences confirms the presence of a region of high similarity in the 3′UTR. We conclude that subsegments of the 3′UTRs of both skeletal- and cardiac-actin genes of birds and mammals are under considerable selective pressure. This suggests that these conserved sequences may have functional roles in actin-gene expression or regulation, and that these roles might be different for each actin isoform.

Description / Table of Contents: Abstract Based on distinctive stratigraphic and/or structural characteristics, the Brazilian continental margin can be divided into two main provinces: (1) The southeastern-eastern province, extending from the Pelotas to the Recife-João Pessoa Basin, presents a tensional tectonic style of Late Jurassic-Early Cretaceous age, paralleling the structural alignements of the Precambrian basement, except in the north-eastern segment where the Mesozoic faults of the Recife-João Pessoa Basin cut across the east-west basement directions. The basin-fill, Upper Jurassic through Recent, consists, where complete, of three stratigraphic sequences, each of a distinct depositional environment: (a) a lower clastic non-marine sequence; (b) a middle evaporitic sequence, and (c) an upper clastic paralic and open marine sequence. (2) The northern province, extending from the Potiguar Basin to the Amazon Submarine Basin, displays both tensional and compressional tectonic styles of Upper Jurassic (?) to Upper Cretaceous age either paralleling or cutting transversally the basement alignments. The stratigraphic column differs from the southeastern-eastern province in lacking the Lower Cretaceous evaporitic rocks. The integration of the stratigraphie and structural data allows one to determine in the eastern Brazilian marginal basins the main evolutionary stages of a typical pull-apart continental margin: a continental pre-rift and rift stage, an evaporitic proto-ocean stage, and a normal marine open ocean stage. In the northern province it is possible to infer a continental rift-valley-stage, a transform stage and an open continental-margin stage. The relationship between the rift-valley and the transform stages is yet not clear.

Notes: Abstract Objective: Develop and validate limited-sampling strategy (LSS) models for estimating the area under the plasma concentration versus time curve (AUC) of amlodipine, using data from a bioequivalence study. Methods: Sixteen healthy volunteers received single 5-mg oral doses of amlodipine, as reference or test formulation, at a 14-day interval, in a randomized, crossover protocol. Plasma concentrations of amlodipine (n = 288), measured by mass spectrometry, were used to develop LSS models. Results: Linear regression analysis of the AUC0–72 and a “jack-knife” validation procedure revealed that LSS models based on two sampling times (12 h and 48 h) predict accurately (R2 = 0.99; bias〈0.01%; precision = 0.03%) the AUC0–72 of amlodipine for each formulation. Validation tests indicate that the 2-point LSS model developed for the reference formulation predicts accurately (R2〉0.90): (a) the individual AUC0–72 for the test formulation in the same group of volunteers; (b) the individual AUC0–72 for the same reference formulation in another bioequivalence study in Brazilian volunteers; (c) the average AUC0–72 reported in seven additional international studies performed under protocols similar to the present investigation; (d) the individual AUC0–72 corresponding to concentration data points provided by a first-order compartmental pharmacokinetic model, when the relative values of either the absorption rate (K abs) or the bioavailability (F) model parameters were set at 0.85 or 0.6, of their respective original values. Conclusions: The 2-point LSS models developed in the current study predict accurately the AUC of amlodipine under a variety of experimental conditions and, thus, may be valuable for exploring the relationships between the pharmacokinetics and pharmacodynamics of this calcium antagonist, at reduced costs of sample acquisition and analysis, and avoiding sampling at “unsociable” hours.

Notes: Abstract A marine strain (BS107), identified as a Roseobacter species, was antagonistic to Vibrio species on agar plates. Results suggested that the inhibitory effect was displayed only in the presence of another bacterium. Quantification of the antibacterial activity showed that 48-hour-coculture supernatants from BS107 and another bacterial strain (V. anguillarum 408) reached the highest titers of bacterial inhibition. The antibacterial substance was also liberated when supernatants from V. anguillarum 408 were added to pure cultures of the inhibition-productive bacterium. The presence of a proteinaceous molecule may induce BS107 to display the inhibitory effect. The antibacterial substance was sensitive to trypsin (8000 U/ml) and stable at 100°C. Cell extracts of the isolate BS107 (106 cells/ml) significantly enhanced scallop larval survival, thus being beneficial to the rearing process.

Notes: Abstract The influence of Main Sequence stellar rotation on horizontal branch stars and on RR Lyrae pulsational properties is studied. Synthetic HB's are computed for a wide range of original chemical compositions taking into account evolutionary effects based on Fusi Pecci-Renzini's evolutionary scheme. Theoretical properties of RR Lyrae pulsators are discussed: it is shown that main sequence rotation is able to induce in HB structures sensitive variations, so that some fundamental relations obtained in the classical frame (i.e., no rotation) have to be released. Consequences on the decodification of observational properties in galactic globular clusters are discussed.
