    Notes: Abstract. We have chemically characterized a preparation of halitoxins, (1,3 alkyl-pyridinium salts) isolated from the marine sponge Callyspongia ridleyi. At concentrations of 50 and 5 μg/ml the halitoxin preparation caused irreversible membrane potential depolarization, decreased input resistance and inhibited evoked action potentials when applied to cultured dorsal root ganglion neurones. Under whole cell voltage clamp the halitoxins produced an increase in cation conductance that was attenuated by replacing sodium with N-methyl-d-glucamine. Fura-2 fluorescence ratiometric calcium imaging was used to directly measure calcium flux into neurones after exposure to halitoxins. Calcium influx, evoked by the halitoxins, persisted when the neurones were bathed in medium containing the voltage-activated calcium channel antagonists cadmium and nickel. Experiments on undifferentiated F-11 cells showed little or no calcium influx in response to depolarizing concentrations of potassium and indicated that halitoxins evoked massive calcium influx in the absence of voltage-activated calcium channels. The halitoxins also produced transient increases in intracellular calcium when F-11 cells were bathed in calcium-free medium suggesting that the toxins could release calcium from intracellular stores. The pore-forming action of the halitoxins was identified when the toxins were applied to artificial lipid bilayers composed of phosphatidylcholine and cholesterol. Halitoxins evoked channel-like activity in the lipid bilayers, with estimated unitary conductances of between 145pS and 2280pS, possibly indicating that distinct channels could be produced by the different components in the preparation of halitoxins.

    Notes: Abstract  We have identified the major histocompatibility complex class II β-chain (B-LB) genes present in the B-F/B-L region of the B complex of nine well-characterized lines of chickens and have cleared up much of the confusion concerning numbers and location of B-LB genes in this region. By amplifying DNA sequences between adjacent genes, we found two B-LB genes that lie on either side of Tapasin. The dominantly expressed 'major' B-LB gene in all haplotypes lies between Tapasin and RING-3, and belongs to the B-LBII family of class II β-chain genes. The poorly expressed 'minor' B-LB gene in all haplotypes lies between B-lec1 and Tapasin, and belongs either to the B-LBII family or to the previously unmapped B-LBVI family of class II β-chain genes. The data suggest that the B-LBII and B-LBVI genes are two lineages of B-LB genes and we propose that they all be termed B-LB genes. The location of a third B-LB gene in the B12 haplotype (and possibly other haplotypes as well) has yet to be determined. The structural organization and expression of the class II β-chain genes in the B-F/B-L region is similar to that of chicken class I (B-F) genes, one functional result of which is differential resistance to disease and response to vaccines.

    Notes: Summary The bioavailability of a rapidly dissolving tablet of theophylline and three brands of standard aminophylline tablets was estimated in a four way cross-over study involving 8 healthy adult volunteers. The relative extent of bioavailability as assessed by the measurement of the total area under the plasma concentration time curves showed no difference between the products (P〉0.05). Computed estimates of the rate of drug absorption were similar for all 4 products tested. The results indicate that the rapidly dissolving tablet offers no advantage in respect to rate and extent of absorption over conventional aminophylline tablets.

    Notes: Abstract For Markov models of single channel kinetics, a sojourn time in a class of states has a density function which is usually a linear combination of exponential densities. There are many instances in the single channel literature where the time constants of exponentials fitted to sojourn time data have been used as estimated mean sojourn times in individual states, though the two may be very different. In the present study the nature and magnitude of this difference in the case of a two state class is illustrated analytically and numerically. The time constants should be viewed at best as approximations, possibly poor, to the estimated mean sojourn times. Estimates of kinetic parameters cannot in general be obtained explicitly from the fitted parameters of the density alone. However, this is shown to be possible in some special cases and enables direct estimation of, for example, the channel opening rate constant β (or an upper limit to the estimate of β in the case of multiple channels) in standard sequential three or four state models of nicotinic receptor kinetics, using only the fitted parameters of the closed-time density.

    Notes: Abstract A structural study of glass-ionomer cement (GIC) dental restoratives has been completed. Transmission electron microscopy, selected area electron diffraction, and X-ray diffraction studies indicate domain-like microstructure in a new experimental material, whereas a featureless amorphous gel-like microstructure exists in the conventional GIC. Nuclear magnetic resonance studies were also conducted. The new experimental GIC contains domains of (i) bonelike material (apatite), (ii) mesoporous material and (iii) other framework structures (aluminium phosphate in the high cristobalite structure), with its setting chemistry a restructuring of the aluminosilicate glass around the template of poly(acrylic acid). Conventional glass-ionomer cement may set by a similar but slower process. Leaching properties of glass-ionomer cements are also explained.

    Notes: Abstract A lead zirconate titanate (PZT) precursor sol was prepared by a diol sol-gel route and used as a binder for assisting the uniaxial die pressing of PZT powders. The powders, of composition Pb(Zr0.52Ti0.48) O3, were prepared by a two step mixed oxide route using lead carbonate, zirconia and titania starting powders. The densification characteristics of the powders, which were not milled after the final calcination step, were compared to samples prepared using PVA as a conventional binder. Improved sintering behaviour was observed for the novel sol-gel binder system. For example, using a sol concentration, equivalent to 7 wt% of PZT derived from the binder, pellets of 95% theoretical density were obtained after firing at 1150°C for 4 h, whereas for the same firing conditions, samples blended with PVA resulted in a density of only 78% theoretical.

    Notes: Abstract Simple analytic models for the passive evolution of arcade-like magnetic fields through a series of force-free equilibria are presented. At the photospheric boundary, the normal magnetic field component is prescribed together with either the longitudinal field component or the photospheric shear. Analytic progress is made by considering either cylindrically symmetric solutions or using the separation of variables technique. Two distinct cylindrically symmetric force-free fields are obtained that possess the same normal field component and photospheric shear. The scond field contains a magnetic bubble. As the shear increases beyond a critical value, so the magnetic energy of the first configuration exceeds that of the second. The possibility is therefore suggested of an eruption of the first field outwards towards the second. Such an eruptive instability is proposed as the origin of a two-ribbon solar flare. A new analytic solution to the force-free field equations, of separable form, is discovered and it is pointed out that the existence of shear in a magnetic field does not preclude it from being potential.

    Notes: Abstract A conception of probability as an irreducible feature of the physical world is outlined. Propensity analyses of probability are examined and rejected as both formally and conceptually inadequate. It is argued that probability is a non-dispositional property of trial-types; probabilities are attributed to outcomes as event-types. Brier's Rule in an objectivist guise is used to forge a connection between physical and subjective probabilities. In the light of this connection there are grounds for supposing physical probability to obey some standard set of axioms. However, there is no a priori reason why this should be the case.
