    Notes: Abstract Sequential imaging of the temporal changes inP-wave velocity offers a practical tool to monitor a rock mass. Using established correlations between the location of seismic events and velocity structure, the temporal seismic potential characteristics of the rock may be monitored. Furthermore, the temporal velocity differences isolate the time dependent factors effecting velocity such as stress, while cancelling static factors such as lithology. Various sequential imaging techniques were compared with respect to accuracy. Differences between successive velocity images were found to have relatively high associated error estimates. However, images of velocity differences calculated from measured travel time delays between successive velocity surveys were found to have lower error estimates. In particular, travel time delays measured using cross-correlation techniques resulted in the most accurate sequential image. Two 3D sequential imaging studies were carried out at Strathcona Mine in Sudbury, Ontario, Canada. Results of the average static images indicated an association between the location of induced microseismicity and a zone of both high velocity and high gradient. Additional examples are described from the global seismology literature which also show a similar correlation between seismicity and velocity structure. We attribute this association to an interrelated stress and strength effect. The Strathcona Mine sequential images show zones of significantly decreased velocity in regions of concentrated microseismic activity, which are postulated to be indications of localized destressing and relaxation of the clamping forces resulting in the microseismicity. The zones of decreased velocity corresponded to an increase in the velocity gradient. One of the case studies also shows an increase in velocity in a zone of high static velocity, which is later the site of am N 2.5 mining-induced seismic tremor. The increase in velocity is believed to correspond to a region of stress concentration, resulting in the subsequent seismic tremor.

    Notes: Abstract The study of microseismicity in mines provides an ideal method for remote volumetric sampling of rock masses. The nature and uniqueness of microseismic monitoring is outlined in the context of acquisition hardware and software requirements. Several topics are used to highlight the potential for novel applications of microseismicity and to outline areas where further study is required. These topics reflect some of the current interest areas in seismology, namelyb values and source parameters, fault-plane solutions, modes of failure and moment tensor inversion, imaging and seismicityvelocity correlations. These studies suggest potential correlations between zones of high seismic velocity, high microseismic activity and maximal stress drops, which can be interpreted spatially to be the locations of highly stressed ground with a potential for rock bursting. Fault-plane solutions are shown to be useful in determining the slip potential of various joint sets in a rock mass. Source parameter studies and moment tensor analysis clearly show the importance of non-shear components of failure, andb values for microseismicity appear to be magnitude-limited and related to spatial rather than temporal variations in effective stress levels.

    Notes: Abstract Mutation analysis of the superoxide dismutase gene SOD1 in a familial case of amyotrophic lateral sclerosis revealed a T→C transition at codon 151 of exon 5. This mutation results in the substitution of an isoleucine for a threonine. It appears to affect formation of dimers of the protein and is the most C-terminal amino acid change in SOD1 described to date.

    Notes: Abstract Transfection of the wild-type p53 gene into an immortalized human endothelial cell line (ECV-304) by recombinant adenoviral delivery resulted in high level expression of the wild-type p53 protein and induction of apoptosis. Increases in the number of apoptotic cells were observed within 12 h after infection of ECV-304 cells with recombinant p53 adenovirus, as deter-mined by the appearance of internucleosomal DNA fragmentation ladders and by TUNEL and electron microscopic analyses. Control cells infected with a β-galactosidase recombinant adenovirus exhibited little or no increase in apoptosis over uninfected cells. The expression of Waf-1 and Bax gene products were in-creased substantially in apoptotic ECV-304 cells as determined by Northern blot, reverse transcription-PCR and immunoblotting analyses. Lesser increases in the expression of the PCNA gene were detected in ECV-304 cells undergoing apoptosis. Both control and apoptotic ECV-304 cells did not express detectable levels of Bcl-2 mRNA or protein in Northern blotting and immunoblotting analyses, respectively. The data suggest a role for the Bax gene product in p53-mediated apoptosis of endothelial cells.

    Notes: Abstract The natural metabolic byproduct of estradiol, 2-methoxyestradiol (2-MeOE2), induces apoptosis in human lung cancer cells by a p53-dependent mechanism. The expression of wild-type p53 isoforms was investigated in H1299 non-small cell lung carcinoma cells induced into apoptosis by 2-MeOE2. H1299 cells lack endogenous p53 and undergo predominantly a G1 arrest when infected with a recombinant wild-type p53 adenovirus. However, when H1299 cells transfected with p53 were treated with 2-MeOE2, they underwent rapid and extensive apoptosis. H1299 cells expressing mutant his273 p53 were unaffected by 2-MeOE2, indicating a dependence of 2-MeOE2-mediated apoptosis on the presence of a functional p53. Analysis of wild-type p53 phosphoisoforms in H1299 cells by two-dimensional gel electrophoresis revealed that 2-MeOE2 induced a unique group of acidic p53 isoforms. Although most of the wild-type p53 in untreated H1299 cells migrated as at least five diffuse species with isoelectric points from pH 5.5–6.3, as many as nine additional forms migrating toward the acidic region with pI values from 4.4–5.3 were detected in 2-MeOE2-treated apoptotic cells. Two other agents known to induce apoptosis, vinblastine and actinomycin D, induced a similar pattern of acidic p53 species as that observed for 2-MeOE2. The results indicated that the induction of apoptosis in H1299 cells by 2-MeOE2 is dependent on the upregulation of specific p53 isoforms. Identification of the specific p53 phosphoisoforms induced by MeOE2 will be an important step in understanding the regulation and function of p53 in apoptosis.

    Notes: Abstract Overexpression of the normal p53 protein in tumor cell lines is known to induce apoptosis and a potential mediator of this response is the apoptotic inducer, Bax. The expression of Bax mRNA products were investigated in the ECV-304 endothelial cell tumor line and primary human umbilical vein endothelial cells (HUVEC) that were induced to overexpress the p53 protein. Induction of p53 in ECV-304 and HUVEC cells was mediated by infection with a p53 recombinant adenovirus (AdCMV-p53). The expression of Bax transcripts in p53-induced cells was investigated by reverse-transcription polymerase chain reaction (RT-PCR). The Baxα mRNA species was detected in both ECV-304 and HUVEC cells. Surprisingly, Baxα expression was reduced several-fold in ECV-304 endothelial cells overproducing p53 and no change in Baxα was detected in HUVEC cells after induction of p53. However, the Baxδ spliced transcript was observed to be induced by p53 in the ECV-304 tumor cell line. Baxα was the predominant species expressed in normal human endothelial cells but, in contrast to the immortalized ECV-304 endothelial cell line, induction of p53 failed to alter the expression of Baxα or to induce any other Bax transcripts. HUVEC cells were more resistant to p53, since at least 80% of the HUVEC cell population survived the overexpression of p53 after 24 h of infection with AdCMV-p53. An ECV-304-derived cell line (DECV) resistant to p53-mediated apoptosis did not show any changes in expression of Bax mRNA products, even in the presence of high levels of p53. ECV-304 endothelial cells that expressed the Baxδ species underwent apoptosis much more rapidly and more extensively after induction of p53, suggesting that the Baxδ species enhances p53-mediated apoptosis.

    Notes: Abstract A group of five children with kwashiorkor, seven with marasmic kwashiorkor and one underweight child were given an aflatoxin-free diet consisting of maize meal and milk powder. Blood specimens were collected on admission; on day 4 and 10, 24 hour urine and stool samples were collected for the first ten days. Serum, urine and stool samples were analysed for aflatoxins using high performance liquid chromatography with fluorescent detection, after various extraction and clean-up procedures. The children with kwashiorkor and marasmic kwashiorkor excreted aflatoxins in stools for up to 9 and 6 days after admission respectively. No aflatoxins were detected in the stools or urine of the underweight child. In kwashiorkor, urinary excretion ceased after 2 days, while in marasmic kwashiorkor urinary excretion persisted for 4 days. In stools, B1 was the type of aflatoxin detected most frequently in kwashiorkor and least frequently in marasmic kwashiorkor. Aflatoxin M2 was frequently detected in the stools of both groups of children. Estimates of the total amount of aflatoxin excreted by kwashiorkor and marasmic kwashiorkor indicate that these children were harbouring up to 4 μg/kg body weight at the time of admission. These findings establish that aflatoxins accumulate in body fluids and tissues in kwashiorkor and marasmic kwashiorkor which is only slowly eliminated.

    Notes: Abstract Autopsy brain (cerebrum) specimens from 18 kwashiorkor children and 19 children who had died from a variety of other diseases, at the Obafemi Awolowo Teaching Hospital complex, Ile-Ife, Nigeria, were analysed for the presence of aflatoxins using high-performance liquid chromatography. Aflatoxins were detected in 81%, 15 specimens in each group. More than one type of aflatoxin was detected in 14 (37.8%) of all the specimens. Aflatoxin B1 and its reversible metabolite, aflatoxicol, were detected in 11 brain specimens of patients with kwashiorkor and 6 of those who died of other miscellaneous diseases; out of these 6, two died from measles and its complications. The frequent detection of aflatoxins in the brains of these children and sometimes in multiple forms may suggest that aflatoxins are stored in the brain tissue which could be related to the lipophilic nature of these compounds. These findings also suggest that although many children in the tropics are exposed to aflatoxins, the accumulation of aflatoxin B1 and aflatoxicol in the brains of kwashiorkor children may be a result of an impaired metabolism of these compounds by these children.

    Notes: Abstract Upregulation of the p53 tumor suppressor protein by infection with a recombinant p53 adenovirus resulted in extensive apoptosis in ECV-304 cells and the eventual death of almost all the cells. To establish a system to elucidate the molecular mechanisms involved in p53-mediated apoptosis of these cells, we established a variant of ECV-304 that is resistant to p53-induced apoptosis by repeated infections with a recombinant p53 adenovirus. We have designated this variant as the DECV cell line (Differentiated ECV-304). DECV cells expressed similar amounts of nuclear-localized p53 as ECV-304 cells when infected with recombinant p53 adenovirus, but in contrast to ECV-304 cells, greater than 95% of DECV cells survived and remained viable after 24 hours of infection. In further contrast to ECV-304 cells, DECV cells grew less efficiently in soft agar and exhibited contact inhibition in growth assays. Moreover, DECV cells formed unusual lattice or cyst-like structures in culture and formed lumenal structures indicative of epithelial differentiation in three-dimensional collagen matrices, while parental ECV-304 cells showed minimal evidence of these cellular behaviors. A comparative molecular analysis of gene expression in DECV and ECV-304 cells was conducted by cDNA micro-array technology. Protocadherin-1 was found to be expressed in DECV cells but not in ECV-304 cells, while the Id-3 gene was observed expressed in ECV-304 cells but not in DECV cells. Moreover, upregulated expression of p53 in ECV-304 cells induced the EPHB2 (Ephrin) receptor tyrosine kinase and the ephrin-B1 ligand mRNAs compared to DECV cells treated in the same manner. These data demonstrate that a new variant of the ECV-304 cell line, which is resistant to p53-mediated apoptosis, exhibits differential gene expression as well as distinct cell behaviors as compared to the parental ECV-304 cell line. DECV cells should prove to be a useful tool in future studies to elucidate mechanisms of p53-mediated apoptosis and differentiation.
