    Notes: Abstract Core samples were taken at two sites from a peat deposit buried by a sanitary landfill operated by the city of Vancouver since the 1960s and from a third site where the same peat bed is not covered by landfill. Twenty-nine subsamples from the three cores were analyzed by a variety of techniques to determine the concentration of as many as 34 constituents. The content of heavy metals, the principal object of this investigation, is highest in the lower part of the peat succession, in which there is a significant amount of interbedded inorganic sediment, rather than in the upper clean bog peat. Individual layers as little as 2.5 cm thick can hold concentrations of heavy metals ten times that of the nearby layers. The heavy metal contents show a high positive correlation with those of iron and manganese and a very low correlation with sulfur. Iron from the landfill has been transported by downward percolating groundwater in solution or colloidal suspension into the lower layers of peat deposit where the passing heavy metals were sorbed. A comparison of the amounts of heavy metals stored in the peat alone with the amount leaving the whole landfill annually suggests that some metals, notably lead and arsenic, might be retained in the peat for very long periods, whereas other metals such as zinc and mercury might be quickly lost.

The gating cycle of CFTR (Cystic Fibrosis Transmembrane conductance Regulator) chloride channels requires ATP hydrolysis and can be interrupted by exposure to the nonhydrolyzable nucleotide AMP-PNP. To further characterize nucleotide interactions and channel gating, we have studied the effects of AMP-PNP, protein kinase C (PKC) phosphorylation, and temperature on gating kinetics. The rate of channel locking increased from 1.05 × 10−3 sec−1 to 58.7 × 10−3 sec−1 when AMP-PNP concentration was raised from 0.5 to 5 mm in the presence of 1 mm MgATP and 180 nm protein kinase A catalytic subunit (PKA). Although rapid locking precluded estimation of P o or opening rate immediately after the addition of AMP-PNP to wild-type channels, analysis of locking rates in the presence of high AMP-PNP concentrations revealed two components. The appearance of a distinct, slow component at high [AMP-PNP] is evidence for AMP-PNP interactions at a second site, where competition with ATP would reduce P o and thereby delay locking. All channels exhibited locking when they were strongly phosphorylated by PKA, but not when exposed to PKC alone. AMP-PNP increased P o at temperatures above 30°C but did not cause locking, evidence that the stabilizing interactions between domains, which have been proposed to maintain CFTR in the open burst state, are relatively weak. The temperature dependence of normal CFTR gating by ATP was strongly asymmetric, with the opening rate being much more temperature sensitive (Q 10= 9.6) than the closing rate (Q 10= 3.6). These results are consistent with a cyclic model for gating of phosphorylated CFTR.

    Notes: Abstract A human heavy chain variable region subgroup III pseudogene (HV3.3) was isolated, characterized, and sequenced. When HV3. was hybridized to Southern blots of human DNA, two potentially informative polymorphic bands, resulting from 2.7 kb Hind III (HH2.7) and 7.3 kb Eco RI(HE7.3) fragments, were detected with frequencies of 0.553 and 0.606, respectively. These polymorphic bands showed Mendelian segregation in families and appeared to be in tight linkage disequilibrium with each other (χ 1 2 =24.91, P〈0.001). Evidence from sibling-pair data indicated linkage of the Hind III polymorphic band to constant region allotypic and restriction fragment length polymorphism markers. Bands representing alternative forms of the polymorphic restriction sites were not detected for either HH2.7 or HE7.3. This indicates either that the alternative fragments comigrate with homologous fragments resulting from conserved restriction sites, or that the polymorphism is due to a gene duplication or deletion. No band segregating with HH2.7 was found in separate digests using eight other enzymes. Although this indicates that a major deletion is unlikely, it does not exclude the possibility of a gene deletion or duplication affecting the intergenic region(s) of one or more homologous genes. Whatever the precise explanation, these findings support the hypothesis that there is polymorphic variation of V H gene repertoires in man.

    Notes: Summary A physical map of the COB region in mtDNA of yeast has been established. This region harbours a split gene coding for apocytochrome b. It includes restriction sites of seven endonucleases (EcoRI, HindII, HindIII, HaeIII, HpaII, AluI and BamHI). Various mtDNA sequences of this region, retained in a series of genetically characterized rho− clones have been allocated to this map. The combination of this physical map with a genetic map of the rho− clones revealed that 1) cob− mutational sites spread over 8,400 bp, 2) mutations in sequences coding for apocytochrome b map in five distinct segments which are separated by intervening sequences with minimum lengths from 350 to 1,900 bp.

    Notes: Abstract Agricultural management in Cades Cove, an historic district in Great Smoky Mountains National Park, has affected natural resources both within the district and in the adjoining natural areas. Aquatic impacts of haying and cattle grazing included increases in water temperatures, turbidity, nutrient loading, and bacterial counts and decreases in benthic macroinvertebrate density and fish biomass. Wildlife populations, including groundhogs, wild turkeys, and white-tailed deer, have increased in the open fields and around the periphery of the historic district. Intensive deer foraging has removed deciduous seedlings and saplings from woodlots, lowering species diversity and favoring coniferous reproduction. Cades Cove has limestone habitats unique in the park, and both deer browse and cattle grazing may have disturbed populations of rare plant species. Effects on water quality are detectable at a campground 15 stream km from the agricultural area, and the effects of deer foraging extend about 1 km beyond the open fields. Since “historic landscape” preservation is presently a goal of the park, managing for open vistas in Cades Cove will require some sort of continuing disturbance. Conversion of cattle pastures to hayfields would reduce aquatic impacts but the deer herd might increase as a result of reduced competition for forage. Retarding old field succession would increase populations of native plant species dependent on sunlight, but would require government-funded mowing. Other options are discussed. Completely eliminating the effects of the historic district on adjoining areas may be impossible, at least under present economic constraints.

    Notes: 2 Copper Basin in southeastern Tennessee became the site of increasingly extensive and successful reforestation efforts. To determine the effectiveness of more than 50 years of reforestation efforts, we compared rainfall infiltration, sediment detachment, and soil organic matter of reforested sites to those properties of unvegetated sites and forested reference sites outside the basin. Results of 54 rainfall simulation experiments conducted in zones of the basin known to have been planted during different decades demonstrate that hydrologic recovery of soils in the Copper Basin lags significantly behind the establishment of tree cover and the protection offered by vegetation against soil erosion. Soils in new “forests” have significantly less organic matter and lower infiltration than forests more than 50 years old. The long-term persistence of low infiltration rates serves as a reminder that, at sites where the A and B soil horizons have been lost, restoration of the hydrologic function of a landscape requires decades, at least.
