    Notes: Abstract Exposing cells to adverse conditions usually elicits expression of stress-response (heat shock) proteins (srp). Here we show that hyperosmolar growth conditions do not uniformly affect srp expression in MCF-7 and HeLa S3 cells, derived from carcinoma of the breast and cervix, respectively. Thus, whereas srp 27 expression was increased in MCF-7, but not in HeLa S3, the opposite was the case with srp 72. On the other hand, hyperosmolality did not induce αB-crystallin or ubiquitin in either cell line. These findings show that srp expression by the human tumor cells studied is non-coordinate, suggesting that each srp is independently modulated.

    Notes: Abstract Segregation analysis of polymorphic sites within the retinoblastoma (RB) gene and on chromosome 13, as well as the parental origin of the lost allele in the tumor, were analyzed in 24 families with RB patients. Four mutant alleles transmitted through the germ-line and seven de novo germ-line mutant alleles were identified in 11 patients with hereditary RB. Segregation analysis within the RB gene and on chromosome 13 was useful for DNA diagnosis of susceptibility to RB in relatives of hereditary patients, even if mutations were not identified. All seven de novo germ-line mutant alleles were paternally derived. The bias toward the paternal allele for de novo germ-line mutations of the RB gene was statistically significant. Seven paternal alleles and six maternal alleles were lost in 13 non-hereditary RB tumors with no bias in the parental origin of the somatic allele loss. These results suggest that the physical environment or a deficiency in DNA repair during spermatogenesis may be associated with significant risk factors for de novo germ-line mutations.

    Notes: Abstract Objective: Patients with heart failure have abnormal neurohormonal regulation during orthostatic stress, and abnormal arterial baroreflex function. This study investigated the effects of alacepril, a new angiotensin-converting enzyme inhibitor with sulfhydryls, on changes in neurohormonal factors during tilt and on the arterial baroreflex control of heart rate. Methods: Plasma concentrations of noradrenaline, adrenaline, renin activity, angiotensin II, and atrial natriuretic peptide were measured at supine rest and after 30° head-up tilt with measurements of central venous pressure and cardiac dimensions in seven patients with congestive heart failure (65 years, ejection fraction = 34%). Arterial baroreflex control of heart rate was assessed by phenylephrine bolus. The arterial baroreflex test was re-examined 3 h after oral alacepril (37.5 mg). The tilt and arterial baroreflex tests were repeated 12 weeks after alacepril treatment (50 mg␣·␣day−1). Results: Heart rate, blood pressure, and neurohormonal factors did not differ before and after chronic alacepril, except for a trend toward an increase in renin activity (2.0 vs 4.9 ng · ml−1· h−1). Head-up tilt decreased central venous pressure (−2.5 mmHg) with a decrease in cardiac dimensions in the pre-alacepril phase. These changes were accompanied by increases in noradrenaline, adrenaline, and angiotensin II and a decrease in atrial natriuretic peptide. After chronic alacepril, the increase in noradrenaline during head-up tilt tended to be smaller (84 vs 30 pg · ml−1), with similar changes in central venous pressure (−3.4 mmHg) and cardiac dimensions. Both acute (3.6 vs 4.8 ms · mmHg−1) and chronic (3.6 vs 6.7 ms · mmHg−1) alacepril treatment was associated with a trend towards an increase in the arterial baroreflex control of heart rate. Conclusion: These results suggest that treatment with alacepril may cause a reduction of sympathetic activation during orthostatic stress and may enhance arterial baroreflex function in patients with mild to moderate heart failure.

    Notes: Abstract To elucidate the effect of Nicorandil on myocardial energy metabolism and myocardial sympathetic activity, we administered Nicorandil orally to eight patients with angina pectoris prior to exercise testing. Arterial and coronary sinus levels of lactate, ammonia, hypoxanthine (HX), adrenaline and noradrenaline were measured during exercise in order to determine the irrespective myocardial extraction ratios (MER). Compared to placebo, Nicorandil increased the time to development of significant ST depression (322 vs 390 s) while decreasing the maximum amplitude of ST depression (0.244 vs 0.216 mV). Heart rate, systolic blood pressure, and the rate pressure product during exercise were not significantly affected. The MER of lactate, measured during exercise, was significantly higher after Nicorandil than placebo (13.6 vs 27.9). Similarly, the MERs of ammonia and HX were significantly higher after Nicorandil (-46.0 vs 7.4% and −47.0 vs 9.9% respectively). Nicorandil, had no apparent effect on myocardial sympathetic activity as the MERs of adrenaline and noradrenaline were essentially unaffected. We conclude that Nicorandil decreased myocardial ischaemia and suppressed myocardial accelerated purine metabolism (a marker of cellular energy metabolism) during exercise in patients with angina pectoris. This effect appears not to be related to myocardial sympathetic activity.

    Notes: Abstract The Bonin (Ogasawara) Islands are oceanic islands located in the northwest Pacific, and have ten native (nine endemic) bee species, all of which are nonsocial. The European honeybee (Apis mellifera), which was introduced to the islands for apiculture in the 1880s, became naturalized in a few islands shortly after introduction. To detect the impact of the honeybees upon native bee diversity, we analyzed pollen harvest by honeybees and surveyed the relative abundance of honeybees and native bees on flowers on several islands. Both hived and feral honeybee colonies were active throughout the year, harvesting pollen of both native and alien flowers and from both entomophilous and anemophilous flowers. Honeybees strongly depended on the alien plants, especially during winter to spring when native melittophilous flowers were rare. From June to November, honeybees exhaustively utilized native flowers, which had originally been utilized and pollinated by native bees. On Chichi and Haha Islands, where human disturbance of forests has been severe, both native and alien flowers were dominated by honeybees, and native bees were rare or extinct even in well-conserved forests. In contrast, on Ani Island and Haha's satellite islands where primary forests were well conserved and honeybees were still uncommon or absent, native bees remained dominant. These results suggest that competition for nectar and pollen of the native flowers between honeybees and native bees favors honeybees on the disturbed islands, which are thoroughly invaded by alien nectariferous, sometimes aggressive, weedy plants.

    Notes: Abstract The possibility is considered that there exists a universal V-A interaction and complete symmetry in the nonleptonic decay of hyperons. The departure from the expected symmetry that is observed in actual decays Is explained by effects caused by the mesonic structure of hyperons. In close connection with the idea of complete symmetry, a new isotopic spin is defined, which is equal to 1/2 for all baryons and to 0 or 1 for K mesons. The law of conservation of this isotopic spin holds in nonleptonic decays.

    Notes: Abstract The population fluctuation pattern of light-attracted beetles was studied from August 1992 to September 1998 (for 73 months) using ultraviolet light-traps set at three vertical levels in a tropical lowland dipterocarp forest in Sarawak, Malaysia. During our study, a general flowering occurred from April to July in 1996, and flowering on a small scale in 1997 and 1998. We analyzed the data for eight scarabaeid and six meloid species, some of which were anthophilous species. Various fluctuation patterns were observed among the beetle species in aspects of both seasonality and correlation with the supraannual phenological pattern. Three large chafer species (Scarabaeidae, Melolonthini) showed a clear seasonal fluctuation pattern with a peak once from March to May every year, the peak monthly catch greatly fluctuating annually. Other scarabaeid beetles did not show such a clear seasonal population pattern and hardly fluctuated annually. Populations of an anthophilous scarabaeid species, Parastasia bimaculata, a specific pollinator of Homalomena propinqua (Araceae), hardly fluctuated, probably because of its response to the constant flowering of its floral hosts. Monthly catches of an anthophilous scarabaeid, Anomala sp., and meloid beetles showed clear supraannual patterns in response to the general flowering and were significantly correlated with the flowering intensity with or without a lag of a month. The fluctuation pattern of meloids suggests a supraannual population fluctuation pattern of their hosts, i.e., megachilid/anthophorid bees.

    Notes: Abstract Using AnCP (Aspergillus nidulans CCAAT-binding protein) as a CCAAT-specific binding factor model, the possibility that one factor is able to recognize CCAAT sequences in several different genes in A.␣nidulans was examined. DNase I protection analysis showed that AnCP specifically bound to CCAAT sequence-containing regions comprising 21 to 36 bp of the taa, amdS and gatA genes. Furthermore, replacement of the CCAAT sequence with CGTAA was found to abolish the binding of AnCP and to have an inhibitory effect on taa promoter activity. This clearly demonstrates a positive function of the CCAAT element. However, amylase was induced by starch and repressed by glucose in a CCAAT-box disruptant, as in wild-type cells.
