    Notes: Abstract The microprojectile bombardment method was used to transfer DNA into embryogenic callus of asparagus (Asparagus officcinalis L.) and to produce stably transformed asparagus plants. Embryogenic callus, derived from UC 157 and UC72 asparagus cultivars, was bombarded with tungsten particles coated with plasmid DNA that contained genes encoding hygromycin phosphotransferase, phosphinothricin acetyl transferase and β-glucuronidase. Putatively transformed calli were identified from the bombarded tissue after 4 months selection on 25 mg/L hygromycin B plus 4 mg/L phosphinothricin (PPT). By selecting embryogenic callus on hygromycin plus PPT the overall transformation and selection efficiencies were substantially improved over selection with hygromycin or PPT alone, where no transgenic clones were recovered. The transgenic nature of the selected material was demonstrated by GUS histochemical assays and Southern blot hybridization analysis. Transgenic asparagus plants were found to withstand the prescribed levels of the PPT-based herbicide BASTATM for weed control.

    Notes: Abstract Leaf-cutting ants of the generaAcromyrmex andAtta are considered the principal polyphagous pests of the Neotropics Although some members of these genera are of economic importance, have a broad geographic distribution, and are extremely good colonizers, others are endemic and closely interact with native ecosystems. Control is generally practiced against any colony, irrespective of its taxonomic status. Indiscriminate control coupled with habitat destruction threatens endemic species with extinction, and, through habitat simplification, favors other pest species. As nests ofAtta are large, having several square meters of nest surface, the endemic taxa can be easily used as environmental indicators for natural ecosystems Likewise, the pest species can be used to detect environmental disturbance As these ants are keystone species and easily identified by nonspecialists, efforts should be made to integrate these into viable conservation programs

    Notes: Abstract A method for the simultaneous determination of yohimbine and boldine in mixtures by first-derivative synchronous spectrofluorimetry has been developed. The method is based on their native fluorescence in 0.1N sulphuric acid medium. The constant wavelength difference chosen to optimize the determination was Δλ=λem -λem=82 nm. Yohimbine was measured at λex//λem= 285/367 nm, and boldine at λex/λem=272/354 nm. The range of application is 10–500 μg/l for yohimbine and 1–50 μg/l for boldine. The method was applied to the determination of yohimbine and boldine in synthetic mixtures and pharmaceuticals, with errors generally ⩽2%. Relative standard deviations were about 2%.

    Notes: Abstract Molecular dynamics (MD) simulations ofγ-Al2O3, using a pairwise additive interaction potential of Pauling's type with four different radii for aluminum atoms, were analysed in order to determine the influence of the radius on the modification of coordination numbers of A1 relative to the ideal structure, at two different temperatures of 300 and 1500 K. It is found that the best choice is the radius of penta-coordinated aluminum, reproducing structural and vibrational properties of the compound in excellent agreement with experimentally observed properties.

    Notes: Summary 1. The neurotransmitter mechanisms regulating neuroendocrine processes have been traditionally inferred from the effects of drugs purportedly acting through specific transmitter systems. The direct appraisal of changes in endogenous neuromediators had to rely initially on analyses of brain samples obtained post-morten. 2. Currently, a more physiological assessment is available through the monitoring ot the extracellular levels of neurotransmitters and their metabolites in discrete brain areas of living animals. Two methodologies, namely in vivo voltammetry and microdialysis, are being increasingly used for this purpose. This article summarizes their principles, relative merits, and limitations and presents some relevant applications. 3. Thus, microdialysis data show a differential response in the amphetamine-induced dopamine release in the nucleus accumbens in adult male and female rats castrated prepuberally. Given their high time-resolution, in vivo electrochemistry techniques seem especially suited for studying the fast, non-genomic effects of steroid hormones. This is illustrated by the voltammetric detection of a rapid release of dopamine in the corpus striatum induced by progesterone in males. 4. These methodologies should be regarded as complementary tools for the assessment of the neurochemical correlates of neuroendocrine interactions.

    Notes: Abstract Activity coefficient values for NaCl in ethanol-water mixtures containing 20, 40, 60, 70, 80 and 90 weight % ethanol are calculated from the emf for the galvanic cell $$Na - glass|NaCl(m), H_2 O (100 - Y), EtOH(Y), AgCl|Ag$$ in which the weight % of the corresponding solvent is indicated in parenthesis. The results obtained are analyzed by using different theoretical models. Chemical models which take into account the presence of ion pairs in solutions of high ethanol content are also used. There is good agreement between the results obtained from all the models.

At each total ionic strength studied, the measurements were carried out at different ratios of the Na-Formate to NaCl ionic strengths. The experimental activity coefficients were fitted using the Harned equation and the treatments of Scatchard et al. and Pitzer et al. Finally, the excess Gibbs energy of these mixtures was also calculated.

    Notes: Abstract The coiled body is a phylogenetically conserved nuclear organelle whose function is not known. Probes for detection of p80-coilin, an 80 kDa protein enriched in the coiled body, have made possible studies determining the behavior of the coiled body during the cell cycle, in proliferating cells, as well as reports suggesting some relationship of the coiled body to mRNA splicing and to the nucleolus. The objective of this study is to examine the distribution of p80-coilin and nucleolar proteins in cells infected with adenovirus in vitro. HeLa cells grown as monolayers were infected with successive dilutions of type 5 human adenovirus culture and fixed in methanol/acetone at different time points. Single and double indirect immunofluorescence was performed with human autoantibodies to p80-coilin, fibrillarin, NOR-90/hUBF, RNA polymerase I, PM-Scl, and To, as well as rabbit polyclonal serum to p80-coilin (R288) and mouse monoclonal antibody to adenovirus 72-kDa DNA-binding protein. Indirect immunofluorescence (IIF) with anti- p80-coilin antibodies showed that the usual bright dot-like coiled body staining pattern was replaced in infected cells by 1–5 clusters of tiny dots at the periphery of the nucleus. This phenomenon was first detected within 12 h of infection and affected more severely cells with increased length and load of infection. Cells subjected to heat shock presented no such alteration. Double IIF showed that cells with abnormal coiled body appearance expressed the viral 72-kDa DNA-binding protein. Nucleolar proteins RNA polymerase I and NOR-90/hUBF became associated with the p80-coilin-enriched clusters and were no longer detected in the nucleolus. Other nucleolar proteins, like PM-Scl and To, remained associated to the nucleolus and were not detected in the newly formed clusters. Fibrillarin had a heterogeneous behavior, being restricted to the nucleolus in some infected cells while in some others it was associated with the p80-coilin-enriched clusters. Thus our results showed that in vitro adenovirus infection induced radical redistribution of nucleolar and coiled body constituents into newly formed structures characterized by clusters of tiny dots in the periphery of the nucleus. The fact that three major proteins involved in rRNA synthesis and processing colocalized with p80-coilin in these clusters may bring additional support to the idea that the coiled body and p80-coilin may be implicated in functions related to the nucleolus.
