    Notes: Abstract Manila clams, Ruditapes philippinarum, removed from their natural environment and maintained for 9 weeks in continuously immersed conditions exhibited a clear endogenous circatidal rhythm in oxygen consumption. The clams exhibited a semidiurnal rhythmicity in oxygen consumption after showing a diurnal pattern in the first few days (5 to 7 d) of the experiment. The results of the present study indicate that activity rhythms of clams are controlled not only by exogenous factors, but also by an endogenous circatidal periodicity.

    Notes: Abstract A 7050 (Al–6.2 wt% Zn–2.3 wt% Cu–2.3 wt% Mg) Al alloy, conventionally used for wrought products, has been successfully cast to near-net shape using direct squeeze casting. Squeeze casting with an applied pressure of 50 MPa removes the defects observed in gravity die cast billets, in particular, (1) shrinkage pipe, (2) poor die replication and waisting, and (3) microporosity. Squeeze casting results in considerable refinement of the microstructure due to an increase in cooling rate from ∼0.5°C s−1 for gravity casting to ∼11°C s−1 for squeeze casting in a tool steel die lined with porous insulation, and from ∼2.5 to ∼10°C s−1, respectively, in an uninsulated die. A normal segregation pattern of increasing eutectic toward the center of the billet is found for squeeze casting, compared to an inverse segregation pattern of increasing eutectic toward the edge of the billet for gravity casting. This change in segregation pattern is due to a higher radial temperature gradient and reduced time in the semisolid state for squeeze casting.

    Notes: Abstract The effects of initial sol parameters on the pore structure and optical transparency of TiO2–SiO2 binary aerogels are investigated. Different silicon alkoxides, catalysts and water to alkoxide ratios were considered as initial sol parameters to control the hydrolysis and condensation reactions in each reaction stage. Strongly acidic conditions gave rise to the most transparent monolithic aerogels for both tetra-ethyl ortho silicate (TEOS) and tetra methyl ortho silicate (TMOS) based systems. In the TMOS-based system, due to a larger number of monosilicic acids acting as equivalent nucleation centres for further growth, smaller and uniformly distributed particles and interstitial pores were produced during the gellation and supercritical drying. Therefore, with the same catalyst and water content, better transparency was observed in the TMOS-based system than in the TEOS-based system.

    Notes: We investigated the influence of inorganic phosphate concentration on the production of curdlan by Agrobacterium species. A two-step culture method was employed where cells were first cultured, followed by curdlan production under nitrogen-limiting conditions. In the curdlan production step, cells did not grow but metabolized sugar into curdlan. Shake-flask experiments showed that the optimal phosphate concentration for curdlan production was in the range of 0.1–0.3 g l−1. As the cell concentration increased from 0.42 to 1.68 g l−1 in shake-flask cultures, curdlan production increased from 0.44 to 2.80 g l−1. However, the optimal phosphate concentration range was not dependent upon cell concentration. The specific production rate was about 70 mg curdlan g-cell−1 h−1 irrespective of cell concentration. When the phosphate concentration was maintained at 0.5 g l−1 under nitrogen-limiting conditions, as high as 65 g l−1 of curdlan was obtained in 120 h.     Notes: We sought an optimal pH profile to maximize curdlan production in a batch fermentation of Agrobacterium species. The optimal pH profile was calculated using a gradient iteration algorithm based on the minimum principle of Pontryagin. The model equations describing cell growth and curdlan production were developed as functions of pH, sucrose concentration, and ammonium concentration, since the specific rates of cell growth and curdlan production were highly influenced by those parameters. The pH profile provided the strategy to shift the culture pH from the optimal growth condition (pH 7.0) to the optimal production one (pH 5.5) at the time of ammonium exhaustion. By applying the optimal pH profile in the batch process, we obtained significant improvement in curdlan production (64 g L−1) compared to that of constant pH operation (36 g L−1).

    Notes: Abstract As one of the intrinsically polarizable materials used in electrorheological (ER) fluid, polyaniline was synthesized by the chemical oxidation of aniline with ammonium peroxysulfate. ER fluids were prepared by dispersing polyaniline particles in silicone oil, and their rheological properties were measured. The effect of the polymerization temperature of polyaniline on its ER characteristics was investigated. ER fluids with polyaniline particles synthesized at −10°C (PA-10) showed the best ER performance (yield stress) compared with those synthesized at higher temperatures. A difference in the flow behavior of the ER fluids was also investigated through the dielectric spectra of ER fluids.

    Notes: Abstract We isolated a promoter that is inducible by paraquat, a superoxide-generating agent, fromEscherichia coli using the promoter-probe plasmid pRS415. Sequence analysis revealed that the promoter derives from theribA gene encoding GTP cyclohydrolase II, which is the first enzyme in the biosynthetic pathway of riboflavin. We fused thelacZ gene with theribA promoter to monitor the expression of the gene in the single-copy state. LacZ expression from theribA promoter was induced about eight-fold by 200 µM paraquat. Other known superoxide generators, menadione and plumbagin, also induced the expression ofβ-galactosidase in the fusion strain. On the other hand, no significant induction was observed following treatment with hydrogen peroxide, ethanol or heat shock. Induction ofβ-galactosidase was significantly reduced by the introduction of a Δsox-8::cat orsoxS3::Tn10 mutation into the fusion strain, indicating that theribA gene is a member of thesoxRS regulon. The transcriptional start site was determined by primer extension analysis and putative binding sites for SoxS in both orientations were identified. GTP cyclohydrolase II activity in soluble extracts ofE. coli increased more than three-fold on treatment with paraquat. This increase was dependent on thesoxRS locus, and reflects the increase in transcript levels. However, flavin pools did not change significantly. A possible role forribA induction during superoxide stress is discussed.

    Notes: Abstract The ribA gene, encoding GTP cyclohydrolase II in Escherichia coli, is a member of the soxRS regulon, which is induced by superoxide-generating agents. By evaluating lacZ expression driven by the ribA promoter carrying different lengths of upstream region in a monolysogen, we found that the superoxide-responsive element resides between 56 and 94 nucleotides upstream of the transcriptional start site. Purified SoxS protein bound to this region and protected nucleotides between positions −80 and −58 from degradation by DNase I. This region contains a putative SoxS-binding sequence (soxbox) in reverse orientation. The SoxS protein interacted specifically with four guanine residues within the soxbox sequence, as demonstrated by methylation interference analysis. These results clearly indicate that SoxS binds to the reversed soxbox sequence in the ribA gene, while in other known genes of the soxRS regulon it binds to the normally oriented soxbox. Possible modes of interaction between SoxS and RNA polymerase are discussed.

    Notes: Abstract High concentrations of adenosine (Ado), when added to L1210 lymphocytic leukemia cells, resulted in apoptosis or programmed cell death. The apoptotic process was accompanied by distinct morphological changes including chromatin condensation and blebbing of plasma membranes. Extensive DNA fragmentation was correlated with Ado concentrations. Furthermore, apoptosis in these cells was preceded by an early but transient expression of c-myc proto-oncogene, and was not influenced by homocysteine thiolactone added to the cells. Since severe combined immunodeficiency (SCID) is associated with a deficiency of adenosine deaminase, leading to defects in both cellular and humoral immunity, Ado-induced apoptosis may thus be a contributing factor in the pathology of SCID.
