    Notes: Abstract The expression of manganese peroxidase (MnP) in nitrogen-limited cultures of Phanerochaete chrysosporium is regulated by Mn, heat shock (HS), and H2O2 at the level of gene transcription. We have constructed a homologous gene reporter system to further examine the regulation of two mnp genes, mnp1 and mnp2, encoding individual MnP isozymes. Internal deletions of 234 and 359 bp were made within the coding regions of the mnp1 and mnp2 genes, respectively. The truncated mnp genes were subcloned into the shuttle vector pOGI18, which includes the Schizophylum commune ade5 gene as a selectable marker, and transformed into a P. chrysosporium Ade1 auxotrophic mutant. Northern-blot analysis of purified Ade+ transformants demonstrated that both of the truncated mnp genes were regulated in a manner similar to the endogenous mnp genes with respect to nitrogen limitation and induction by Mn, HS, and H2O2.

    Notes: Abstract This paper examines the influence of the materials on the electrical properties of conductors for both high speed MCM structures and microstrip applications. Calculated figures for attenuation in microstrip lines are compared with published results. It is seen that the nature and physical structure as well as the geometrical shape of a conductor track all strongly influence the microwave performance. However, it is also clear that real materials have been developed for these applications which closely approach the behavior predicted for ideal conductors. It is proposed that microwave performance can be predicted by modelling the d.c. conductor materials and extrapolating to high frequencies.

    Notes: Abstract The shear fracture mechanism in a rapidly-solidified aluminium alloy was investigated using both smooth and circumferentially-notched bars under uniaxial tension in a temperature range of 193–423 K and a strain rate range of 10-5–1 s-1 for the alloy under as-extruded and precipitation-hardened states. An analytical procedure was performed to make corrections on the hydrostatic stresses in the necked region of a smooth specimen. A complete shear fracture occurs only for the as-extruded alloy in plain tension at intermediate temperatures and relatively low strain rates. The interfacial delamination is associated with both the gross shear fracture in plain tension and the local shearing in notch tension. Based on experimental observations combined with analytical results, a constraint-releasing mechanism is proposed which fairly accounts for the conditions under which the present shear fracture is evident.

    Notes: Abstract Ti–25Al–10Nb–3V–1Mo alloy has been prepared by gas atomization with atomizing pressure above 8 MPa. The cooling rate of 5×103∼3×105 K s-1 was obtained according to secondary dendritic arm spacing, as the particle size is 30∼300 μm. It is known that in the big particles two dendrite morphologies can be found, cellular dendrite and well-developed dendrite, and in small particles there exists only well-developed dendrite. The surface of particles is characteristic of solidification that is identical to the observation on the cross-sectional views. The micrographs of cross-sectional views show that the big particles, whether of cellular dendrite or well-developed dendrite, solidify by multiple nucleation events while the small particles of well-developed dendrite solidify from a single nucleation event. The microstructure of the alloy consists of single β0 phase.

    Notes: Abstract The flocculation of a dilute titanium dioxide (TiO2) suspension using homopolymers and graft copolymers of acrylamide (AM) and diallyldimethylammonium chloride (DADMAC) was investigated. The graft copolymers produced by γ-irradiating the mixtures of polyacrylamide (PAM) and polyDADMAC gave better flocculating performance than homopolymers, reflecting the higher fractions of large particles and bigger floc size. A kinetic delay in the onset of flocculation was observed after adding the copolymers in the dose range 5–30 [mg polymer]/ [g TiO2]. Increasing dosage resulted in a longer delay period. No significant flocculation was observed when the dose was above 50 [mg polymer]/[g TiO2]. This delay was interpreted in terms of the re-conformation of polymer chains driven by charge neutralization, between the positively charged polymer branches and the negative particle surface. Depending on the dosage used, the flocculation behavior of the graft copolymer has been suggested to be equilibrium and non-equilibrium flocculation. It was also observed that re-conformation is not affected by the ion strength of the media, but a strong shear force significantly reduces the chain reconformation time.

    Notes: Abstract A novel quantitative method for the determination of proteins in aqueous solutions has been based on the quenching of the resonance scattering light of colloidal silver chloride in the presence of proteins. The detection limits for eight kinds of proteins (BSA, HSA, egg albumin, human γ-IgG,α-chymotrypsin, E. Coli. alpsase, myoglobin, α-casein) were at about 8 ng/mL; the linear ranges of the calibration curves were 10–400 ng/mL ¶under optimal conditions,except for human γ-IgG (20–¶400 ng/mL), myoglobin (10–300 ng/mL), and α-casein (10–300 ng/mL). Three wavelengths (398 nm, 475 nm, 499 nm) were all suitable for the determination and any acidity from pH 3.0 to pH 9.0 could be chosen. A few non-protein substances at high concentration levels interfered with this method, but this problem could simply be overcome by diluting the samples before the assay. Mechanism studies showed that the quenching effect of proteins on the scattering light of colloidal silver chloride was mainly due to the coagulation of AgCl particles retarded by protein. The method was employed for the determination of total protein in human serum with sactifactory results.

    Notes: Abstract A fluorescence quenching method was developed for the rapid determination of DNA and RNA using magdala red as fluorescence probe. In weakly acidic ¶medium, the fluorescence of magdala red (λex〉lem = 54055 nm) can be largely quenched by DNA or RNA. The calibration graphs are linear over the range 0.01–¶1.2 μg/mL for both calf thymus DNA (CT DNA) and salmon DNA (SM DNA), and 0.015–1.0 μg/mL for yeast RNA, respectively. The corresponding detection limits are ¶6.0 ng/mL for CT DNA, 7.0 ng/mL for SM DNA and ¶15.0 ng/mL for yeast RNA, respectively. CT DNA could be determined in the presence of 20% (w/w) yeast RNA, and the relative standard deviation of six replicate measurements is 3.18% for 400 ng/mL of CT DNA. Interference from coexisting substances in the determination of DNA was also examined. Real samples were determined with satisfactory results.

    Notes: Abstract. A rice transcript, Rim2, was identified that accumulated in both incompatible and compatible interactions between rice and Magnaporthe grisea. The Rim2 transcript also accumulated in response to treatment with a cell wall elicitor derived from M. grisea. A 3.3-kb RIM2 cDNA clone was isolated and is predicted to encode a protein of 653 amino acids, which shares 32–55% identity with TNP2-like proteins encoded by CACTA transposons of other plants. A 1.05-kb segment of the Rim2 sequence shows 82% nucleotide sequence identity with sequences flanking the A1 and C members of the rice Xa21 disease resistance gene family. The 5'-upstream region of Rim2 was cloned and the transcriptional start sites were identified. The 5' and 3' noncoding termini of Rim2 are AT-rich. A cis-element showing similarity to a sequence that mediates defense-associated transcriptional activation of the tobacco retrotransposon Tnt1, and four motifs that fit the consensus sequence of the elicitor-responsive elements in the promoters of the parsley PR-1 genes were found in the 5'-upstream region. Four imperfect tandem repeats were identified in the 3' noncoding terminus. Southern analysis with genomic DNA from different rice species indicated that Rim2 is present in 3–4 copies per genome. These results suggest that Rim2 may be one component of a large CACTA-like element, whose transcript accumulates in response to attack by pathogens.
