    Notes: Summary Trypsin-catalyzed coupling of porcine desoctapeptide-insulin with synthetic octapeptides produced the [LeuB24]-(I), [LeuB25]- (II) and [LeuB24, LeuB25]- (III)analogues of human insulin. I, II and III displayed respectively 20–30%, 1–2% and 0.5% of the receptor binding activity of the normal hormone. Biological activities of these analogues seemed to be proportional to their binding potencies when assayed in vitro, while in an in vivo assay analogue I was fully active and II exhibited 10–20% of normal activity. III was less active than II in all assays tested.

    Notes: Abstract Oxidant toxicity of indole was demonstrated by the induction of alkylhydroperoxide reductase subunit C (AhpC) in Escherichia coli K12 and by the constitutive overproduction of AhpC in a variant of E. coli JM109 with enhanced resistance to indole. Oxidant toxicity was also indicated in an indole-adapted variant of Brevibacterium flavum by the indole-inducible overproduction of a novel 36-kDa protein with N-terminal sequence similarity to proteins involved in superoxide and singlet oxygen resistance. It is proposed that indole dissolved in membrane lipids, which caused membrane derangement and enabled direct interaction of redox-cycling isoprenoid quinones and dioxygen, resulting in the generation of superoxide. ¶A direct indication of membrane derangement in E. coli may be the indole-inducible overproduction of spheroplast protein y (Spy).

    Notes: Abstract Chlorella ellipsoidea was incubated in nitrogendeficient medium to examine the changes in the relative DNA content per nucleus, which were measured by epifluorescent microspectrophotometry combined with Feulgen-pararosaniline staining. The results were compared with synchronous multiplication in the basal medium. The minimum and maximum measured DNA values per nucleus were 2.5 and 280, which were related to 1 C and 112 C respectively. The mode of the DNA value moved reciprocally between 8 C and 4 C levels under multiplication in basal medium, which one mother cell producing four autospores in each synchronous division. The average value of the modes over 5 days of nitrogen starvation was 8 C. The frequency distribution of the relative DNA value consisted of at most five levels, 1–3.2 C, 4–12 C, 28 C, 32 C, and 40–112 C. The modal changes suggested the occurrence of DNA synthesis caused by some intracellular nitrogen pools. During the nitrogen starvation, 10-fold cell multiplication and a small amount of cell growth occurred.

    Notes: Abstract Magnetic properties of compacted cores which are formed from amorphous alloy powders prepared by a rapid quenching water atomization process are studied. The coercivity of the as-compacted core was estimated to beH c=0.028(λσ/I s)+1200 with the idea that the coercive force depends on the magnitude of the stress variation. The a.c. magnetic properties are remarkably improved by use of the coupling agent and zinc stearate. The use of the amorphous powder core for choke coils of switching power supplies leads to the decrease of cut-off current, that is, an improvement in the efficiency of power supplies.

    Notes: Abstract Second harmonic generation (S.H.G.) in inclusion complexes between dimethyl β-cyclodextrin (dimethyl β-CD) and nitroaniline derivatives occurs. This is due to the destruction of the centrosymmtric crystal structure in nitroaniline derivatives caused by forming inclusion complexes. S.H.G. intensity of dimethyl β-CD complex with N-methyl-nitroaniline is 5.5 times as large as that of urea.

    Notes: Abstract Transparent or translucent fibre-reinforced polymeric wires have been produced in an attempt to reproduce the mechanical properties of the metallic wires in current use in orthodontics. Two methods were employed: mould polymerization, and hot-drawing. Both methods produced wires of 0.5 mm diameter. Two polymers were investigated, poly(methyl methacrylate) and epoxy resin, and these were filled with either long silane-coated alumina fibres or fibres made from CPSA glass. Whilst mould-polymerized wires showed a linear increase in Young's modulus with fibre content, they did not obey the rule of mixtures. However, the hot-drawn wires did, and they also demonstrated the rigidity, strength and good elastic recovery needed for use in orthodontics.

    Notes: Abstract TZT-1027, a newly synthesized dolastatin 10 derivative, is a potent antitumor agent which inhibits microtubule polymerization and perturbs microtubule dynamics. In this report, we investigated whether TZT-1027 inhibited the growth of various human cancer cells, and the cell death caused by TZT-1027 was due to apoptosis. In addition, we elucidated the apoptosis machinery induced by treatment with TZT-1027. The 50% growth-inhibitory concentrations (IC50 values) of TZT-1027 on cancer cells derived from various sources were not more than 5.9 ng/ml. TZT-1027 showed superior cytotoxicity than any other antitumor agents. Next, we evaluated morphological nuclear change, namely, chromatin condensation and DNA fragmentation. We used three cancer cell lines derived from different types in view of having apoptosis related protein, human leukemia HL-60 (in the presence of both Caspase-3 and Bcl-2), human breast cancer MCF-7 (in the absence of Caspase-3), and human prostate cancer DU145 (in the absence of Bcl-2). TZT-1027 induced DNA fragmentation in the presence but not absence of Caspase-3. Nevertheless, apoptic chromatin condensation was observed in all cancer cells even if there was no Caspase-3. Furthermore, we examined whether TZT-1027, microtubule-disrupting agent, influenced cell cycle progression. Flow cytometric analysis revealed the cells treated with TZT-1027, and with the other antimicrotubule agents, to be arrested at the G2/M phase and subsequently to show fragmented DNA smaller than that of G1 phase cells. Moreover, we tested TZT-1027 for its ability to induce Bcl-2 phosphorylation in human cancer cell lines. TZT-1027 and other agents which interacted with microtubules induced Bcl-2 phosphorylation, whereas DNA-damaging agents did not. The present results suggested an association of the growth-inhibitory effect of TZT-1027 with the induction of apoptosis and indicated that the apoptosis induced by TZT-1027 was followed by G2/M arrest even if there was no Caspase-3 or Bcl-2.
