    Notes: Abstract Under starvation conditions, 50% survivorship times displayed no significant relationship with cell size in 2 ciliate species in this study and 5 protozoan species from the literature. Differences in survival ability were attributed to differences in weight-specific respiratory rate and relative motility among these 7 species. At low bacterial levels, 4 ciliate species in this study displayed significant differences in survivorship. High survivorship ofEuplotes patella relative to that ofParamecium caudatum andParaurostyla sp. at low ciliate densities was attributed to the lower individual energy requirements of this smaller species. High survivorship ofStentor coeruleus was interpreted as an effect of its large quantity of reserves and low respiratory rate.     Notes: Abstract We consider secret sharing schemes which, through an initial issuing of shares to a group of participants, permit a number of different secrets to be protected. Each secret is associated with a (potentially different) access structure and a particular secret can be reconstructed by any group of participants from its associated access structure without the need for further broadcast information. We consider ideal secret sharing schemes in this more general environment. In particular, we classify the collections of access structures that can be combined in such an ideal secret sharing scheme and we provide a general method of construction for such schemes. We also explore the extent to which the results that connect ideal secret sharing schemes to matroids can be appropriately generalized.

    Notes: Abstract To test the hypothesis that the T-cell receptor (Tcr) λ gene encodes a natural killer (NK) cell receptor molecule, three human NK clones and fresh peripheral blood lymphocytes with NK activity from two patients with a CD16+ lymphocytosis were analyzed for rearrangements and expression of the human Tcr α, β, and λ genes. Two of the clones displayed distinct rearrangements of their Tcr β and λ genes and expressed mature Tcr α, β, and αl RNA. However, one of the clones and both patient samples displayed marked NK activity but failed to rearrange or express any of their Tcr genes. These findings demonstrate that human natural killer activity is not dependent on Tcr λ gene rearrangement and expression. In addition, they confirm previous findings concerning the lack of Tcr α and β gene expression in some natural killer cells. Thus, they suggest the existence of additional NK-specific recognition molecules.

    Notes: Abstract This paper discusses an effective clock methodology for the design of high-performance microprocessors. Key attributes include the clustering and balancing of clock loads, multiple clock domains, a balanced clock router with variable width wires to minimize skew, hierarchical clock wiring, automated verification, an interface to the Cadence Design Framework II™ environment, and a complete network model of the clock distribution, including loads. This clock methodology enabled creation of the entire clock network, including verification, in less than three days with approximately 180 ps of skew.

    Notes: Summary Murine interleukin-3 (Mu IL-3) cDNA was previously expressed inEscherichia coli using atac promoter and a constitutive high copy number plasmid vector. We found that significant increases in expression levels could be realized by using thetac promoter for the expression of Mu IL-3 in a plasmid vector possessing a temperature-inducible runaway-replicon. In contrast, use of anlpp promoter under similar conditions did not result in an increase in the Mu IL-3 expression level. Significant differences were observed when the expression levels of IL-3 were monitored in variousE. coli hosts having different genetic backgrounds. A mutant ofE. coli which lacks the protease La was found to increase the level of IL-3 produced. This report describes the effect of a specific protease-deficientE. coli host strain, as well as the effect of different promoters and plasmid replicons on the expression levels and stability of a heterologous gene product.

    Notes: Abstract A secret sharing scheme for an incomplete access structure (Γ,Δ) is a method of distributing information about a secret among a group of participants in such a way that sets of participants in Γ can reconstruct the secret and sets of participants in Δ can not obtain any new information about the secret. In this paper we present a more precise definition of secret sharing schemes in terms of information theory, and a new decomposition theorem. This theorem generalizes previous decomposition theorems and also works for a more general class of access structures. We demonstrate some applications of the theorem.

    Notes: Abstract Given a set of participants we wish to distribute information relating to a secret in such a way that only specified groups of participants can reconstruct the secret. We consider here a special class of such schemes that can be described in terms of finite geometries as first proposed by Simmons. We formalize the Simmons model and show that given a geometric scheme for a particular access structure it is possible to find another geometric scheme whose access structure is the dual of the original scheme, and which has the same average and worst-case information rates as the original scheme. In particular this shows that if an ideal geometric scheme exists then an ideal geometric scheme exists for the dual access structure.

    Notes: Abstract Although placental trophoblasts, the only fetal cells in direct contact with infectious maternal blood, can be infected with HIV, the precise cause for the low transmission rate of virus across the placental barrier is unknown. One of the most common conjectures is that maternal anti-HIV antibodies (Abs) contribute to the protection of the fetus. This hypothesis has been tested in vitro by infecting the CD4-negative placental trophoblast line, BeWo, with HIV-1IIIB in the presence of serial dilutions of neutralizing monoclonal Abs against the V3 loop (No. 694) or CD4-binding conformational domain (No. 588). The results, based on measurement of p24 production from virus-exposed cells, reveal that the titers of Abs, adequate in preventing the infection of control MT-4 T lymphocytes, were less effective in protecting trophoblasts. Furthermore, PCR analysis of HIV DNA formed after a single round of infection has shown no significant decrease in the number of viral copies in Ab-protected BeWo cells. An anti-HIV serum from a pregnant woman did also have no effect. Although our in vitro observations do not necessarily apply to the in vivo situation, the results suggest that the humoral immune response sustained by neutralizing Abs may be able to protect T lymphocytes, but not placental trophoblasts. The findings are consistent with recent clinical studies demonstrating a lack of correlation between the presence of neutralizing anti-HIV Abs in pregnant women and HIV transmission in utero.

    Notes: Abstract The human immunodeficiency virus type-1 regulatory protein Rev is absolutely required for the production of viral structural proteins. Splice sites have been seen to function ascis-acting repressor sequendes (CRS) and inhibit expression of the Rev-dependent RNAs. In order to analyze the role of a splice donor in Rev dependence, the wild-type 5′ splice donor of HIV-1 was mutated in the context of othergag sequences. Following transient transfection, RNA expression by RT-PCR was analyzed. The unspliced RNA produced by the mutant construct still required Rev for the cytoplasmic accumulation of the RNA. Despite deletion of the wild-type 5′ splice donor and thetat splice acceptor was used. A cryptic splice donor was identified by PCR and subsequent cloning of the spliced RNA. The cryptic site is 5/9 to the consensus sequence and located immediately downstream of the initiation codon (ATG) for Gag. Analysis of the RNA product containing the cryptic splice donor revealed that the Rev was required for the cytoplasmic accumulation of unspliced RNA, while spliced RNA was Rev independent. Transfection of a wild-type construct also demonstrated usage of the cryptic splice donor. These results indicate that a cryptic splice donor can be activated when the wild-type splice donor is inactivated and that the cryptic splice donor may retain Rev regulation. The findings also suggest the potential for cryptic splice sites to serve as CRS in the determining the Rev dependence of viral RNAs.
