    Notes: Summary Transport of the nucleoside analog cytosine-arabinoside (CAR) in transformed hamster cells in culture has been studied in conditions of minimal metabolic conversion. Uptake (zero-trans in) properties at 20°C over a limited range of CAR concentrations were characterized by aK m of 350 μm and a maximal velocity (V) of 780 μm·min−1 (V/K m =2.28 min−1). Equilibrium exchange at 20°C over a wider range of concentrations was best described by a saturable component with aK m of 500 μm and av of 1230 μm·min−1 (V/K m =2.26 min−1) and either a saturable component of highK m or a nonsaturable component ofk=0.3 min−1. For the saturable component, thev/K m values were similar in both procedures. CAR transport was inhibited by various metabolizable nucleosides. Uptake of some of these nucleosides was inhibited by CAR. CAR transport and uridine uptake were inhibited in a reversible but partially competitive fashion by high affinity probes like S-(p-nitrobenzyl-6-mercaptoinosine (NBMI) (K i 〈0.5nm) and in an irreversible fashion by SH reagents such as N-ethylmaleiimide (NEM). The organomercurialp-hydroxymercuribenzene sulfonate (pMBS) markedly stimulated transport of these nucleosides, but also markedly potentiated the inhibitory effects of either NBMI or NEM. These effects are interpreted either in terms of models which invoke allosteric properties or in terms of two transport systems which display distinct chemical susceptibilities to externally added probes.

    Notes: Summary Cortical bone can be modeled as a complex hierarchical composite interrelating both structure and material properties on four levels of structural organization: molecular, ultrastructural, microscopic, and macroscopic. In young animals, the microstructural systems are long parallel lamellar units, plexiform bone, which in older or more mature animals converts by internal remodeling into multiple concentric lamellar units, secondary osteons, forming haversian bone. Ultrasonic wave propagation measurements performed on both plexiform and haversian bone clearly show a definitive relationship with microstructure; haversian bone can be described as a transversely symmetric material whereas plexiform bone appears to be orthotropic in nature. The anisotropy of the elastic constants are found to reflect the tissue symmetry; moreover, plexiform bone is stiffer and more rigid in all directions than is haversian bone. Similar experiments were performed on osteoporotic and osteopetrotic bone. While the results for osteoporotic bone are understandable in terms of the increased porosity, the results for the osteopetrotic bone are anomalous with respect to its density. Since Wolff, the remodeling of bone has been interpreted as a way of altering the mechanical properties to suit some need. For haversian remodeling from plexiform bone, the argument that adaptation occurs to optimize properties requires additional clarification since haversian bone appears to have inferior mechanical properties to plexiform bone.

    Notes: Abstract The microstructure of cortical bone may exhibit either transverse isotropic or orthotropic symmetry, thus requiring either five or nine independent elastic stiffness coefficients (or compliances), respectively, to describe its elastic anisotropy. Our previous analysis to describe this anisotropy in terms of two scalar quantities for the transverse isotropic case is extended here to include orthotropic symmetry. The new results for orthotropic symmetry are compared with previous calculations using the transverse isotropic analysis on the same sets of anisotropic elastic constants for bone, determined either by mechanical or by ultrasonic experiments. In addition, the orthotropic calculation has been applied to full sets of orthotropic elastic stiffness coefficients of a large variety of wood species. Although having some resemblance to plexiform bone in microstructural organization, there is a dramatic difference in both the shear and the compressive elastic anisotropy between the two materials: wood is at least one order of magnitude more anisotropic than bone.

Durch eine neue Art Drehaufnahmen wurde es im höchsten Grade wahrscheinlich gemacht, daß die „kristallinische“ Materie aus der „amorphen“ entsteht, wenn Kautschuk gedehnt wird. 2. Es muß unterschieden werden zwischen Gleichrichtung der Moleküle und Bildung eines dreidimensionalen Gitters; das letztere liegt hier vor. Ob das echte Kristalle sind, blieb unentschieden. 3. Aus dieser Annahme findet der Joule-Effekt des Kautschuks (Erwärmung bei der Dehnung) seine ungezwungene Erklärung: jede amorphe Substanz entwickelt Wärme, wenn sie kristallisiert. 4. Ebenso läßt sich charakteristisches im Verlauf des Dehnungsgrades mit der Spannung und im Verlauf der optischen Anisotropie mit dem Dehnungsgrad auf Grund dieser Annahme erklären. 5. Ein Feld für weitere experimentelle Untersuchungen ergibt sich auf Grund dieser Tatsachen.
