    Notes: Abstract Three chrysanthemum (Dendranthema grandiflora) cultivars were cocultivated with 2 Agrobacterium tumefaciens strains in combination with 4 pBIN19 derived binary plasmids, all carrying the Nosnptll selection gene and 35Sgus(intron) reporter gene. All binary plasmids transferred DNA to chrysanthemum explants but only pMOG410 gave good stable expression of GUS. This plasmid differs from the other plasmids in 2 aspects: 1) It carries a restored nptll gene and 2) the selection gene is positioned at the left border side of the reporter gene. Cocultivation with AGLO(pMOG410) yielded up to 13 GUS positive shoots per 100 explants. The presence of the gus and nptll gene in recovered shoots was confirmed by PCR and Southern blot analysis.

    Notes: Summary To develop an Agrobacterium mediated transformation protocol for chrysanthemum we studied the transformation efficiency of commonly used A.tumefaciens strains on 14 genotypes by comparing tumour size and frequency. One genotype was analyzed in detail using 14 strains of both A.tumefaciens and A.rhizogenes. Only a few genotype/strain combinations resulted in significant tumour formation. Especially 0-type strains were highly efficient. An 0-type strain was used to transfer genes for neomycine phosphotransferase (NPT II) and ß-glucuronidase (GUS) to a susceptible cultivar. Transfer of the GUS gene was confirmed by using the Polymerase Chain Reaction (PCR).

    Notes: Abstract The objective of the study was assess the utility during anaesthesia of noninvasive continuous blood pressure measurement techniques which use intermittent oscillometric blood pressure measurement for their calibration. The assessment was performed by comparing noninvasive blood pressure with intra-arterial blood pressure. The noninvasive blood pressure measurement device used for evaluation was the NCAT N-500 which uses tonometry for its continuous measurements. Fifteen patients were studied. In 10 patients the intra-arterial blood pressure curve (IBP) was recorded from the radial artery (radial artery group), and in 5 patients it was recorded from the brachial artery (brachial artery group). In all patients the oscillometrically calibrated tonometric blood pressure (OTBP) was recorded from the other arm. To discriminate between calibration dependent measurement error and tonometric measurement error, the OTBP signal was recalibrated against the IBP signal to get the intra-arterial calibrated tonometric pressure curve (ITBP). OTBP-IBP reflected the overall measurement error, ITBP-IBP the error of the tonometric measurement, and OTBP-ITBP the calibration dependent measurement error. According to criteria formulated in the discussion the accuracy and agreement of the ITBP-IBP measurements were clinical acceptable. Accuracy and agreement of OTBP-IBP and of OTBP-ITBP were not clinical acceptable. Correlation of dynamic behavior was lower for OTBP than for ITBP. A significant effect of site difference between calibration measurements and continuous measurements was not found. It is concluded that the approach of continuous noninvasive blood pressure measurement based on the combination of two different measurement methods, in which the continuous method is calibrated by the oscillometric method, lead to clinical unacceptable accuracy and agreement in the patient group studied.

    Notes: Summary The vase life duration and stem stiffness of 12 parent clones and 77 progenies of Gerbera jamesonii were studied both in summer and in winter. In winter, vase life was shorter and the stems were weak. Few exceptions from this general pattern were detected. The average vase life duration ranged from 16.0 days for the best to 8.9 days for the worst progeny. Individual plants were selected with a vase life of 20 or more days. The curvature of the stem, measured after 24 hrs dry storage, is associated with the folding of the stem in the vase. Selection for low curvature may decrease the incidence of folding and with it the variation in vase life of Gerbera.

    Notes: Summary An incomplete diallel cross with selfs and reciprocals was made with twelve cultivars of Gerbera jamesonii. Very significant differences occurred between GCA's of the parents for cut flower yield, earliness and number of lateral shoots. The selfs were mainly responsible for the significant SCA's. A positive genetic correlation occurs between the number of lateral shoots at anthesis of the first flower and total flower production. The phenotypic performance of parents (measured on cuttings) was poorly related to their breeding value (measured on seedlings). It is suggested that this is due to physiological differences between cuttings and seedlings.

    Notes: Summary The feeding, fecundity, mortality and rate of development of Liriomyza trifolii (Burgess) on 12 cultivars of Dendranthema grandiflora (Tzvel.) (Syn. Chrysanthemum morifolium (Ramat.)) were determined in ‘no choice’ experiments. Number of feeding punctures, number of eggs laid and total number of larvae varied between cultivars but these characters were not strongly associated with leafminer population increase. Good indicators of host plant resistance were larval mortality, number of pupae and duration of the larval period. Of these three, larval mortality which could be as high as 98% was most suitable. Based on life history data it was predicted that Liriomyza trifolii cannot survive on the resistant cultivar Penny Lane. This prediction was verified experimentally.

    Notes: Abstract Explants from leaves of in vitro-grown chrysanthemum (Dendranthema grandiflora Tzvel.) cultivars regenerated adventitious shoots without an intermediate callus phase. Puncturing explants with a brush increased regenerations, but in combination with cocultivation with Agrobacterium tumefaciens it had an adverse effect on shoot formation. The negative effect of brushing and cocultivation could be overcome by preculturing explants for 8 days. Preculture altered the location of transformed sites but did not inhibit transformation. Regeneration following cocultivation with Agrobacterium is also encouraged if alternative regeneration protocols are used that do not require brushing.

    Notes: Abstract The Cortronic APM 770 (Cortronic, Ronkonkoma, NY) is a commercial device that claims to measure blood pressure noninvasively and continuously with the use of a standard blood pressure cuff. The aim of our study was to assess the performance of the continuous-mode blood pressure readings of the Cortronic during anesthesia and surgery. We recorded blood pressure in 5 patients bilaterally. An intraarterial pressure (IAP) curve was recorded from 1 arm and the Cortronic pressure curve (CPC) was recorded from the other. For statistical analysis the period between 2 Cortronic recalibrations was defined as the intercalibration interval. The duration of these intervals ranged from 20 to 0.5 minutes. Four paired samples were drawn from each interval. The first sample in an interval represented the recalibration blood pressure; the other samples represented the continuous blood pressure. A total of 1,232 samples were taken, of which 308 were recalibration. The median of the differences and the 2.5th and 97.5th percentile limits of agreement were determined. Their respective values for diastolic and systolic recalibration measurements were 5, −17, and 34 mm Hg, and 6, −12, and 38 mm Hg. Their values for continuous measurements were 4, −23.5, and 32 mm Hg, and 6, −30, and 70 mm Hg. Changes in CPC were evaluated against changes in the corresponding IAP by plotting them in 4-quadrant graphs. In these graphs the Spearman rank correlations were betweenr=−0.17 andr=0.01. We observed opposite CPC and IAP trends on 24 occasions during this study. We performed a simple simulation study to better understand the measurement method of the Cortronic. The study showed a positive relationship between pulsation volume and CPC amplitude, and between pulsation rate and CPC amplitude. We conclude that during anesthesia and surgery continuous-mode blood pressure readings of the Cortronic are unreliable, and suggest that the phenomenon of the two pressures' moving in opposite directions is inherent to the measurement principles of the device.
