Notes: Abstract: The binding of polymerised actin--a prototype of semi-flexible macromolecule--to lipid monolayers is studied by neutron reflectivity to deduce the average thickness, the interfacial roughness and the polymer volume fraction of the adsorbed film. Electrostatic interaction between actin filaments (F-actin) and the lipid monolayer is mediated through a cationic lipid (1,2-dimyristoyl-3-trimethylammonium-propane, DMTAP). The adsorbed F-actin forms a monolayer with an average thickness of 69 to 84 Å, depending on the ionic strength of the buffer and surface charge density of the monolayer. The volume fraction of F-actin in the adsorbed layer can be as high as 0.29. The thickness and high volume fraction of the actin layer suggest that actin filaments lie flat on the surface and form nematic ordering. The binding-unbinding equilibrium of F-actin is controlled by the ionic strength and exhibits a strong hysteresis. In contrast to the results obtained for filamentous actin, monomeric actin (G-actin) shows no detectable binding to the positively charged lipid layers.

Notes: Abstract A noninvasive optical method is described which allows the measurement of the vertical component of the instantaneous displacement of a surface at one or more points. The method has been used to study the motion of a passive compliant layer responding to the random forcing of a fully developed turbulent boundary layer. However, in principle, the measurement technique described here can be used equally well with any surface capable of scattering light and to which optical access can be gained. The technique relies on the use of electro-optic position-sensitive detectors; this type of transducer produces changes in current which are linearly proportional to the displacement of a spot of light imaged onto the active area of the detector. The system can resolve displacements as small as 2 μm for a point 1.8 mm in diameter; the final output signal of the system is found to be linear for displacements up to 200 μm, and the overall frequency response is from DC to greater than 1 kHz. As an example of the use of the system, results detailing measurements obtained at both one and two points simultaneously are presented.

Notes: Abstract The Azospirillum 10SW used in our experiments was isolated from roots of wheat growing in nitrogen-poor soil of a hilly region of Nepal, where inorganic nitrogen fertilizers were never used. The main objectives of this work were to assess the effects of inorganic nitrogen fertilization in the yield responses of wheat grown in association with the bacteria. The in vitro experiments were done in laboratory, whereas the pot experiments were performed in a greenhouse. The nitrogenase activities of in vitro grown Azospirillum were repressed by nitrate. The magnitude of repression was lower when the bacteria were growing in association with wheat. The number of roots per plant was increased significantly in inoculated plants irrespective of the nitrate concentration of the medium. Inoculation with Azospirillum 10SW also increased the yield of wheat grown in pots with medium levels of nitrogen fertilization. These data show the possibility of inoculation of this Azospirillum spp. in combination with nitrogen fertilizer to improve the yield of wheat. Azospirillum inoculation enhanced the development of roots and shoots in the early growth stages of wheat. It may be one of the factors responsible for the yield increases.

Notes: Summary We have exploited the spatial resolution of the magnetic sensor to provide numerical estimates of the atrial contribution to magnetic fields recorded just prior to ventricular depolarization. The contribution is found to be small, strenghening our original interpretation that such fields originate mostly in the conduction system of the heart.

Notes: Summary We have studied the magnetic field produced by the brain when attending to a variety of rare events. The temporal character of the response to a rare auditory stimulus is reported. The magnetic field appears to offer a useful new probe of endogeneous activity in the human brain.

Notes: Summary A transfilter culture of the non-leguminous plantPortulaca grandiflora var. JR andRhizobium sp. ‘cowpea’ 32H1 was established. Using15N2-analysis we demonstrated that15N-containing substances produced by the bacteria passed through the membrane and15N was enriched in the plant cells.

Notes: Summary In isolated mitochondria of heart muscle from rabbits and oxen there is, under suitable conditions, an accumulation of Ca++, which is significantly enhanced by elevating the K+/Na+ quotient of the incubation medium. K-strophanthine (10−5−10−7) does not influence the accumulation of Ca++ by the mitochondria of heart muscle. Therefore the intracellular increase in exchangeable Ca++ observed after digitalis-glycosides could be explained by a decrease of the intracellular K+/Na+ quotient, which is caused by inhibition of the membrane ATPase and diminishes the capacity for Ca++ accumulation in mitochondria.

Notes: Summary In transfilter associations ofPortulaca grandiflora callus tissues withRhizobium sp. ‘cowpea’ 32H1 the nitrogenium fixed by the bacteria was channelled into the normal pathway of nitrogen metabolism of higher plants. In associations kept in an15N2 containing atmosphere 10% of the15N taken up into the plant cells was incorporated intoPortulaca proteins. The results demonstrate that cells of nonleguminous plants are profiting from the quasi-symbiotic situation.

Notes: Summary Plant nuclei prepared from protoplasts can be used as a cell-free system for testing the template activity of procaryotic DNA for plant polymerases. We were able to demonstrate that plant polymerases ofPetunia hybrida are capable of transcribing linear bacterial DNA, whereas supercoiled DNA could not be used as a template.
